Abstract
NEW MATERIAL:A monoclonal antibody reactive with carcinoembryonic antigen (CEA), nonspecific cross-reactive antigen (NCA) and biliary glycoprotein I (BGP-1), especially with the CEA. USE:Serological assay of CEA, cytological and histopathological diagnosis of cancer and mutual discrimination of the CEA, NCA and BGP-1, etc. PREPARATION:A mouse is initially immunized with cancerous cells, etc. The amount inoculated is preferably 10<5>-10<8> cells per mouse at a time. The spleen is then extirpated from the mouse and subjected to centrifugation to provide cells capable of producing antibodies. The resultant cells are subsequently fused to cells having the self-propagation ability to afford fused cell, which are then separated by a limiting dilution method. After propagating the separated fused cells, antibodies produced in respective holes (wells) are reacted with various cells, etc., by a fluorescent antibody technique, etc. Sorted fused cell are subsequently cultivated in an incubator to provide the aimed antibody from the supernatant liquid.
