Description
本発明の一側面は、血管新生抑制活性を有する新規ペプチドを提供する。
前記ペプチドは、ペプチド結合で連結された2個以上のアミノ酸からなるポリマーを意味するところ、ペプチド自体の大きさがあまりにも大きいため、標的組織または細胞に効果的に流入されることができないか、半減期が短くて短期間に体内で消滅するという短所があるため、本発明の前記ペプチドは、血管新生抑制活性を有する、20個以下、好ましくは15個以下、さらに好ましくは10個以下のアミノ酸からなる。
本発明のまた他の側面は、配列番号1または配列番号2のアミノ酸配列を含むペプチドを有効成分として含む血管新生関連疾患の予防または治療用薬学的組成物を提供する。
本発明のまた他の側面は、配列番号1または配列番号2のアミノ酸配列を含むペプチドを有効成分として含有する血管新生関連疾患の予防または改善用健康機能食品を提供する。
但し、下記実施例及び実験例は本発明を具体的に例示するものであり、本発明の内容が下記実施例及び実験例により限定されない。
自動ペプチド合成基(Milligen 9050、Millipore、米国)を用いて、下記表1に記載された配列番号1のアミノ酸配列を有するペプチド及び配列番号2のアミノ酸配列を有するペプチドをそれぞれ合成し、C18逆相高性能液体クロマトグラフィー(HPLC)(Waters Associates、米国)を用いて、これら合成されたペプチドをそれぞれ純粋分離した。カラムは、ACQUITY UPLC BEH300C18(2.1mm×100mm、1.7μm、Waters Co,米国)を用いた。
*血管内皮細胞の増殖抑制効果を確認するために、血管内皮細胞に前記製造例1で製造されたペプチドをそれぞれ処理した後、それぞれMTT分析を行った。
血管内皮細胞の分化抑制効果を確認するため、血管内皮細胞に前記製造例1で製造された2種のペプチドをそれぞれ処理した後、脈管形成分析(tube formationassay)を行った。
Claims (10)
- 配列番号1または配列番号2のアミノ酸配列のみからなる血管新生(angiogenesis)抑制用ペプチド。
- 前記ペプチドのN-またはC-末端は、アセチル基、フルオレニルメトキシカルボニル基、ホルミル基、パルミトイル基、ミリスチル基、ステアリル基、及びポリエチレングリコール(PEG)よりなる群から選択される保護基と結合されている、請求項1に記載の血管新生抑制用ペプチド。
- 前記ペプチドは、血管内皮細胞増殖、分化及び移動を抑制する、請求項1に記載の血管新生抑制用ペプチド。
- 前記ペプチドは、VEGF(vascular endothelial growth factor)受容体と結合する、請求項1に記載の血管新生抑制用ペプチド。
- 前記VEGF受容体は、VEGFR2(Vascular endothelial growth factor receptor 2)である、請求項4に記載の血管新生抑制用ペプチド。
- 前記ペプチドは、VEGF受容体に対してVEGFと競争的に結合する、請求項1に記載の血管新生抑制用ペプチド。
- 請求項1に記載のペプチドを有効成分として含有する、血管新生関連疾患の予防または治療用薬学的組成物。
- 前記組成物は、血管内皮細胞増殖、分化及び移動を抑制する、請求項7に記載の血管新生関連疾患の予防または治療用薬学的組成物。
- 前記血管新生関連疾患は、癌、糖尿病性網膜症、未熟児網膜症、新生血管緑内障、紅皮症、増殖性網膜症、滲出型黄斑変性症、乾癬、血友病性関節、アテローム性動脈硬化プラーク内における毛細血管の増殖、ケロイド、肉芽創、血管接着、リウマチ関節炎、骨関節炎、自己免疫疾患、クローン病、再狭窄、アテローム性動脈硬化、ネコ引っかき病、潰瘍、糸球体腎炎、糖尿病性腎症、悪性腎硬化症、血栓性微小血管症及び糸球体腎症からなる群から選択される何れか一つ以上である、請求項7に記載の血管新生関連疾患の予防または治療用薬学的組成物。
- 請求項1に記載のペプチドを有効成分として含有する血管新生関連疾患の予防または改善用健康機能食品。
