Claims (8)
- 단백질 또는 폴리펩티드가 내포되고, 폴리에틸렌글리콜로 이루어지는 친수성 세그먼트와, 산성 아미노산, 그 소수성 유도체 및 산성 아미노산과 그 소수성 유도체의 혼합물로부터 선택되는 폴리아미노산으로 이루어지는 소수성 세그먼트를 가지는 블록 코폴리머를 함유하는 고분자 미셀 조성물.
- 제 1 항에 있어서,상기 산성 아미노산의 소수성 유도체가 산성 아미노산 알킬에스테르 또는 산성 아미노산 알킬아미드인 조성물.
- 제 1 항에 있어서,상기 산성 아미노산이 아스파라긴산 또는 글루타민산인 조성물.
- 제 1 항에 있어서,블록 코폴리머가 하기 화학식 1 또는 화학식 2인 조성물:[화학식 1]또는[화학식 2][상기 각 식 중, R1 및 R3은 각각 독립적으로 수소원자 또는 보호되어 있어도 되는 관능기가 치환된 또는 치환되지 않은 저급 알킬기를 나타내고, R2는 소수원자, 포화 또는 불포화의 C1~C29 지방족 카르보닐기 또는 아릴카르보닐기를 나타내며, R4는 수산기, 포화 또는 불포화의 C1~C30 지방족 옥시기 또는 아릴-저급 알킬옥시기를 나타내고, R5는 -O- 또는 -NH-를 나타내며, R6은 수소원자, 페닐기, -(CH2)4-페닐기, 치환되지 않은 또는 아미노기 또는 카르복시기로 치환된 C4~C16 알킬기, 또는 벤질기를 나타내고, R7은 메틸렌기를 나타내며, n은 10~2500의 정수이고, x는 10~300이 되는 정수이며, m은 0~300이 되는 정수이고(단, m이 존재하는 경우, (COCHNH)의 유닛과 (COR7CHNH)의 유닛은 랜덤하게 존재하고, R6은 1개의 블록 코폴리머 내의 각 아미노산 유닛에서 임의로 선택가능하고, 랜덤하게 존재하는데, R6이 수소원자인 경우에는 R6 전체의 60% 이하임), y는 1 또는 2의 정수를 나타내고, L1은 -NH-, -O-, -O-Z-NH-, -CO-, -CH2-, -O-Z-S-Z- 및 -OCO-Z-NH-(여기서, Z는 독립적으로 C1~C6 알킬렌기임)로 이루어지는 군으로부터 선택되는 연결기를 나타내고, L2는 -OCO-Z-CO- 및 -NHCO-Z-CO-(여기서, Z는 C1~C6 알킬렌기임)로부터 선택되는 연결기를 나타낸다]
- 제 4 항에 있어서,상기 블록 코폴리머가 폴리아미노산 측쇄의 에스테르화 또는 아미드화율 40~100%인 조성물.
- 제 1 항 내지 제 5 항 중 어느 한 항에 있어서,상기 단백질 또는 폴리펩티드의 등전점(pI)이 3~11.5인 조성물.
- 제 1 항 내지 제 6 항 중 어느 한 항에 기재된 고분자 미셀 조성물의 조제방법으로서, 상기 블록 코폴리머와 상기 단백질 또는 폴리펩티드를 혼합하고, 상기 단백질 또는 폴리펩티드의 등전점(pI)과는 다른 pH로 상기 혼합액의 pH를 조정함으 로써 상기 블록 코폴리머로 구성되는 미셀의 소수성 코어영역에 상기 단백질 또는 폴리펩티드를 내포시키는 공정을 포함하여 이루어지고,여기서 상기 단백질 또는 폴리펩티드의 pI, 상기 블록 코폴리머의 소수성 세그먼트 안의 산성 아미노산 및/또는 그 유도체의 등전점(pI') 및 상기 pI와는 다른 pH는,pI>pH>pI'의 관계에 있고, 따라서 상기 pH에서 상기 블록 코폴리머의 소수성 세그먼트는 음으로 대전하고, 상기 단백질 또는 폴리펩티드는 양으로 대전하거나, 또는pI'>pH>pI의 관계에 있고, 따라서 상기 pH에서 상기 블록 코폴리머의 소수성 세그먼트는 양으로 대전하고, 상기 단백질 또는 폴리펩티드는 음으로 대전하는 것을 특징으로 하는 방법.
- 제 7 항에 있어서,상기 pH가 상기 단백질 또는 폴리펩티드의 pI로부터 1 이상 떨어져 있는 방법.
