内部標準物質としてp−トルイル酸0.1gを採取し、次いで反応液0.3gを採取し、メタノールで均一に溶解した後、条件の整ったHPLCに注入した。得られたHPLCチャートからp−ヒドロキシ桂皮酸とp−トルイル酸のピーク面積比、およびp−ヒドロキシスチレンとp−トルイル酸のピーク面積比を読み取った。別途標準物質を用いて作成した検量係数を用い、p−ヒドロキシ桂皮酸の残存量およびp−ヒドロキシスチレンの生成量を計算した。
内部標準物質としてp−トルイル酸0.1gを採取し、次いでp−ヒドロキシ桂皮酸0.03gを採取し、メタノールで均一に溶解した後、条件の整ったHPLCに注入した。得られたHPLCチャートからp−ヒドロキシ桂皮酸とp−トルイル酸のピーク面積比を読み取った。p−ヒドロキシ桂皮酸とp−トルイル酸のピーク面積比と重量比の相関係数、すなわち検量係数を算出した。
内部標準物質としてp−トルイル酸0.1gを採取し、次いでp−アセトキシスチレン0.05gを採取し、48%水酸化ナトリウム0.2gおよびメタノール10mlを加えて室温で30分撹拌することで、p−アセトキシスチレンを加水分解させ、定量的にp−ヒドロキシスチレンを生成させた。p−ヒドロキシスチレンの生成量はp−アセトキシスチレンの採取量から100%の収率で生成する量として算出した。この混合物をメタノールで均一に溶解した後、条件の整ったHPLCに注入した。得られたHPLCチャートからp−ヒドロキシスチレンとp−トルイル酸のピーク面積比を読み取った。p−ヒドロキシスチレンとp−トルイル酸のピーク面積比と重量比の相関係数、すなわち検量係数を算出した。
攪拌機、温度計、還流冷却器を備えた50mlフラスコにp−ヒドロキシ桂皮酸3.28g(20mmol)、重合禁止剤としてp−メトキシフェノール0.12g(1mmol)、および溶媒として水分率を3重量%に調整した含水N,N−ジメチルホルムアミド20mlを仕込み、撹拌下、150℃に温調したオイルバスに浸して加熱した。温度上昇に伴い還流が観察され、反応液の温度は136℃に達した。3時間加熱を継続した後、速やかに冷却し、反応混合物を、HPLCを用いて上述の方法で分析したところ、p−ヒドロキシ桂皮酸が0.545g(3.32mmol)残存し、p−ヒドロキシスチレンが1.929g(16.06mmol)生成し、脱炭酸反応成績は転化率83.4%、収率80.3%であった。
実施例1において溶媒を3%含水N,N−ジメチルホルムアミドから、種々のアルコール類とN,N−ジメチルホルムアミドを含む混合溶媒、および3%含水N,N−ジメチルアセトアミドに変えた以外は同様に実施した。脱炭酸反応成績を表1に示した。
実施例1において溶媒を3%含水N,N−ジメチルホルムアミドから、種々の単独溶媒、および3%の水を含むアミド結合非含有溶媒に変えた以外は同様に実施した。脱炭酸反応成績を表2に示した。
実施例1において含水N,N−ジメチルホルムアミドの水分率を変化させた以外は同様に実施した。脱炭酸反応成績を表3に示した。
攪拌機、温度計、還流冷却器を備えた50mlフラスコにp−ヒドロキシ桂皮酸9.85g(60mmol)、重合禁止剤としてp−メトキシフェノール0.12g(1mmol)、および溶媒として水分率を3重量%に調整した含水N,N−ジメチルホルムアミド20mlを仕込み、撹拌下、150℃に温調したオイルバスに浸して加熱した。4時間加熱を継続した後、HPLCを用いて上述の方法で反応液を分析し、原料のp−ヒドロキシ桂皮酸が消失していることを確認した。次いで反応液をトルエン50gで希釈し、50gの水を加えて良く撹拌した後、水層と油層を分離した。油層をさらに25gの水で洗浄し、水層と油層を分離した。油層から溶媒を減圧留去することにより黄褐色固体6.97gを得た(理論収量の97%)。HPLCを用いて上述の方法で化学純度を分析したところ、p−ヒドロキシスチレンの化学純度は96.2%(面積百分率)であった。
攪拌機、温度計、還流冷却器を備えた100mlフラスコに4−ヒドロキシ−3−メトキシ桂皮酸23.30g(120mmol)、重合禁止剤としてp−メトキシフェノール0.15g(1.2mmol)、および溶媒として水分率を3重量%に調整した含水N,N−ジメチルホルムアミド40mlを仕込み、撹拌下、150℃に温調したオイルバスに浸して加熱した。4時間加熱を継続した後、HPLCを用いて上述の方法で反応液を分析し、原料の4−ヒドロキシ−3−メトキシ桂皮酸が消失していることを確認した。次いで反応液をトルエン50gで希釈し、100gの水を加えて良く撹拌した後、水層と油層を分離した。油層から溶媒を減圧留去することにより黄色液体16.95gを得た(理論収量の94%)。HPLCを用いて上述の方法で化学純度を分析したところ、化学純度は96.8%(面積百分率)であった。得られた化合物はガスクロマトグラフィ−質量分析法により4−ヒドロキシ−3−メトキシスチレンと同定した。図1にガスクロマトグラフィ−質量分析法で得られた、主生成物の質量スペクトルを示す。
Claims (4)
- 下記の一般式(1)
(式中、R1、R2、R3およびR4は、互いに独立して、H、OH基、または、OCH3基のいずれかである)で示されるヒドロキシ桂皮酸誘導体を、水および/またはアルコール類の存在下、少なくとも1種のアミド結合含有溶媒を含む溶媒中で、脱炭酸反応させる下記の一般式(2)
(式中、R1、R2、R3およびR4は、互いに独立して、H、OH基、または、OCH3基のいずれかである)で示されるスチレン誘導体の製造方法であって、水および/またはアルコール類の使用量が、水および/またはアルコール類と、少なくとも1種のアミド結合含有溶媒を含む溶媒を合計した量に対して、10重量%以下であるスチレン誘導体の製造方法。 - 脱炭酸反応における反応温度が100℃以上の温度である請求項1に記載のスチレン誘導体の製造方法。
- アミド結合含有溶媒が、N,N−ジメチルホルムアミド、または、N,N−ジメチルアセトアミドから選ばれる少なくとも1種の溶媒である請求項1または2に記載のスチレン誘導体の製造方法。
- ヒドロキシ桂皮酸誘導体が、p−ヒドロキシ桂皮酸、または、4−ヒドロキシ−3−メトキシ桂皮酸である請求項1〜3のいずれかに記載のスチレン誘導体の製造方法。
