Description
化学反応のために熱の伝導を制御する装置は、核酸の部分を増幅する熱サイクルPCRのような合成反応に適用できる。この系統的分類法では、DNAの鋳型(テンプレート)が、熱的に安定なDNAポリメラーゼ、例えば、TaqDNAポリメラーゼ、ヌクレオチド三燐酸塩、および異なる配列をもつ2つのオリゴヌクレオチドとともに使用されて、鋳型DNAの対向する鎖上にある配列を補間し、増幅されるべきDNAの部分の側面に位置する(「プライマー」)。反応成分は高温(例えば、95℃)と、次のより低い温度(プライマーのアニーリングに対しては40〜60℃、重合に対しては70〜75℃である。)との間でサイクルする。脱交雑温度とアニール温度と重合温度との間で繰返されるサイクルは、鋳型DNAの指数関数的な増幅をもたらす。
図1は本発明による熱交換反応チャンバの内部を示し、熱交換反応チャンバは化学反応のための試料を収容するようになっている。この反応チャンバは、最良の熱伝導性および試料の有効な光学的観察のために設計されている。この薄肉形状は、熱伝導のために大きな表面を与えることによって最適な熱反応速度に貢献している。表面は温度調節要素と密接するようになっている。さらに、小さな壁または大きな壁が反応チャンバ内に窓を備えて、反応試料の全てが光学的に検出され得る。加えて、この反応チャンバは広範囲の反応容積に適する。
反応容器は、反応チャンバ内の混合物を加熱または冷却するための熱スリーブと和合できる。熱スリーブは、反応チャンバの壁と密接させることによって、反応チャンバ内の温度変化を引き起こすように設計されている。
光学アセンブリは、熱スリーブを受け入れるように形成されたユニットの中に作られ得る。このユニットは、付加的に、冷却システムのような環境温度を維持するためのシステムと、熱交換反応チャンバ内で行なわれる作業を調整するための様々な制御機構とを有し得る。
このような付加的な反応は、例えばポリメラーゼ連鎖反応は、熱的に循環する。或いは、付加的な反応は、例えば核酸配列をベースにした増幅(NASBA)は、単一の温度で行なわれる。さらに、このような反応は、広範で様々な増幅試薬と酵素を使用し、とりわけ、DNAリガーゼ,T7RNAポリメラーゼおよび/または逆転写酵素を含んでいる増幅試薬と酵素を使用する。加えて、ポリヌクレオチドの変性は、既知の化学的または物理的方法によって、単独あるいは熱的変化と組合せて達成される。この発明の装置で実施され得るポリヌクレオチド増幅反応は、限定するものではないが、(1)自立配列複製(3SR)および鎖置換増幅(SDA)のような標的ポリヌクレオチド増幅方法、(2)「有枝鎖」DNA増幅のような標的ポリヌクレオチドに取り付けられた信号の増幅に基づいた方法、(3)リガーゼ連鎖反応(LCR)およびQBレプリカーゼ増幅(QBR)のようなプローブDNAの増幅に基づいた方法、(4)配位子結合活性転写(LAT)および核酸配列ベース増幅(NASBA)のような転写ベースの方法、(5)修復連鎖反応(RCR)および循環プローブ反応(CPR)のような様々な他の増幅方法を含んでいる。
Claims (26)
- a)2つの対向する主壁を備え、少なくとも1つの上記主壁がシートまたはフィルムからなり、上記シートまたはフィルムは、加熱表面または冷却表面の形状に一致するのに充分な可撓性を有し、
b)反応チャンバを形成するために、上記主壁を互いに接合する複数の副壁を有する剛性フレーム備え、上記反応チャンバに光学的窓を提供するために、少なくとも2つの上記副壁は光透過性であり、
c)上記反応チャンバ内に流体を導入するためのポートを備えていることを特徴とする反応容器。 - 請求項1に記載の反応容器において、

上記副壁の表面積に対する上記主壁の表面積の比が、少なくとも2:1であることを特徴とする容器。 - 請求項13乃至25のいずれか1つに記載の容器において、
上記反応チャンバ内に試薬をさらに備えていることを特徴とする容器。
