Abstract
1. Способ введения представляющей интерес молекулы в клетку растения, имеющую клеточную стенку, предусматривающий ! обеспечение клетки растения, имеющей клеточную стенку; ! покрытие наночастицы представляющей интерес молекулой; ! приведение клетки, имеющей клеточную стенку, и имеющей покрытие наночастицы в контакт друг с другом; и ! позволение поглощения наночастицы и представляющей интерес молекулы в клетку, содержащую клеточную стенку. ! 2. Способ по п.1, где покрытие наночастицы представляющей интерес молекулой предусматривает иммобилизацию представляющей интерес молекулы посредством нековалентной абсорбции на поверхности наночастицы. ! 3. Способ по п.1, дополнительно предусматривающий абсорбцию представляющей интерес молекулы в наночастицу. !4. Способ по п.1, дополнительно предусматривающий позволение поглощения наночастицы в компартмент клетки растения, содержащей клеточную стенку. ! 5. Способ по п.4, дополнительно предусматривающий покрытие наночастицы нацеливающим на субклеточный компартмент белком. ! 6. Способ по п.4, где компартмент выбран из группы, состоящей из цитозоля, ядра, тонопластов, пластиды, этиопласта, хромопласта, лейкопласта, элайопласта, протеинопласта, амилопласта, хлоропласта и просвета двойной мембраны. ! 7. Способ по п.1, где клетка растения, содержащая клеточную стенку, выбрана из группы, состоящей из клеток табака, моркови, кукурузы, канолы, рапса, хлопчатника, пальмы, арахиса, сои, Oryza sp., Arabidopsis sp., Ricinus sp. и сахарного тростника. ! 8. Способ по п.1, где клетка растения происходит из ткани, выбранной из группы, состоящей из зародыша, меристемы, каллуса, пыльцы, листьев, пыльников, корней, кон�
Claims (21)
1. Способ введения представляющей интерес молекулы в клетку растения, имеющую клеточную стенку, предусматривающий
обеспечение клетки растения, имеющей клеточную стенку;
покрытие наночастицы представляющей интерес молекулой;
приведение клетки, имеющей клеточную стенку, и имеющей покрытие наночастицы в контакт друг с другом; и
позволение поглощения наночастицы и представляющей интерес молекулы в клетку, содержащую клеточную стенку.
2. Способ по п.1, где покрытие наночастицы представляющей интерес молекулой предусматривает иммобилизацию представляющей интерес молекулы посредством нековалентной абсорбции на поверхности наночастицы.
3. Способ по п.1, дополнительно предусматривающий абсорбцию представляющей интерес молекулы в наночастицу.
4. Способ по п.1, дополнительно предусматривающий позволение поглощения наночастицы в компартмент клетки растения, содержащей клеточную стенку.
5. Способ по п.4, дополнительно предусматривающий покрытие наночастицы нацеливающим на субклеточный компартмент белком.
6. Способ по п.4, где компартмент выбран из группы, состоящей из цитозоля, ядра, тонопластов, пластиды, этиопласта, хромопласта, лейкопласта, элайопласта, протеинопласта, амилопласта, хлоропласта и просвета двойной мембраны.
7. Способ по п.1, где клетка растения, содержащая клеточную стенку, выбрана из группы, состоящей из клеток табака, моркови, кукурузы, канолы, рапса, хлопчатника, пальмы, арахиса, сои, Oryza sp., Arabidopsis sp., Ricinus sp. и сахарного тростника.
8. Способ по п.1, где клетка растения происходит из ткани, выбранной из группы, состоящей из зародыша, меристемы, каллуса, пыльцы, листьев, пыльников, корней, кончиков корней, цветков, семян, стручков и стеблей.
9. Способ по п.1, где наночастица выбрана из группы, состоящей из наночастиц золота, покрытых золотом наночастиц, пористых наночастиц, мезопористых наночастиц, наночастиц диоксида кремния, наночастиц полимера, вольфрамовых наночастиц, желатиновых наночастиц, нанооболочек, наносердцевин (нанокоров), наносфер, наностержней, магнитных наночастиц и их комбинацией.
10. Способ по п.1, дополнительно предусматривающий дериватизацию поверхности наночастицы.
11. Способ по п.1, где представляющая интерес молекула содержит компонент, выбранный из группы, состоящей из молекул нуклеиновых кислот, ДНК, РНК, RNAi, генов, плазмид, космид, YAC, BAC, полипептидов, ферментов, гормонов, гликопептидов, сахаров, жиров, сигнальных пептидов, антител, витаминов, мессенджеров, вторичных мессенджеров, аминокислот, цАМФ, лекарственных средств, гербицидов, фунгицидов, антибиотиков и их комбинаций.
12. Способ по п.10, где представляющая интерес молекула содержит ген.
13. Способ по п.11, где ген является геном чужеродного белка, агрономическим геном или маркерным геном.
14. Способ по п.11, дополнительно предусматривающий отбор клеток, которые стабильно интегрировали представляющую интерес молекулу.
15. Способ по п.13, где отобранные клетки являются регенерируемыми клетками.
16. Способ по п.14, дополнительно предусматривающий регенерацию растения из регенерируемых клеток.
17. Способ экспрессии гена, предусматривающий
обеспечение клетки растения, имеющей клеточную стенку;
покрытие наночастицы геном;
приведение клетки, имеющей клеточную стенку, и имеющей покрытие наночастицы в контакт друг с другом;
позволение поглощения наночастицы и представляющей интерес молекулы в клетку растения, содержащую клеточную стенку; и
экспрессию гена.
18. Способ по п.16, где ген экспрессируется в хлоропласте.
19. Способ по п.16, дополнительно предусматривающий отбор на клетки, экспрессирующие ген.
20. Способ переноса молекулярного вещества в клетку растения, предусматривающий
покрытие наночастицы плазмидной ДНК; и
контактирование имеющей покрытие наночастицы с интактными, несущими клеточную стенку клетками растения при условиях, допускающих поглощение имеющих покрытие наночастиц в клетку растения.
21. Способ по п.19, где наночастица содержит золото.
