Claims (14)
- 대장균 라이실-tRNA 합성효소 유전자에 더하여 연결 펩타이드 유전자, 단백질 분해효소 인지부위 유전자, 표지 서열 유전자 또는 제한효소 인지부위를 포함하는 발현 플라스미드.
- 제 1항에 있어서, 라이실-tRNA 합성효소 유전자는lys S유전자인 것을 특징으로 하는 발현 플라스미드.
- 제 2항에 있어서, 발현 플라스미드 pGE-lysRS 인 것을 특징으로 하는 발현 플라스미드.
- 제 1항에 있어서, 아미노 말단은 아미노산 번호 1번에서 13번 또는 29번까지가 제거된 것을 특징으로 하는 발현 플라스미드.
- 제 1항에 있어서, 아미노 말단은 OB 폴드 부분인 것을 특징으로 하는 발현 플라스미드.
- 제 5항에 있어서, OB 폴드 부분은 아미노산 번호 1번에서 65번까지가 제거된 것을 특징으로 하는 발현 플라스미드.
- 제 2항에 있어서, 발현 플라스미드는 pGE-lysN 인 것을 특징으로 하는 발현 플라스미드.
- 제 7항의 발현 플라스미드로 대장균 HMS 174 균주를 형질전환시킨 형질전환체 (수탁번호 : KFCC-10918).
- 제 1항의 발현 플라스미드에 외래 단백질 유전자를 삽입한 발현 플라스미드.
- 제 9항에 있어서, 외래 단백질은 GMcsf 단백질인 것을 특징으로 하는 발현 플라스미드 plysN-GMcsf.
- 제 9항에 있어서, 외래 단백질은 Gcsf 단백질인 것을 특징으로 하는 발현 플라스미드 plysN-Gcsf.
- 제 9항에 있어서, 외래 단백질은 TIMP 2 단백질인 것을 특징으로 하는 발현 플라스미드 plysN-TIMP2.
- 제 9항의 발현 플라스미드로 숙주세포를 형질전환시켜 얻은 형질전환체.
- 제 13항의 형질전환체를 배양하여 단백질의 발현을 유도한 다음, 이로부터 융합 단백질 형태의 수용성 외래 단백질을 대량 생산하는 단백질의 제조방법.
