アルミ箔
1.透過性アッセイが行われる2週間前に、コラーゲンコーティングしたウェルインサート上にCaco−2細胞を播く。100%エタノールとコラーゲンの体積比を1:1にすることによって、コラーゲンコーティングプレートを作製する。無菌フード内で表面を乾燥するまで一晩乾燥させる。
2.フェノールレッド不含α−MEM培地において対象のFITC共役分子(BSA、IgGなど)の溶液を0.1mg/mL作る。アルミ箔にくるんで光から保護する。
3.抵抗を測定することによって、Caco−2細胞の密集度をチェックする。密集度に対して抵抗は約〜600Ωであるべきである。
4.頂端(アピカル)側及び基底外側においてセルカルチャーインサートから古い培地を吸引する。PBSでリンスして、残りのフェノールレッド色素を除去する。
5.各インサートの頂端側において0.5mLのFITC共役溶液を加える。
6.セルカルチャーインサートを備えた別の24ウェルプレートにおいて、0.5mLの加温したPBSを加える。
7.インサートをPBSの付着したプレートに移す。キムワイプでインサートの底部を拭き取ることにより、残りのフェノールレッドを除去する。
8.t=0の時点:インサートの基底外側から75mLを採取し、底部が黒色の96ウェルプレートに移す。上記体積を75mLの加温したPBSに置き換える。「チョップスティック型」電極を用いて各ウェルの抵抗を記録する。
9.メンブレンを適切にラベルを付けたウェルに注意深く加える。対照は、インプリントされていないメンブレン及び細胞単独である。顕微鏡下で、メンブレンが細胞と直接接触することをチェックする。細胞との接触を示すはっきりした円を見ることができるはずである。
10.t=0の時点:ステップ7を繰り返し、その後インキュベータ内に1時間載置する。
11.t=1の時点:ステップ7を繰り返し、その後インキュベータ内に1時間載置する。
12.t=2の時点:ステップ7を繰り返す。
13.分光蛍光光度計プレートリーダーを用いて蛍光シグナルを測定する。FITC(励起=490nm、発光=520nm)
使用したフィルム及び得られた結果を要約したものを下表6に示す。
Claims (23)
- 関節リウマチ薬を皮膚バリアを透過して送達するためのデバイスであって、
チャンネルを含むマイクロニードルのアレイと、
少なくとも1つの前記マイクロニードルの外表面上に形成されかつ互いに離間して所定のパターンで配列された複数のナノ構造体と、
前記マイクロニードルの前記チャンネルに流体連通する貯蔵部と、
前記貯蔵部に保持された関節リウマチ薬とを含むことを特徴とするデバイス。 - 前記所定のパターンが、複数のマイクロ構造体をさらに含み、
前記ナノ構造体が、前記マイクロ構造体よりも小さい断面寸法を有することを特徴とする請求項1に記載のデバイス。 - 前記マイクロ構造体の断面寸法より小さくかつ前記複数のナノ構造体の断面寸法より大きい断面寸法を有する複数の別のナノ構造体をさらに含むことを特徴とする請求項2に記載のデバイス。
- 前記複数のナノ構造体のうちの少なくとも一部のナノ構造体が、約5nm〜500nmの断面寸法を有することを特徴とする請求項1ないし請求項3のいずれか1項に記載の医療デバイス。
- 前記複数のナノ構造体のうちの少なくとも一部のナノ構造体が、約50nm〜1μmの中心間距離を有することを特徴とする請求項1ないし請求項4のいずれか1項に記載の医療デバイス。
- 前記複数のナノ構造体のうちの少なくとも一部のナノ構造体が、約10nm〜20μmの高さを有することを特徴とする請求項1ないし請求項5のいずれか1項に記載の医療デバイス。
- 前記複数のナノ構造体のうちの少なくとも一部のナノ構造体が、約0.15〜30のアスペクト比を有することを特徴とする請求項1ないし請求項6のいずれか1項に記載の医療デバイス。
- 前記関節リウマチ薬が、疾患修飾性抗リウマチ薬であることを特徴とする請求項1ないし請求項7のいずれか1項に記載のデバイス。
- 前記関節リウマチ薬が、タンパク質治療薬であることを特徴とする請求項8に記載のデバイス。
- 前記関節リウマチ薬が、TNF−α阻害薬またはIL−1阻害薬であることを特徴とする請求項9に記載のデバイス。
- 前記関節リウマチ薬が、抗炎症薬であることを特徴とする請求項1ないし請求項8のいずれか1項に記載のデバイス。
- 前記関節リウマチ薬が、鎮痛薬であることを特徴とする請求項1ないし請求項8のいずれか1項に記載のデバイス。
- 関節リウマチ薬を皮膚バリアを透過して送達するためのデバイスを形成する方法であって、
マイクロニードルアレイを作製するステップと、
前記マイクロニードルのうちの少なくとも1本のマイクロニードルの外表面上に互いに離間して配置される複数のナノ構造体のパターンを作製するステップと、
貯蔵部を前記マイクロニードルに流体連通するように前記マイクロニードルと関連付けるステップと、
前記貯蔵部内に間接リウマチ薬を保持させるステップとを含むことを特徴とする方法。 - 前記貯蔵部が、前記マイクロニードルアレイが配置されている基板に取り付けられていることを特徴とする請求項13に記載の方法。
- 前記貯蔵部が、前記マイクロニードルアレイに取り外し可能に結合可能であることを特徴とする請求項13に記載の方法。
- 前記パターンが、1より大きいフラクタル次元を有することを特徴とする請求項1に記載のデバイス。
- 複数の前記ナノ構造体の少なくとも一部の断面寸法が100〜300nmであることを特徴とする請求項1に記載のデバイス。
- 複数の前記ナノ構造体は、互いに概ね等しい断面寸法を有することを特徴とする請求項1に記載のデバイス。
- 互いに隣接する2つの前記ナノ構造体の平均断面寸法の、その2つの前記ナノ構造体の中心間距離に対する比は、1:1から1:4であることを特徴とする請求項1に記載のデバイス。
- 前記ナノ構造体間の距離は、少なくとも一部において等距離であることを特徴とする請求項1に記載のデバイス。
- 複数の前記ナノ構造体の少なくとも一部は、複数のピラーによって形成されたことを特徴とする請求項1に記載のデバイス。
- 前記チャンネルの断面寸法は、1〜100μmであることを特徴とする請求項1に記載のデバイス。
- 前記チャンネルの長さは、約10〜800μmであることを特徴とする請求項22に記載のデバイス。
