Description

【発明の詳細な説明】 技術分野 本発明は、有効成分としてピロキシカムを含有する消炎
鎮痛外用貼付剤に関する。
従来技術 ピロキシカムは、Pfizer社により開発された薬剤であ
り、その化学名は４−ヒドロキシ−２−メチル−Ｎ−
（２−ピリジル）−2H−1,2−ベンゾチアジン−３−カ
ルボキシアミド−1,1−ジオキシドである。
ピロキシカムは、優れた抗炎症作用および鎮痛作用を有
する非ステロイド性消炎鎮痛剤であるが、一般に非ステ
ロイド性消炎鎮痛剤は、胃腸障害等を始めとする副作用
がある。そこで、症状の軽快化に伴って局所療法に切り
替えることが望ましく、また、軽症あるいは患部が限定
されている疾患においても局所療法が適当である。
軟膏やクリームは局所療法に好適であるが、塗布量の不
明確さや使用時の煩雑さがあり、また、塗布後に衣服等
に付着してしまうという問題があった。
発明の目的 本発明は、局所療法に好適なピロシキカム製剤を提供す
ることを目的とする。
また、本発明は、このようなピロキシカム製剤の皮膚か
らの吸収を高めることを目的とする。
発明の構成 本発明は、ピロキシカムを貼付剤とすることによって、
上記の欠点を補い、薬効の長時間持続性を可能とするも
のである。
しかし、含水率20〜80％の基剤を用いる通常のパップ剤
では、分散媒中での放出性が低く、必要な血中濃度ある
いは薬理効果を必ずしも期待できない傾向がある。そこ
で、本発明者らは更に、ピロキシカムの放出性を高める
観点から粘着性を上げ、しかも放出性を阻害すると考え
られる無機物等を配合しないテープ材（実質的に無水の
粘着剤層中に有効成分を含有する貼付剤）に着目し、ピ
ロキシカムの経皮吸収性を高めるべく鋭意検討を行なっ
た。
さらに、ピロキシカムは、水および通常の媒体には溶け
難く、また溶解性の良い数種の有機溶媒も知られている
が、これらは揮発性であったり、皮膚に適用した場合に
支障があるなどの難点がある。また、ピロキシカムを懸
濁または微粉化して使用することも考えられるが、この
場合には皮膚からの吸収性が悪く効果が十分に発揮され
ないなどの難点がある。
そこで、本発明者らは、ピロキシカムを溶解分散し、経
皮吸収性に優れた貼付剤として製剤すべく鋭意検討した
結果、ピロキシカムを水溶性のアルカリ剤に溶解、分散
させることにより、経皮吸収性を改善しうることを見出
し、本発明を成すに至った。
すなわち、本発明は、ピロキシカムを水溶性のアルカリ
剤に溶解して基剤中に配合し、実質的に無水の粘着剤層
中に含有せしめてなることを特徴とする。
以下、本発明についてさらに詳細に説明する。
本発明の貼付剤は、プラスチックフィルム、不織布など
の支持体上に、ピロキシカムおよび水溶性のアルカリ剤
や他の成分を配合した基剤からなる実質的に無水の粘着
剤層を形成することによって得られ、各成分を配合した
基剤を剥離ライナー上に塗布乾燥したのち塩化ビニルフ
ィルム等の支持体に展着したり、塩化ビニルフィルム等
の支持体上に上記基剤を塗布乾燥したのち剥離ライナー
を張り合わせることにより製造される。
基剤としてはドライ状態で良好な粘着性を得られるもの
が用いられる。粘着性基剤としては、アクリル酸エステ
ル系のポリマー、コポリマーなどのアクリル樹脂；天然
ゴム、スチレン−ブタジエン共重合体、シリコンゴム等
の合成ゴムなどのゴム弾性基剤；石油樹脂、ロジン、水
添ロジン、エステルガム等の樹脂類；ポリブテン、流動
パラフィン、イソプロピルミリステートの如き高級エス
テル類等の軟化剤などが挙げられる。
ピロキシカムの配合量は特に限定されないが、0.1〜2.0
重量％程度が好ましい。
基剤中にピロキシカムに加えて水溶性のアルカリ剤を配
合することにより、ピロキシカムの経皮吸収性が向上す
る。水溶性のアルカリ剤としては、ジイソプロパノール
アミン、ジエタノールアミン等の有機アミン、Ｌ−アル
ギニン、Ｌ−リジン、水酸化ナトリウム、アンモニア等
が用いられる。アルカリ剤は、ピロキシカム１重量部に
対して0.5〜10重量部配合することが好適である。
また、ピロキシカムにアルカリ剤を添加し、更にベンジ
ルアルコール、クロタミトン（クロトノイル−Ｎ−エチ
ル−Ｏ−トルイジン）、プロピレングリコール、ハッカ
油、ポリエチレングリコール、エチレングリコールモノ
エチルエーテル等の溶剤に再溶解分散させ、さらに必要
に応じてポリオキシエチレンソルビタンモノステアレー
ト等の界面活性剤を添加し、基剤と均一に練り合わせる
ことにより貼付剤を得ることができる。上記溶剤は、ピ
ロキシカム１重量部に対し、０〜20重量部用いることが
好適である。
アジピン酸ジイソプロピル、ジエチルセバケート、オレ
イン酸等の浸透剤を配合することもできる。
また、水および／またはアルコールに可溶なメチルセル
ロース、ヒドロキシエチルセルロース、ヒドロキシプロ
ピルセルロース、カルボキシメチルセルロースナトリウ
ム、ポリビニルピロリドン、ポリアクリル酸ナトリウ
ム、ポリビニルアルコール、カルボキシビニルポリマ
ー、アルギン酸ナトリウム、アルギン酸プロピレングリ
コール等を加えることができる。
各成分を配合した基剤は、pH4.0〜9.0の範囲となるよう
に調製することが好ましく、これにより皮膚刺激をなく
することができる。
発明の効果 本発明によれば、ピロキシカムを有効成分として用い実
質的に無水の粘着剤層中に配合して貼付剤とすることに
より、被服等への付着もなく手軽に使用でき、塗布量の
管理も容易で、しかも薬効を長期にわたって持続させる
ことができる。
しかも、ピロキシカムをアルカリ剤に溶解して貼付剤の
基剤中に配合することにより、経皮吸収性を改善するこ
とができる。
実施例１ 下記組成物を均一に混合した後、剥離ライナー上に塗
布、乾燥し、塩化ビニルフィルムに展着して本発明の貼
付剤を得た。
さらに、実施例１と同様にして、下記組成で実施例２〜
５の貼付剤を作成した。
実施例２ 実施例３ 実施例４ 実施例５ 実施例６ 以上のようにして得られた本発明の実施例１の貼付剤に
ついて経皮吸収試験および薬理試験を行った。
経皮吸収試験 体重200g前後のWister系ラット（雄）を、前日に電気バ
リカンで腹部除毛後に、実施例１の貼付剤を貼付し、一
定時間貼付後に腹大動脈より採血した血清を用い、高速
液体クロマトグラフ法により血中濃度を測定し、その結
果を表−１に示した。
また、合わせて下記の比較例１の貼布剤、および下記組
成の通常パップ基剤（参考例）を用いたものについても
同様に経皮吸収試験を行ない、これらの結果を表−１に
示した。
比較例１ 下記組成物を均一に混合した後、剥離ライナー上に塗
布、乾燥し、塩化ビニルフィルムに展着して比較用の貼
付剤を得た。なお、この貼布剤は実施例１の本発明の貼
布剤からジイソプロパノールアミン（アルカリ剤）を除
いた以外は、実施例１の貼布剤と同じである。
パップ剤組成 薬理試験（紫外線紅斑抑制試験） 体重250〜300gのモルモット（雄）を１群５匹として用
いた。
実施前日にモルモットの腹部を脱毛し、脱毛腹部に実施
例１のピロキシカム貼付剤を貼付した後、伸縮性包帯で
被覆保護し、貼付１時間後に常法により紫外線を照射
し、照射２時間後、５時間後の紅斑の程度を３段階で評
価した。
１点：照射部位が明瞭な境界をもったもの 0.5点：境界不明瞭な紅斑 ０点：ほとんど紅斑を認められないもの

  1. 【請求項１】有効成分として下記構造式で表されるピロ
    キシカム〔４−ヒドロキシ−２−メチル−Ｎ−（２−ピ
    リジル）−（2H−1,2−ベンゾチアジン−３−カルボキ
    シアミド−1,1−ジオキシド〕を、水溶性のアルカリ剤
    に溶解して基剤中に配合し実質的に無水の粘着剤層中に
    含有せしめてなることを特徴とする消炎鎮痛外用貼付
    剤。
