알로파노일 피페라진 화합물의 제조방법
[발명의 상세한 설명]
본 발명은 신규의 알로파노일 피페라진 화합물의 제조방법에 관한 것이다.
본 발명에 따른 알로파노일 피페라진 화합물의 하기 일반식(1)로 나타낸다.

상기 식에서,
R1은 저급알킬기 또는 페닐기이며 :
R3는 수소원자 또는 저급알킬기이며 :
R4는 페닐기 또는 치환기로서 할로겐원자 또는 메틸, 트리플루오로메틸, 히드록실, 메톡시, 메틸렌디옥시, 니트로 또는 카르복실기를 갖는 치환된 페닐기 ; 피리딜기, 피리미딜기, 티아조일기, 벤질기, 신나밀기, 시클로헥실기, 저급알킬기, 치환기로서 염소원자 또는 히드록실기를 갖는 치환된 저급알킬기 ; 또는 저급알케닐기이다.
본 발명에 의한 화합물은 문헌에 기술된 바 없는 신규의 화합물이며 진통작용을 갖고 진통제로서 유용할 뿐만 아니라 소염작용을 갖는다. 종래에는 식(1)로 나타내는 알로파노일피페라진 화합물은 말할 것도 없고 하기식으로 나타내는 구조를 갖는 화합물에 대한 기록이 전혀 없었다.

본 발명자는 진통작용을 갖는 화합물의 발전을 위해 상술한 형의 알로파노일-피페라진 화합물에 대해 광범위한 연구를 한 결과 목적하는 바에 적합한 상기 일반식(1)의 화합물을 발견함으로써 본 발명을 완성하였다.
본 발명에 의한 알로파노일피페라진은 다음의 예시한 방법으로 제조한다.
제조 방법
본 방법은 일반식(2)의 이소시아네이트를 일반식(3)의 카바모일 피페라진 화합물과 다음 반응도식에 따라 반응시키는데 특징이 있다. (단, 본 방법은 상기 식(1)에서 R2가 수소원자인 경우에만 적용)

본 발명에 사용된 일반식(2)의 이소시아네이트는 공지화합물로서, 참고문헌 [condenced Chemical Dictionary 9thed 1977 : page 571, Van Nostrandreinhold co.]에 그의 제법 등이 기재되어 있고, 일반식(3)의 카르바모일 피페라진 화합물도 또한 공지화합물로서 참고문헌 [Z. Chem. 21(1) 25∼27 (1981)]에 기재되어 있다.
이소시아네이트(2)와 카르바모일피페라진 화합물(3)의 반응은 일반적으로 촉매의 존재하에서 용매 중에서 수행시킨다. 적당한 촉매는 무수염화 알루미늄, 무수염화 제2주석 및 사염화티타늄과 같은 루이스산이다. 반응에 참가하지 않는 어떠한 용매도 사용할 수 있지마는, 일반적으로 염화메틸렌, 클로로포름 및 사염화탄소와 같은 할로겐화 탄화수소 및 벤젠, 톨루엔 및 크실렌과 같은 방향족 탄화수소가 적당하다. 이소시아네이트(2), 카르바모일 피페라진 화합물(3) 및 촉매 사이의 비는 적당히 선택할 수 있지만, 반응물은 대략 등몰량의 양으로 사용하는 것이 유리하다.
반응온도는 적당하게 선택할 수 있으나, 반응은 일반적으로 -20℃ 내지 실온에서 유리하게 진행된다.
상기 방법에 의해 형성된 알로파노일 피페라진 화합물(1)은 통상의 분리방법으로 용이하게 단리시킬 수 있다.
본 진통제의 1일 복용량은 어른에게 1 내지 4회 분량으로 일반식(1)의 알로파노일 피페라진에 관하여 0.5 내지 1, 000mg이고 1 내지 500mg이 바람직하다. 특별한 경우에 복용량은 환자의 임상특징 및 연령에 따라 적당히 조정한다. 이는 경구투여제, 주사제, 직장용좌제 및 외용제와 같은 여러가지 형태로 투여한다.
본 발명에 의한 진통제는 일반적으로 조성물로서 이를테면, 탄산칼슘, 인산칼슘, 녹말, 슈크로오스, 락토오스, 활석, 마그네슘 스테아레이트, 젤라틴, 폴리비닐피롤리돈, 아라비아고무, 소르비톨, 미소결정질셀룰로오스, 폴리에틸렌글리콜, 카르복시메틸셀룰로오스, 실리카, 폴리비닐아세탈디에틸아미노-아세테이트, 히드록시프로필메틸셀룰로오스 및 셀락과 같은 방법에서 사용되는 부형제를 함유하는 의약용으로 처방된다. 정제는 공지의 방법으로 코우트한다. 경구투여의 액상제제는 일반적으로 공지된 방법으로 조제된 물 또는 오일 중의 현탁액, 용액, 시럽, 엘릭서이다.
주사제로는 물 또는 오일 중의 현탁액 또는 용액. 또는 사용하기 전에 용해시킬 수 있는 미소분말 또는 동결건조된 분말이다. 주사제는 관행의 방법으로 제조한다. 직장용으로, 본 진통제는 이를테면, 폴리에틸렌글리콜, 라놀린, 카카오버터 및 지방산 트리글리세리드와 같은 방법에서 공지된 약학 부형제를 함유하는 좌제조성물로 제공된다.
의용제는 통상의 방법으로 연고제 기제 등에 본 발명에 따른 유효성분을 혼합하여 제조한 연고제 또는 크림제로 사용하는 것이 바람직하다.
본 발명은 일반식(1)로 나타내는 알로파노일피페라진 화합물의 합성실시예, 이와 같이 얻은 화합물의 진통작용에 대한 시험결과 및 동일물을 함유하는 약제의 실시예에 관해서 하기에서 상세히 기술된다.
방법에 의한 일반식(1)의 알로파노일피페라진 화합물의 합성 실시예는 하기에 기술한 바와 같다.
본 실시예 및 본 실시예와 비슷한 방법의 다른 실험으로 얻은 화합물의 특징은 표 1에서 볼 수 있다.
[상기 방법에 의한 합성 실시예 :]
[실시예 1]
(표 1에서 화합물번호 1의 합성)
70ml의 디클로로메탄에 4.1g의 4-페닐피페라진-1-카르복스아미드 및 1.2g의 메틸이소시아네이트를 용해시킨다. 용액에 교반 및 빙냉하면서 5.2g의 염화 제2주석을 점적 가한다. 실온에서 15시간 반응시킨 후, 물을 첨가하여 유기층을 분리한다. 유기층을 무수황산나트륨으로 탈수시키고 용매를 유거한다. 잔기를 에탄올로 재결정하여 융점이 198o∼200℃인 2.7g(수율 : 51%)의 1-(4-메틸알로파노일)-4-페닐피페라진을 얻는다.
[실시예 2]
(표 1에서 화합물번호 12의 합성)
70ml의 디클로로메탄에 4.3g의 1-(메틸카르바모일)-4-(m-트리플루오로메틸페닐) 피페라진 및 0.9g의 메틸이소시아네이트를 용해시킨다. 용액에 교반 및 빙냉하면서 0.3g의 무수염화 제2주석을 점적 가한다. 실온에서 15시간 반응시킨 후, 침전물을 여과로 모으고 디클로로메탄 및 물의 혼합물에서 교반한다. 유기층을 분리하고 무수황산나트륨으로 탈수시킨다. 용매를 유거한 후. 잔기를 에탄올 및 석유 에테르의 혼합물로 재결정하여 3.2g(수율 : 62%)의 1-(2, 4-디메틸알로파노일)-4-(m-트리플루오로메틸페닐)-피페라진을 얻는다.
본 발명의 방법에 의해 제조된 화합물들은 다음 표 1과 같다.

약리시험
진통작용 및 급성독성을 시험하는 방법은 하기에서 기술하고 시험결과는 표 2에서 볼 수 있다.

진통작용 :
1. 초산-유도 신축 방법
Koster 등의 방법 [Fed. Pro., 18, 412 (1959)]에 따라, ddy 계통 수컷생쥐(각각 몸무게 20∼25g)을 시험용으로 사용한다. 100mg/kg의 시험약의 경구투여 1시간 후 0.3ml의 0.6% 초산용액을 각 생쥐의 복강내에 투여하고 신축증후를 관찰하고 억제비율(in%)를 계산한다. 표 2에서 별표(*)가 표시된 숫자를 평균 효력복용량 ED50(mg/kg)이고 NT는 시험되지 않은 것(not tested)를 의미한다.
2. 하프너(Haffner) 방법
ddy 계통 수컷생쥐(각각 몸무제 20∼25g)를 사용하여 후지무라 등의 개량된 방법 [Bulletin of the Institute for Chemical Research, kyoto University, 25, 36(1951)]에 의해 시험한다.
100mg/kg의 시험약의 경구투여 30분 후 역치복용량(2.0mg/kg)의 몰핀히드로클로라이드를 피하에 투여한다. 클램핑에 의한 생쥐의 진통반응을 1시간 동안 관찰하여 억제비율(in%)을 결정한다. 표 2에서, 별표(*)가 표시된 숫자는 평균 효력복용량 [ED50(mg/kg)]이고, NT는 "Not teasted"를 의미한다.
급성 독성 :
급성독성시험은 ddy 계통 수컷생쥐(각각 몸무제 20∼25g)을 사용하여 수행한다. 시험약 경구투여 1주일 후 각 쥐의 일반적인 증후를 관찰한다.
복용량(mg/kg) 및 대응비

는 표 2에서 볼 수 있고, 별표(*)가 표시된 숫자는 평균 치사복용량 [LD50(mg/kg)]이다.
상기 시험중에서, 약은 0.1∼0.25% 카르복시메틸-셀룰로오스 중에서 용액 또는 현탁액으로 사용된다.
결국, 본 발명에 따른 알로파닐피페라진 화합물을 함유하는 약제의 약간의 실시예는 하기에서 주어진다.

  1. 일반식(2)로 표시되는 이소시아네이트 화합물을 일반식(3)로 표시되는 카르바모일피페라진 화합물과 반응시킴을 특징으로 하는 일반식(1)로 표시되는 알로파노일피페라진 화합물의 제조방법.
