Abstract
Description
(1)優れたFGF−7産生促進作用を有し、天然物由来の安全性の高いFGF−7産生促進剤が得られる。
(2)100万ダルトン以上のプロテオグリカンを選択することにより安定した作用効果が得られる。
(3)優れた毛乳頭細胞増殖促進作用を有し、天然物由来の安全性の高い毛乳頭細胞増殖促進剤が得られる。
(4)毛乳頭細胞の増殖が促進されると共に、増殖した毛乳頭細胞それぞれのFGF−7産生量が促進されるFGF−7産生促進剤及び毛乳頭細胞増殖促進剤が得られる。
1.毛乳頭細胞増殖試験
ヒト毛乳頭細胞(白色人種22歳男性、頭頂部由来、製品番号:CA60205a、東洋紡績株式会社製品)を1ウェルあたり、1.2×104細胞/0.3mLとなるように48ウェル培養プレート(製品番号:677180、株式会社グライナー・ジャパン製品)に播種した。なお、培地は、毛乳頭細胞専用培地(製品番号:TMTPGM−250、東洋紡績株式会社製品)を用いた。CO2インキュベーター内(5%CO2、37℃)で1日培養して50%コンフルエントとした後、以下表1に示す被験物質を含む培地に置換した。
2.FGF−7産生試験
実施例1において回収した培養1日目及び培養3日目の各試験区の培養上清を用い、この培養上清中に含まれているFGF−7量を測定した。FGF−7量の測定はFGF7Human ELISA Kit(製品番号:ab100519、アブカム株式会社製品)を用いて行った。得られた測定値に関し、比較試験区間での有意差検定を行った。検定はstudentのT検定として行い、P<0.05(帰無仮説が5%未満)のものを有意差ありと判断し、図中に*印にて記した。培養1日目の結果を以下表4及び図3に、培養3日目の結果を以下表5及び図4に示す。なお、図及び表中の「サンプル濃度」とは、プロテオグリカンの濃度を示している。

- 分子量100万〜140万ダルトンの鮭鼻軟骨由来のプロテオグリカンを含有することを特徴とする毛乳頭細胞のFGF−7産生促進剤。
