Description
(a)有機溶媒と有機金属前駆体との混合物を提供する工程、
(b)調製物を調製するために、工程(a)の前記混合物に、粒子状の充填材を添加する工程、
(c)工程(b)で調製された調製物を、インプラントにコーティングを提供するために、前記インプラントに適用する工程、および、
(d)前記適用したコーティングを乾燥する工程
(a)有機溶媒と有機金属前駆体との混合物を提供する工程、
(b)調製物を調製するために、工程(a)の前記混合物に、粒子状の充填材を添加する工程、
(c)工程(b)で調製された調製物を、インプラントにコーティングを提供するために、前記インプラントに適用する工程、および、
(d)前記適用したコーティングを乾燥する工程
ヒドロキシアパタイト充填材によるコーティング
69.5gのテトラブトキシチタネートを、室温で、500gのn−ブタノールに溶解させ、2時間攪拌した。つぎに、提供されたゾルの全重量(569.5g)を基準として、17.1g(3重量%に相当)の市販のリン酸カルシウムパウダー(Merck社、D50<2μm)を少しずつ添加した。1重量%の分散剤(DB194、無水物、Byk Chemie社、ウェーゼル)を、分散目的で添加し、さらに2時間攪拌した。
酸化チタニウム充填材によるコーティング
41.7gのテトラブトキシチタネートを、室温で、300gのn−ブタノール中に溶解させ、2時間攪拌した。提供されたゾルの全重量(341.7g)を基準として、1.71g(0.5重量%に相当)、10.26g(3重量%に相当)および20.52g(6重量%に相当)の市販の酸化チタンパウダー(Alfa Aesar社、カルルスルーエ;D50:0.3μm)を少しずつ添加した。1重量%の分散剤(DB194、無水物、Byk Chemie社、ウェーゼル)を分散の目的で添加し、さらに2時間攪拌した。
作用機序の記述
パウダー充填コーティングの作用機序を明らかにするために、実施例1に従って、ガラス基材(板状、直径14.5mm)に、異なる量の市販カルシウムパウダー(Merck社、D50<2μm)を含むコーティングを提供した。一方は、生理的条件下でのカルシウム放出を決定し、他方は、得られた表面上での細胞増殖を、充填したパウダーの機能として決定した。
光度計試験によってカルシウム濃度を決定した。これは、O−クレゾールフタレインコンプレクソン(CPC、Fa.Fluka社)がアルカリpHでカルシウムと反応することによる複合体の形成に関与している。形成された色素複合体の紫色は、575nmの吸光度で測定した。CPC試薬は、以下のように調製した。625mgの8−ヒドロキシキノリン(Fluka社)を25mlのジメチルスルホキシド(Serva社)に溶解し、つぎに、10mgのO−CPCおよび250μlの36%塩酸(Fluka社)を添加し、250mlの蒸留水を追加した。ジメチルアミン緩衝液を調製するために、2mlのジメチルアミンおよび25mgの塩化カリウムを40mlの蒸留水に溶解し、つぎに体積が50mlになるように蒸留水を追加した。試験のために、50μlのサンプル、標準品およびブランク(純粋な溶解緩衝液)を、2.5mlのPSマイクロセル(Brand社)に提供し、960μlの蒸留水を添加した。つぎに、1mlのCPC試薬および1mlのジエチルアミン緩衝液を各ケースにおいてピペットを用いて添加、混合し、そして、5分間のインキュベーション後、分光光度計(Beckmann Coulter社)で測定した。濃度を測定するために、カルシウム標準品を用いた。
培養用培地:90% PRMI1640(=2.05mmol/L グルタミン含有血清)、10% FCS(ウシ胎仔血清);インキュベーション:48時間、37℃、5% 二酸化炭素、静置培養、暗条件)
細胞系:MC3T3−E1(マウス胎仔骨芽細胞)
培養器:24ウェルペトリディッシュ、ポリスチレン製
接種数:120,000細胞/mlおよびウェル、G1期
ミトコンドリア活性:WST試験(Roche社、マンハイム)を行った。
Claims (22)
- インプラントにおける粒子が充填された酸化金属コーティングの製造方法であって、
(a)有機溶媒と有機金属前駆体との混合物を提供する工程、
(b)調製物を調製するために、工程(a)の前記混合物に、粒子サイズが0.01〜10μmである粒子状の充填材を添加する工程、
(c)工程(b)で調製された調製物を、インプラントにコーティングを提供するために、前記インプラントに適用する工程、および、
(d)前記適用したコーティングを乾燥する工程
を含み、
前記工程(b)における前記調製物の調製時に、前記粒子状の充填材は、前記工程(a)の前記混合物の前記前駆体材料および前記溶媒の全重量を基準として、重量割合が15%を超えないことを特徴とする製造方法。 - 前記混合物が、さらに、水、鉱酸、有機酸、または、それらの混合物を含む請求項1記載の製造方法。
- 前記溶媒が、C2〜C8の直鎖状もしくは分枝状アルコール、または、環状、芳香族もしくは複素環式芳香族の炭化水素、またはそれらの混合物である請求項1または2記載の製造方法。
- 前記有機金属前駆体が、多座配位型金属原子、ならびに、酸素原子およびRによって金属原子と配位する−ORリガンドを有する有機金属化合物であり、Rが、1つ以上の酸素および/または窒素原子が任意的に挿入且つ/または置換されたC2〜C8のアルキル基またはアルキレン基である請求項1から3のいずれか一項に記載の製造方法。
- 前記有機金属前駆体が、テトラエトキシチタネート、テトラ(n−プロポキシ)チタネート、テトラ(イソプロポキシ)チタネート、テトラ(n−ブトキシ)チタネート、テトラ(イソブトキシ)チタネート、テトラ(tert−ブトキシ)チタネート、テトラエトキシオルトケイ酸塩、テトラ(n−ブトキシ)オルトケイ酸塩、テトラ(イソブトキシ)オルトケイ酸塩およびテトラ(tert−ブトキシ)オルトケイ酸塩からなる群から選択される請求項4記載の製造方法。
- 前記有機金属前駆体の前記金属原子が、チタニウム、シリコン、アルミニウムおよびジルコニウム元素から選択される請求項1から5のいずれか一項に記載の製造方法。
- 前記充填材が、金属、金属合金、プラスチック材料、ガラス、ガラスセラミック、セラミック、複合材料、難溶性塩またはこれらの材料の組合せから形成される請求項1から6のいずれか一項に記載の製造方法。
- 前記工程(b)における前記調製物中の充填材量が、前記有機金属前駆体および前記溶媒の混合物の全重量を基準として15重量%以下である請求項1から7のいずれか一項に記載の製造方法。
- 前記充填材が、生理的条件下で活性物質を放出可能性のある請求項1から8のいずれか一項に記載の製造方法。
- 前記インプラントが、カテーテル、骨形成材、人工器官、外部固定器、内部固定器、爪、ねじもしくはワイヤー、心臓弁、人工血管もしくはシャント、顔面手術/プラスチックインプラント、中耳インプラントまたは歯科インプラントである請求項1から9のいずれか一項に記載の製造方法。
- 前記インプラントが、金属、金属合金、ガラス、セラミック、プラスチック材料、複合材料または骨代替材料である請求項1から10のいずれか一項に記載の製造方法。
- 前記適用が、浸漬コーティング、回転コーティング、ドクターブレード法による適用、印刷もしくは塗布である請求項1から11のいずれか一項に記載の製造方法。
- 前記乾燥工程が、空気中、不活性ガス中、または超臨界乾燥条件下で行われる請求項1から12のいずれか一項に記載の製造方法。
- 前記工程(d)後、適用した前記コーティングを、さらなる工程(e)において、100〜1000℃で加熱する請求項1から13のいずれか一項に記載の製造方法。
- 前記工程(b)の前記調製物をゾル形態で適用し、前記充填材が均一に分散された前記ゾルが、前記工程(c)における適用工程の間またはその後に、前記充填材が均一に分散されたゲルに変化する請求項1から14のいずれか一項に記載の製造方法。
- 前記工程(d)の前記乾燥工程および前記工程(e)の任意の加熱工程の後、層の厚みが50〜1000nmとなる層の厚みを得るために、前記工程(b)の前記調製物が適用される請求項14または15記載の製造方法。
- 前記工程(b)〜(d)および前記任意の工程(e)が、複数回行われる請求項14から16のいずれか一項に記載の製造方法。
- 前記工程(b)による前記調製物の前記充填材量が、異なるコーティングが異なる濃度の充填材を含むように、各回で異なる請求項17記載の製造方法。
- 適用された異なる前記コーティングが異なる充填材を含む請求項17または18記載の製造方法。
- 前記充填剤が、不溶性の充填剤である、請求項1から19のいずれか一項に記載の製造方法。
- 請求項1から20のいずれか一項に記載の製造方法により製造できるコーティングを有するインプラント。
- 前記コーティングに含まれる前記活性物質が、生理的条件下で周囲の媒質に溶出可能であることを特徴とする請求項21記載のインプラント。
