Claims (12)
- 프로모터, 리컴비나제 유전자 및 폴리 (A) 서열을 포함하는 동물 세포의 형질 감염용 재조합 아데노바이러스 벡터.
- 제 1 항에 있어서, 상기 리컴비나제 유전자가 대장균 P1 파지로부터 유래하는 리컴비나제 Cre 유전자임을 특징으로 하는 재조합 아데노바이러스 벡터.
- 제 1 항 또는 제 2 항에 있어서, 상기 프로모터 및 폴리 (A) 서열이 사이토메갈로바이러스 인핸서 (enhancer), 닭의β- 액틴 프로모터, 및 토끼β- 글로빈스 플라이싱 (splicing) 억셉터 및 폴리 (A) 서열을 포함하는 하이브리드 프로모터 CAG에 포함됨을 특징으로 하는 재조합 아데노바이러스 벡터.
- 두 개의 리컴비나제 - 인식 서열, 동물 세포에서 작용할 수 있는 복제 기원, 프로모터, 외부 유전자 및 폴리 (A) 서열을 함유하며, 상기 복제 기원, 프로모터, 외부 유전자 및 폴리 (A) 서열 모두가 두 리컴비나제 - 인식 서열의 사이에 위치하는 동물 세포의 형질 감염용 재조합 아데노바이러스 벡터.
- 제 4 항에 있어서, 두 리컴비나제 - 인식 서열중의 하나의 상류 (upstream) 쪽에서 상기 복제 기원, 프로모터, 외부 유전자 및 폴리 (A) 서열이 그 순서대로위치함을 특징으로 하는 재조합 아데노바이러스 벡터.
- 제 4 항에 있어서, 두 리컴비나제 - 인식 서열중의 하나의 상류쪽에서 외부 유전자, 폴리 (A) 서열, 복제 기원 및 프로모터가 그 순서대로 위치함을 특징으로 하는 재조합 아데노바이러스 벡터.
- 제 4 항 내지 제 6 항 중 어느 한 항에 있어서, 상기 리컴비나제 - 인식 서열이 리컴비나제 Cre 의 기질인 loxP 를 코딩하는 DNA 서열임을 특징으로 하는 재조합 아데노바이러스 벡터.
- 제 4 항 내지 제 6 항 중 어느 한 항에 있어서, 상기 복제 기원이 바이러스 또는 동물 세포로부터 유래함을 특징으로 하는 재조합 아데노바이러스 벡터.
- 제 8 항에 있어서, 상기 복제 기원이 파포바바이러스, 헤르페스바이러스, 아데노바이러스, 천연두 바이러스 및 파르보바이러스로부터 유래하는 복제 기원으로 구성되는 군으로부터 선택됨을 특징으로 하는 재조합 아데노바이러스 벡터.
- 제 4 항 내지 제 6 항 중 어느 한 항에 있어서, 상기 프로모터 및 폴리 (A) 서열이 사이토메갈로바이러스 인핸서 (enhancer), 닭의β- 액틴 프로모터, 및 토끼β- 글로빈 스플라이싱 (splicing) 억셉터 및 폴리 (A) 서열을 포함하는 하이브리드 프로모터 CAG 에 포함됨을 특징으로 하는 재조합 아데노바이러스 벡터.
- 하기 단계를 포함하는 것을 특징으로 하는 외부 유전자를 인간 세포를 포함하는 동물 세포로 in vitro 에서 형질 도입하거나 외부 유전자를 인간 세포가 배제된 동물 세포로 in vivo 에서 형질도입하는 방법:프로모터, 리컴비나제 유전자 및 폴리 (A) 서열을 포함하는 재조합 아데노바이러스 벡터, 및 두개의 리컴비나제 - 인식 서열, 동물 세포에서 작용할 수 있는 복제 기원, 프로모터, 외부 유전자 및 폴리 (A) 서열을 포함하며 그 복제 기원, 프로모터, 외부 유전자 및 폴리 (A) 서열 모두는 두 리컴비나제 - 인식 서열 사이에 위치하는 재조합 아데노바이러스 벡터 모두를, 상기 동물 세포로 공 - 형질 감염시키고;상기 복제 기원, 프로모터, 외부 유전자 및 폴리 (A) 서열을 함유하는 DNA 절편을 절단하여 환상의 DNA 분자를 만들고;상기 환상의 DNA 분자를 공 - 형질 감염된 동물 세포 내에서 자가 복제시킨다.
- 유전자 치료에서 사용하기 위해 제 11 항에 따른 형질 감염 방법을 사용하는 것을 포함하는 것을 특징으로 하는, 세포로 인간 유전자를 in vitro 에서 형질 도입하는 방법.
