Description
Claims (11)
- 종양괴사인자를 융합파트너로 이용하여 미생물에서 인성장호르몬을 얻는 제조방법.
- 제 1항에 있어서, 융합파트너는 종양괴사인자 전체 아미노산 서열 또는 그의 서열중 일부가 치환된 종양괴사인자 뮤테인인 것을 특징으로 하는 제조방법.
- 제 2항에 있어서, 융합파트너는 종양괴사인자-α의 N-말단의 8번째에서 12번째에 해당하는 아미노산으로 구성되고 그의 C-말단에 폴리히스티딘 서열을 포함하는 펩타이드인 것을 특징으로 하는 제조방법.
- 제 3항에 있어서, 융합파트너와 결합한 인성장호르몬을 생산하는 대장균 형질전환체를 배양하여 세포를 파쇄하고 단백질 침전체를 얻어 알칼리 용액으로 용해한 다음 칼럼 크로마토그라피를 이용하여 융합단백질을 분리하고 이로부터 인성장호르몬을 정제하는 제조방법.
- 제 4항에 있어서, 분리한 융합단백질을 엔테로키나제로 절단하여 인성장호르몬을 정제하는 제조방법.
- 종양괴사인자-α의 N-말단의 8번째에서 12번째에 해당하는 아미노산으로 구성되고 그의 C-말단에 폴리히스티딘 서열이 첨가된 유전자 서열을 포함하는 발현 벡터.
- 제 6항의 발현 벡터에 엔테로키나제 절단 부위 DDDDK 및 인성장호르몬을 코우딩하는 유전자가 삽입된 발현 벡터.
- 제 7항에 있어서, 발현 벡터 pT7T2GH.
- 제 7항에 있어서, 발현 벡터 pT2GH.
- 제 7항의 발현 벡터로 대장균을 형질전환시켜 얻은 형질전환체.
- 제 10항에 있어서, 발현 벡터 pT2GH 로 대장균 BL21(DE3)을 형질전환시켜 얻은 형질전환체 (수탁번호 : KCTC 8786P)
