Claims (7)
- 인터루킨-2(IL-2)의 처음 96 내지 114개의 아미노산에 상응하는 절편 및 이 절편의 C- 또는 N-말단에 위치되는 히루딘 또는 프로인술륜으로 이루어진 융합 단백질을 암호화하는 유전자를 숙주세포에서 발현시킴을 특징으로 하여, 융합 단백직을 제조하는 방법.
- 제 1 항에 있어서, 융합 단백질을 암호화하는 유전자가 하기 일반식(Ia) 또는 (Ib)의 융합 단백질을 암호화 하는 방법.Met-X-Y-Z (Ia)Met-Z-Y-X (Ib)상기식에서, X는 사람 인터루킨-2의 처음 96 내지 114개의 아미노산의 아미도산 서열을 나타내고, Y는 직접 결합을 나타내거나, 유전적으로 암호화 가능한아미노산으로 이루어지고 아미노산 서열 Z를 절단시킬수 있게 하는 연결성분 (bridging element)을 나타내며, Z는 히루딘 또는 프로인슐린의 서열이다.
- 제 2 항에 있어서, Z에 인접한 Y가 Met, Cys, Trp, Arg또는 Lys, 또는 이들 아미노산을 포함하는 아미노산 서열인 방법.
- 제 2 항에 있어서, Z에 인접한 Y가 아미노산 서열 Asp-Pro, 또는 이 아미노산 서열을 포함하는 아미노산 서열인 방법.
- 제 1 항 내지 제 4 항 중의 어느 한 항에 있어서, 융합 단백질이 원심분리에 의해 가용성 단백질로부터 회수되는 방법.
- 제 1 항 내지 제 4 항 중의 어느 한 항에 있어서, 숙주 세포가 세균인 방법.
