Claims (6)
- 微生物、動物組織、植物組織、血液、分泌物及びこれらの試料を組み換え遺伝子で形質転換させた試料から選択される1種以上の生物試料から核酸を分離する方法において、前記生物試料から起因した非溶解性凝集物の凝集又は沈降を促進させる粒子状物質を使用する核酸分離方法であって、
前記粒子状物質は平均粒径が10nm乃至500μmであり、
前記粒子状物質は平均密度が2g/cm 3 乃至10g/cm 3 であり、
前記粒子状物質は鉄、二酸化チタン、ニッケル、シリコン、及び白水晶からなる群から選ばれるものであり、
前記粒子状物質を使用する段階は、
(a)生物試料に直接入れる段階;又は
(b)生物試料溶解のための緩衝溶液に共に混ぜて入れる段階;
であることを特徴とする核酸分離方法。 - 前記粒子状物質は水溶液に分散されることを特徴とする請求項1に記載の核酸分離方法。
- 前記水溶液はアルコキシレート、アルカノールアミド、エステル、アミンオキシド、アルキルポリグリコシド、ポリアクリレート、ポリメタクリレート、ポリビニルピロリドン、ポリエチレンアミン、ポリビニルアミン、ベタイン、グリシネート、イミダゾリン及びグリセロールからなる群から選ばれる分散剤をさらに含有することを特徴とする請求項2に記載の核酸分離方法。
- 前記核酸と非溶解性凝集物の分離は遠心分離法、真空分離法(vacuum manifold type)、フィルター分離法、重力分離法、又はクロマトグラフィ法から選ばれることを特徴とする請求項1に記載の核酸分離方法。
- (a)生物試料に直接入れる段階;又は
(b)生物試料溶解のための緩衝溶液に共に混ぜて入れる段階;
において使用される粒子状物質を含有する核酸分離用組成物において、
前記粒子状物質は平均粒径が10nm乃至500μmであり、
前記粒子状物質は平均密度が2g/cm 3 乃至10g/cm 3 であり、
前記粒子状物質は鉄、二酸化チタン、ニッケル、シリコン、及び白水晶からなる群から選ばれるものであることを特徴とする核酸分離用組成物。 - 前記核酸分離用組成物はアルコキシレート、アルカノールアミド、エステル、アミンオキシド、アルキルポリグリコシド、ポリアクリルレート、ポリメタクリレート、ポリビニルピロリドン、ポリエチレンアミン、ポリビニルアミン、ベタイン、グリシネート、イミダゾリン及びグリセロールからなる群から選ばれる1種以上の分散剤を0.1乃至1.0体積%の量で含有する水分散液であることを特徴とする請求項5に記載の核酸分離用組成物。
