Abstract
1. Способ получения печеночных стволовых клеток из непеченочных стволовых клеток, включающий ! (a) обеспечение стволовых клеток, полученных из непеченочных тканей; и ! (b) культивирование стволовых клеток на внеклеточном матриксе, включающем, по меньшей мере, один фибриллярный белок матрикса, и в свободной от сыворотки культуральной среде, включающей печеночный фактор роста (HGF), ! в котором стволовые клетки, полученные из непеченочных тканей, дифференцируются в печеночные стволовые клетки. ! 2. Способ по п.1, в котором непеченочная ткань представляет собой жировую ткань. ! 3. Способ по п.1, в котором непеченочная ткань представляет собой ткань от взрослого млекопитающего. ! 4. Способ по п.1, в котором, по меньшей мере, один фибриллярный белок матрикса выбирают из группы, состоящей из коллагена I и коллагена III. ! 5. Способ по п.4, в котором, по меньшей мере, один фибриллярный белок матрикса представляет собой коллаген I. ! 6. Способ по п.1, в котором культуральная среда дополнительно включает онкостатин М, DMSO или оба вещества. ! 7. Способ по п.1, в котором внеклеточный матрикс дополнительно включает фибронектин, Матригель, Витроген или их комбинации. ! 8. Способ по п.1, в котором примерно более чем 70% стволовых клеток, полученных из непеченочных тканей, дифференцируются в печеночные стволовые клетки. ! 9. Способ по п.1, в котором стволовые клетки, полученные из непеченочных тканей, дополнительно получают из пассажа в культуре. ! 10. Способ индукции экспрессии HGF из непеченочных стволовых клеток, включающий ! (a) обеспечение стволовых клеток, полученных из непеченочных тканей; и ! (b) культивирование стволовых клеток на внеклеточном матрик�
Claims (13)
1. Способ получения печеночных стволовых клеток из непеченочных стволовых клеток, включающий
(a) обеспечение стволовых клеток, полученных из непеченочных тканей; и
(b) культивирование стволовых клеток на внеклеточном матриксе, включающем, по меньшей мере, один фибриллярный белок матрикса, и в свободной от сыворотки культуральной среде, включающей печеночный фактор роста (HGF),
в котором стволовые клетки, полученные из непеченочных тканей, дифференцируются в печеночные стволовые клетки.
2. Способ по п.1, в котором непеченочная ткань представляет собой жировую ткань.
3. Способ по п.1, в котором непеченочная ткань представляет собой ткань от взрослого млекопитающего.
4. Способ по п.1, в котором, по меньшей мере, один фибриллярный белок матрикса выбирают из группы, состоящей из коллагена I и коллагена III.
5. Способ по п.4, в котором, по меньшей мере, один фибриллярный белок матрикса представляет собой коллаген I.
6. Способ по п.1, в котором культуральная среда дополнительно включает онкостатин М, DMSO или оба вещества.
7. Способ по п.1, в котором внеклеточный матрикс дополнительно включает фибронектин, Матригель, Витроген или их комбинации.
8. Способ по п.1, в котором примерно более чем 70% стволовых клеток, полученных из непеченочных тканей, дифференцируются в печеночные стволовые клетки.
9. Способ по п.1, в котором стволовые клетки, полученные из непеченочных тканей, дополнительно получают из пассажа в культуре.
10. Способ индукции экспрессии HGF из непеченочных стволовых клеток, включающий
(a) обеспечение стволовых клеток, полученных из непеченочных тканей; и
(b) культивирование стволовых клеток на внеклеточном матриксе, включающем, по меньшей мере, один фибриллярный белок матрикса, и в свободной от сыворотки культуральной среде, включающей EGF, bFGF или оба фактора,
в котором стволовые клетки, полученные из непеченочных тканей, продуцируют HGF.
11. Способ по п.10, в котором культуральная среда включает EGF и bFGF.
12. Способ по п.10, в котором культуральная среда дополнительно включает аскорбил-2-фосфат натрия.
13. Способ по п.11, в котором культуральная среда дополнительно включает аскорбил-2-фосфат натрия.
