Description
本発明は、変性剤で前処理されている天然ジェランガムを含む、殺菌ミルク組成物の製造方法に関する。本発明はさらに、結果として生じる殺菌ミルク組成物およびミルクを主成分とするジェラン食製品に関する。
ジェランガムは、細菌スフィンゴモナス・エロデア(Sphingomonas elodea)が産生する菌体外多糖類である。多くのS.エロデア(S. elodea)株は、S-60、LPG-2、PDG-1、およびPDG-4を含む、異なる特質のジェランガムを産生する。S.エロデアが産生するジェランガムは、カリフォルニア州サンディエゴのCP Kelcoより、Kelcogel LT100(登録商標)として販売されている。
本発明は、変性剤で前処理されている、天然ジェランガムを含む殺菌ミルク組成物の製造方法に関する。その方法は、天然ジェラン/ミルク組成物を混合および殺菌する前に、一般的酸化剤またはアルカリ腐食剤などの変性剤で天然ジェランガムを前処理する段階を含む。好ましくは、変性剤処理により12ヶ月保存後の殺菌天然ジェラン/ミルク組成物中のパラ-クレゾールレベルが十億分の25単位 (ppb)よりも低下する結果となり、最も好ましくは、味覚でパラ-クレゾールが検出できないレベルになる。
本明細書に定義されているように、殺菌とは138 ℃以上、2秒以上の超高温処理をさす。最も好ましくは、約140〜150 ℃で約4〜6秒間殺菌を行う。殺菌および密閉容器に詰めた後、殺菌ミルクは室温で12か月まで変質することなしに保存することができる。
パラ-クレゾール産生を低下させる方法は、天然ジェランガムに直接行われる。好ましくは、その方法は沈殿前の天然ジェラン発酵液中、または既に沈殿済みの場合には再構成液中などの液体試料中で行われる。発酵または再構成後に、例えば、カバーされた水槽中の温度調節、または蒸気の直接注入などの当技術分野において周知の技術によりジェラン溶液を約25℃から約100℃の範囲の温度まで加熱する。好ましくは、加熱時間を減らすために蒸気注入が用いられる。溶液の望ましい温度範囲は、用いる変性剤によって異なる。次亜塩素酸ナトリウムのような酸化剤を変性剤に用いる場合には、温度範囲は約5℃から約100℃であり、好ましくは、約20℃から約50℃である。アルカリ腐食剤を変性剤に用いる場合には、温度範囲は約25℃から約70℃であり、好ましくは、約40℃から約60℃である。
クエン酸塩、EDTA、または様々なポリリン酸塩などの選択的なキレート剤をジェラン溶液中に変性剤とともに添加してもよい。キレート剤の好ましい範囲は0〜3000 ppmの間であり、最も好ましくは約1000〜2000 ppmの間である。キレート剤による処置は、好ましくは約1時間であり、その後、下記のような酵素処理が行われる。
本発明の最も好ましい態様は、残存する細胞破片を破壊するために、ジェラン溶液に対して選択的な工程である酵素処理を行う。変性剤(および、選択された場合にはキレート剤)処理の後に、ジェラン溶液の酵素処理を行う。1つまたはそれ以上の酵素は、連続的または同時に添加可能であり、一般的にはそれぞれ1時間から3時間処理を継続する。最も好ましくは、酵素処理はリゾチームおよびプロテアーゼの両者を含み、それぞれ約2時間処理する。リゾチーム濃度は1〜100活性単位の間で変動し、好ましくは約22活性単位である。プロテアーゼ濃度は220〜440活性単位の範囲であり、好ましくは約220活性単位である。リゾチームおよび/またはプロテアーゼによる処理は、同時または順番に行ってよく、いかなる順序であってもよい。
上述のように、変性剤は過酸化水素、オゾン、もしくは次亜塩素酸塩などの一般的な酸化剤でよく、または変性剤は水酸化物塩、炭酸塩、およびアルカリリン酸塩などのアルカリ腐食剤でよい。変性剤がアルカリ腐食剤の場合には、第二のプロトコールが好ましい。第二のプロトコールでは、リゾチーム処理をアルカリ腐食剤処理の前に行い、プロテアーゼ処理を最後に行う。この第二のプロトコールは、リゾチームが、酸性から中性の至適pH条件においてジェラン溶液中で反応することを可能にする。アルカリ腐食剤添加およびそれに続く中和の後では、それによってpHが上昇し、プロテアーゼを約pH 8またはそれ以上という好ましいpHにおいて反応に加えることが可能になる。第一のプロトコールと同様に、腐食剤の攪拌時間は一般的にはそれぞれ0.5時間から2時間であり、最も好ましくは0.5時間から約1.0時間である。アルカリ腐食剤の一般的なレベルは上記と同様で、約500 ppmから約2000 ppmの間であり、最も好ましくは約500 ppmから約1500 ppmの間である。キレート剤濃度の範囲は上述と同様で、リゾチームおよびプロテアーゼ濃度も同様である。
変性剤、ならびに選択的なキレート剤および酵素処理の後に、試料は80℃またはそれ以上で殺菌され、望ましい量でミルクと混合させる前に、約pH 5から7の範囲の軽度酸性のpHに調整される。ミルクと混合させたら、天然ジェラン/ミルク組成物は超高温殺菌にかけられる。

5ガロンの温かいKelcogel LT100(登録商標)(ジェラン溶液)を40℃に加熱した。その後、下記の表Aに提示される酸化剤をジェラン溶液中に導入し、高速攪拌機により約5分間攪拌した。その後、溶液は時間を変えて24℃で保存した。
変性剤としてアルカリ腐食剤を用いた処理によるパラ-クレゾールの低減
5ガロンのKelcogel LT100(登録商標)(ジェラン溶液)を50リットルの湯沸かし中の振とうしたスチーム炊事道具に移し、40℃に加熱した。この時点で、ジェラン溶液の一部を対照試料として注ぎ出し、実施例1に記載したように沈殿させた。沈殿の後、天然ジェランの対照試料を0.12%でミルクと混合し、殺菌して一ヶ月保存した。その後、味覚と臭いによりパラ-クレゾールを検査した。
Claims (1)
- ミルクと混合する前に、天然ジェランガムを変性剤で処理する段階を含み、該変性剤処理が、天然ジェランガムを加熱する段階及び該加熱されたジェランガムに変性剤を添加する段階によって行われる、ジェランガムを含む殺菌されたミルク組成物の製造方法であって、
該変性剤が、過酸化水素および次亜塩素酸塩からなる群より選択される酸化剤、または水酸化物塩から選択されるアルカリ腐食剤である、方法。
