【発明が解決しようとする課題】
この使用感触を得るために、これらの製品において、カルボキシビニルポリマーが、増粘及び安定化剤として一般的に用いられている。しかしながら、アスコルビン酸リン酸マグネシウム,塩酸アルギニンのようなイオン化する薬剤を配合した製品や、顔料を配合したメーキャップ製品等においては、イオン化する薬剤や顔料等によってカルボキシビニルポリマーの増粘性が著しく阻害される。その結果、製品の保存中に、粘度低下,さらには、分離,沈降が起こる等、これらの製品は、長期安定性の面で問題を有している。

この点を改善する目的で、キサンタンガムやヒドロキシエチルセルロース等の増粘剤が用いられる場合もある。しかしながら、このような増粘剤の使用により製品の長期安定性を満足させると、今度は、ぬめり感やべたつき感が生じてしまう。

これらのことは、製品の使用感触や剤形の画一化につながり、消費者の多様化する嗜好を必ずしも満足させ得ていないのが現状である。
そこで、本発明は、消費者の多様化する嗜好に対応できる新たな使用感触を有し、長期安定性にも優れている、化粧料等の外用組成物を提供することを課題とする。

【課題を解決するための手段】
前記課題を解決するために本発明者らが鋭意検討を重ねた結果、増粘及び安定化のために、カルボキシビニルポリマーに代わって、サクシノグルカン及び水膨潤性粘土鉱物を用いることによって、水々しい上に肌なじみがよく、非常にさっぱりとした使用後感を与える等、従来の化粧料にない使用感触を有し、しかも、イオン化する薬剤や顔料を配合しても、粘度低下や分離,沈降が起こらず、長期安定性にも優れている、特に化粧料として有用な外用組成物が得られることを見い出し、本発明を完成するに至った。

すなわち、本発明は、サクシノグルカンと水膨潤性粘土鉱物とを含有し、かつ、アクリル酸メタクリル酸アルキル重合体を組成物全体に対して0.005〜5.0重量%含有する、外用組成物を提供する発明である。この組成物に油性成分を配合すると、より好ましい使用感触を有する外用組成物が得られる。また、この組成物、特に油性成分を含有するものに、界面活性剤を配合すると、特に本発明の所期の効果を有効に発揮し得る。

【発明の実施の形態】
以下、本発明の実施の形態について説明する。
本発明外用組成物(以下本発明組成物という)は、サクシノグルカンと水膨潤性粘土鉱物とを含有する。

本発明組成物に配合されるサクシノグルカンは、微生物に由来する多糖類の一種であり、ガラクトース及びグルコースから誘導される糖単位に加え、コハク酸及びピルビン酸及び随意成分としての酢酸又はこれらの酸の塩から誘導される単位を含む、微生物に由来する多糖類を意味する。

より具体的にはサクシノグルカンは、平均分子量が約600万で、以下の構造式を有する、ガラクトース単位1:グルコース単位7:コハク酸単位0.8:ピルビン酸単位1を含み、場合により随意成分である酢酸単位を含む、水溶性高分子である。

上記式中、Glucはグルコース単位を、Galac はガラクトース単位を表す。また、括弧内の表示は、糖単位同士の結合様式を表す。例えば(β1、4)は、β1−4結合を表す。

このサクシノグルカンの供給源となる微生物としては、例えば、シュードモナス属、リゾビウム属、アルカリゲネス属又はアグロバクテリウム属の微生物が挙げられるが、アグロバクテリウム属に属する細菌であるアグロバクテリウム・ツメファシエンスI−736(ブタペスト条約に従い1988年3月1日に微生物培養締結国収集機関(CNCM)に寄託され、I−736の番号で公に入手できる)が、特に好ましい。

サクシノグルカンは、これらの微生物を培地中で培養することによって製造することができる。
より具体的には、概ね、グルコース,蔗糖,デンプンの加水分解物等の炭素源;カゼイン,カゼイネート,野菜粉末,酵母エキス,コーンスティープリカー(CSL)等の有機窒素源;金属の硫酸塩,リン酸塩,炭酸塩等の無機塩類;及び、随意微量元素等を含む培地中で、上記微生物を培養することによって、サクシノグルカンを製造することができる。

また、一般にこの培養は、1〜4バールの圧力下、25〜35℃の温度で、撹拌培養等の好気条件下で行うことが好ましい。また、培地のpHは5〜9、好ましくは6〜8である。

培養終了後、培養培地を熱処理後、イソプロピルアルコール等の有機溶媒と連続的に接触させることにより、サクシノグルカンを析出させることができる。サクシノグルカンを培養培地から単離する際には、好ましくは、これを濾過し、遠心分離を施し、加圧、乾燥する。このようにして、本発明組成物に配合し得るサクシノグルカンを得ることができる。

なお、本発明組成物には、このようにして製造されたサクシノグルカンをそのまま配合し得ることは勿論、必要に応じて、これに、酸分解,アルカリ分解,酵素分解,超音波処理等を施して得られる分解処理物も同様に配合することができる。

本発明組成物中のサクシノグルカンの含有量は、組成物全体に対して0.01〜15.0重量%であることが好ましい。特に、サクシノグルカンの含有量が、組成物全体に対して、0.1〜10.0重量%である場合には、本発明組成物はジェル状を呈し、長期安定性により優れたものとなる。

本発明組成物に配合される水膨潤性粘土鉱物は、水膨潤性を有する粘土鉱物であれば特に制限されず、天然のものでも合成のものでもよく、また、いかなる組成のものでもよい。

水膨潤性粘土鉱物としては、例えば、モンモリロナイト,サポナイト,ヘクトライト,バイデライト,ノントロナイト,ソーコナイト,スチブンサイト等のスメクタイト;膨潤性の雲母等が挙げられる。

本発明組成物には、水膨潤性粘土鉱物の1種のみを配合しても、2種以上を組み合わせて配合してもよい。
本発明組成物中の水膨潤性粘土鉱物の含有量は、組成物全体に対して0.01〜15.0重量%であることが好ましく、0.1〜10.0重量%であることが特に好ましい。水膨潤性粘土鉱物の含有量が、組成物全体に対して0.01重量%未満であると、得られる組成物の長期安定性が悪くなり好ましくなく、同15.0重量%を超えると、本発明の特徴を有する組成物の製造が困難となり好ましくない。

本発明組成物に、サクシノグルカン及び水膨潤性粘土鉱物に加え、さらに油性成分を配合することが好ましい。油性成分を配合すると、皮膚に塗布する際ののびが非常によくなり、また、うるおい感が向上する等、特に化粧料として、より好ましい使用感触を有するものが得られる。

本発明組成物中の油性成分の含有量は、組成物全体に対して0.1〜40.0重量%であることが好ましく、0.3〜30.0重量%であることが特に好ましい。油性成分の含有量が、組成物全体に対して0.1重量%未満であると、皮膚に塗布する際ののびの向上が認められず好ましくなく、同40.0重量%を超えると、さっぱり感が失われ、使用感触が悪くなり好ましくない。

本発明組成物は、皮膚,毛髪等に適用することができる。本発明組成物は、水溶液系、可溶化系、乳化系、油液系、ゲル系、軟膏系、エアゾール系、水−油2層系、水−油−粉末3層系等の種々の剤形を採り得る。本発明組成物は、化粧料として特に有用であるが、医薬品や医薬部外品等としても使用することができる。例えば、本発明組成物が基礎化粧料である場合、スキンケアを目的としたエッセンス(美容液),乳液,クリーム,ジェル等の製品形態を採り得る。本発明組成物がメーキャップ化粧料である場合、ファンデーション,アイシャドー,マスカラ,化粧下地等の製品形態を採り得る。本発明組成物が毛髪化粧料である場合、育毛剤,ヘアスタイリング剤等の製品形態を採り得る。本発明組成物がボディーケア化粧料である場合、サンスクリーン,サンタン,アフターサンケア,ハンドクリーム等の製品形態を採り得る。本発明組成物が医薬品又は医薬部外品である場合、各種の軟膏剤等の製品形態を採り得る。本発明組成物は、これらの剤形及び製品形態に限定されるものではない。

以下、実施例等を挙げて本発明をさらに詳細に説明するが、本発明はこれらの実施例に限定されるものではない。

サクシノグルカンの製造例
サクシノグルカンを、特公平6−74283号の記載に従い製造した。すなわち、後述する組成の培養培地で、アグロバクテリウム・ツメファシエンスI−736株を用いて発酵を行った。

具体的には、培養培地にアグロバクテリウム・ツメファシエンスI−736株を接種して、使用容量15Lを有する20LのBiolaffite(登録商標)容器中で、28℃で、Ruston(登録商標)タイプ撹拌機を用いて400rpm での撹拌培養を行い、その際、培地を825L/時の風量で曝気した。蔗糖の全消費乃至事実上全消費に相当する90時間の撹拌培養後、使用蔗糖重量に関して定義されるへテロ多糖類66重量%が得られた。30rpm での円筒型スピンドルNo.4を用いたブルックフィールドLTV(登録商標)粘度計により測定した発酵液の粘度は6800Pa/sであった。

サクシノグルカンは、90℃で3分間熱処理された上記発酵液2kgから回収された。すなわち、このようにして熱処理された発酵液に、イソプロピルアルコール2300mLを加え、硫酸ナトリウム150gで沈殿を生じさせた。次いで、この沈殿から生じた繊維について、イソプロピルアルコール1200mLで脱水処理を2回行い、この脱水済繊維を加圧してばらばらに引き裂いて、85℃のオーブンで乾燥させた。得られた乾燥物を粉砕した。このようにして、クリーム色のサクシノグルカン粉末を得た。
