Description
本発明は、炎症性状態/障害、とりわけ喘息、COPDおよび鼻炎のような呼吸疾患の処置における使用のためのホルモテロールまたはホルモテロールのエナンチオマー、グルココルチコステロイドおよび担体または希釈剤を含む安定な粉末製剤に関する。
安定性は、化合物または化合物の混合物が治療上有用な医薬製品へと発展し得るかどうかを決定する最も重要な要因の1つである。医薬製剤において異なる成分を混合する場合に、構成成分の間で起こる相互作用の可能性が存在する。さらに、各構成成分は異なる分解特性を有し得る。
本発明にしたがって、混合物中に、微粉化された(micronised)ホルモテロールまたはそのエナンチオマーである第1の活性成分、微粉化されたグルココルチコステロイドである第2の活性成分および担体または希釈剤を含む固体の状態の医薬組成物が提供され、該組成物は高い保存安定性を有する。
1. 微粉化された第1の活性成分および微粉化された担体/希釈剤の混合物を調製すること
2. 所望により、さらに混合物に微粉化された担体/希釈剤を加えること
3. 前もって微粉化された疎水性の第2の活性成分を添加および混合すること(第2の活性成分は、所望により、微粉化された担体/希釈剤と前もって混合されていてもよい。)および
4. 混合物を凝集化および球状化するか、または粗大な担体/希釈剤を加えること、
が含まれる。
本発明は下記の実施例により例示されるが、これらは本願の範囲を限定することを意図するものではない。実施例において、微粉化は各活性成分の粒子径の範囲が吸入による投与に適するように実施される。ホルモテロールの分解生成物の測定は、LiChrospher 60 RP-select Bを用いる2カラムシステムの逆相液体クロマトグラフィーにより実施された。固定相としてオクチルシランを有する5μmの粒子。UV検出器は214nm。分解生成物がよく分からないので、評価は面積比(area-%)で行った。
下記の実施例は、製剤を通常の方法で製造する参考実施例である。
フマル酸ホルモテロール二水和物(26g)およびラクトース一水和物(4.974kg)を回転型混合機(tumbling mixer)中で1または2時間混合する。この混合物を、吸入に適した粒子径を得るためにスパイラルジェットミル中で微粉化した。低ミクロン範囲(1〜5μm)への物質の微粉化により、該物質の結晶化が阻害され得る。アモルファスの領域が、特に微粉化された物質の表面に導入され得る。物質のこの形態的変化は湿度に対する感受性を増大させ、それは安定性の問題に対する手段となり得る。物質の混合物の結晶構造は制御された方法で米国特許第5.874.063号または米国特許第5.709.884号にしたがって再現された。
凝集性の粉末の流動性を改善するために、それを室温にて50%未満の制御された相対湿度で球状化して凝集体とした。
下記の実施例は、製剤を通常の方法で製造する参考実施例である。
微粉化され、球状化された実施例1のフマル酸ホルモテロール二水和物/ラクトース一水和物製剤を粉末吸入器Turbuhaler(登録商標)(AstraZeneca)中に充填し、6ヶ月間40℃にて75%の相対湿度で保存した。結果は図2(A)を参照。
フマル酸ホルモテロール二水和物(0.2kg)およびラクトース一水和物(34kg)を回転型混合機中で1または2時間混合する。この混合物を、吸入に適した粒子径を得るためにスパイラルジェットミル中で微粉化した。結晶構造は制御された方法で米国特許第5.874.063号または米国特許第5.709.884号にしたがって再現された。このコンディショニングした生成物を微粉化したブデソニド(3kg)と回転型混合機中で30〜60分間混合する。第2の混合ステップとして粉末を改良型スパイラルジェットミルに送り、非常に低い混合圧にて高い窒素気流中で操作した。これにより粒子径のさらなる減少を引き起こすことなく凝集体を粉砕し(それによりアモルファス領域が形成され、続いて安定性の減少が起こる。)、粉末中のブデソニドの均一な分散が改善される。
凝集性の粉末の流動性を改善するために、それを室温にて50%未満の制御された相対湿度で球状化して凝集体とした。
実施例3の微粉化され球状化されたフマル酸ホルモテロール二水和物(5mg/g)/ブデソニド(90mg/g)/ラクトース一水和物(905mg/g)を粉末吸入器Turbuhaler(登録商標)(AstraZeneca)中に充填し、6ヶ月間40℃にて75%の相対湿度で保存した。結果は図2(B)を参照。
Claims (9)
- 混合物中に、所望により塩または溶媒和物または塩の溶媒和物の形態であってもよい微粉化されたホルモテロールである第1の活性成分、微粉化されたグルココルチコステロイドである第2の活性成分および薬学的に許容される担体/希釈剤を含む医薬組成物であって、
工程1. 微粉化された第1の活性成分および微粉化された担体/希釈剤の混合物を調製すること
工程2. 所望により、さらに混合物に微粉化された担体/希釈剤を加えること
工程3. 前もって微粉化された第2の活性成分を添加および混合すること(第2の活性成分は、所望により、微粉化された担体/希釈剤と前もって混合されていてもよい。)および
工程4. 工程3の後に粗大な担体/希釈剤を加えること、
を含む方法により得ることができ、そして
ホルモテロールの含有量が製剤の1.0%(w/w)未満である場合に開放容器で40℃および75%の相対湿度にて6ヶ月保存したときの製剤中のホルモテロールの分解が10%未満であるか、または粉末吸入器(dry powder device)内で貯蔵した場合に同じ条件下での分解が2.5%未満である高い保存安定性を有する組成物。
- ホルモテロールがフマル酸塩二水和物の形態である、請求項1に記載の医薬組成物。
- ホルモテロールが単一のR,R−エナンチオマーの形態である、請求項1または2に記載の医薬組成物。
- 第2の活性成分がブデソニドである、請求項1〜3のいずれかに記載の医薬組成物。
- 第2の活性成分がブデソニドの22R−エピマーである、請求項1〜3のいずれかに記載の医薬組成物。
- 担体/希釈剤がラクトースである、請求項1〜5のいずれかに記載の医薬組成物。
- 活性成分の粒子径が約10μm未満である、請求項1〜6のいずれかに記載の医薬組成物。
- 呼吸障害の処置または予防に使用するための請求項1〜7のいずれかに記載の医薬組成物。
- 喘息、鼻炎またはCOPDの処置または予防に使用するための請求項1〜7のいずれかに記載の医薬組成物。
