Description
Claims (11)
- 脱保護ステップを実施するための少なくとも1つの脱保護ユニットと、カップリングステップを実施するための少なくとも1つのカップリングユニットと、酸化又はチオレーションステップを実施するための少なくとも1つの酸化/チオレーションユニットと、洗浄ステップを実施するための少なくとも1つの洗浄ユニットとを含む装置を使用する核酸合成方法であって、核酸合成のための複数の反応容器が、所望の核酸配列の合成スキーム(synthesis scheme)に従って前記ユニットに移動され、少なくとも2つの反応容器が、一列に(in series)並べられ、いくつかの前記ユニットに同時に作用され、複数の前記ユニットのうちの1つから前記少なくとも2つの反応容器のうちの第1の反応容器に導入された材料は、その後、前記第1の反応容器から、前記少なくとも2つの反応容器のうちの第2の反応容器に受け取られる、核酸合成方法。
- それぞれの前記ユニットが1種類の核酸又は処理用の化学物質に対してのみ使用される、請求項1に記載の核酸合成方法。
- アミダイトが、反応容器への1回の添加において、3:1を超える化学量論的条件で添加される、請求項1に記載の核酸合成方法。
- 全ての反応容器が同時に移動する、請求項1に記載の核酸合成方法。
- 選択されたユニットにおいて、前記反応容器は移動するが、同じユニットと流体を流通可能なままである、請求項1に記載の核酸合成方法。
- 前記カップリングユニットに使用される材料はホスホロアミダイト(phosphoramidite)であり、前記ホスホロアミダイトが1.5~3.0当量(equivalents)の量で1つの反応容器に添加される、請求項1に記載の核酸合成方法。
- 前記カップリングユニットに使用される材料は活性剤(activator)であり、前記活性剤は、前記合成スキームの異なるポイントで異なる濃度で添加され、前記反応容器及び前記洗浄ユニットで使用される材料は、前記合成スキームの異なるポイントで異なる濃度で添加される、請求項1に記載の核酸合成方法。
- ユニットで反応容器から回収された材料が、前記合成スキームにおける後の反応容器の反応又は処理のために、前記ユニットにおいて再使用される、請求項1に記載の核酸合成方法。
- 前記反応容器内の反応生成物が、いくつか又は全てのユニットで評価され、不満足な結果を示す反応容器は合成プロセス(synthesis procedure)から除去される、請求項1に記載の核酸合成方法。
- 選択された核酸配列を構築するための合成スキームに従って、複数の反応容器がコンベヤ装置を介して前記ユニットに移動され;
少なくとも1つのユニットが、他のユニットと比較して最長の時間(R)を必要とする反応を行い;
前記ユニットが並列で(in parallel)運用され;
前記反応容器の少なくとも大部分は、設定された時間(T)に従って前記ユニット間で一斉に移動され、当該反応容器は、Tの倍数の時間、R未満の反応時間のユニットに留まる、
請求項1に記載された、核酸合成方法。 - 前記反応容器が、それぞれのステーションに少なくとも1つの反応容器が関連付けられるまで、Tの分数である反応時間の期間の後に移動されるスタートアップモードを含み、前記スタートアップモードの後に、複数の反応容器は1つ以上のステーションで一列に連結され、当該反応容器は、Tに等しい時間又はTに近づく時間で移動する、請求項10に記載の核酸合成方法。
