- 配列番号1のアミノ酸配列で表される、Taq DNAポリメラーゼに選択的に結合するポリペプチドであって、
前記アミノ酸配列において、
45番残基はQ、S、R、Y、V、またはN;
47番残基はI、T、K、A、またはQ;
49番残基はN、H、L、MまたはI;
67番残基はY、R、AまたはK、
69番残基はAまたはK;
72番残基はGまたはA;
91番残基はI、またはS;
93番残基はTまたはW;
94番残基はGまたはL;
115番残基はVまたはS;
117番残基はVまたはH;及び
118番残基はEまたはWである、ポリペプチド。 - 下記の配列:

118番残基はWを有するTaq DNAポリメラーゼに選択的に結合する、請求項1に記載のポリペプチド。 - 67番残基はR;

上記の配列を有するTaq DNAポリメラーゼに選択的に結合する、請求項1または2に記載のポリペプチド。 - 前記ポリペプチドは、配列番号3〜13のいずれか一つのアミノ酸配列または前記アミノ酸配列の残基のうち45番、47番、49番、67番、69番、72番、91番、93番、94番、115番、117番、及び118番残基のうちの一つ以上が表1の保存的置換残基で置き換えられたアミノ酸配列を有する、請求項1〜3のいずれか一つに記載のポリペプチド。
- 配列番号1のアミノ酸配列で表されるポリペプチドであって、前記アミノ酸配列において、
45番残基はS、T、A、またはN;
47番残基はK、R、Q、E、またはN;
49番残基はI、L、V、M、またはF;
67番残基はR、K、Q、またはH;
69番残基はK、R、Q、E、またはN;
72番残基はA、S、G、またはT;
91番残基はS、T、A、またはN;
93番残基はW、Y、またはF;
94番残基はL、I、V、M、またはF;
115番残基はS、T、A、またはN;
117番残基はH、N、Q、R、またはY;及び
118番残基はW、Y、またはFである、ポリペプチド。 - 前記ポリペプチドは、ホモまたはヘテロダイマー、または三つのホモまたはヘテロトライマーを形成するものである、Taq DNAポリメラーゼに選択的に結合する、請求項1〜5のいずれか一つに記載のポリペプチド。
- 請求項1〜5のいずれか一つに記載のポリペプチドをコードするポリヌクレオチド。
- 前記ポリヌクレオチドは、配列番号14〜24で表されるものである、請求項6に記載のポリヌクレオチド。
- 請求項7または8に記載のポリヌクレオチドを含む組換えベクター。
- 請求項7に記載のポリヌクレオチドまたは請求項9の組換えベクターが導入された形質転換細胞。
- 請求項1〜5のいずれか一つに記載のポリペプチドを含むHot Start用PCR(Polymerase Chain Reaction)組成物。
- 請求項1〜5のいずれか一つに記載のポリペプチドまたは請求項11に記載の、PCR組成物を利用したHot start PCR方法。
- 請求項1〜5のいずれか一つに記載のポリペプチドまたは請求項11に記載のPCR組成物を鋳型及びPCRに必要な成分と混合する段階及び前記混合物をPCRが可能な条件で反応する段階を含む、Hot start PCR方法。
- 請求項1〜5のいずれか一つに記載のポリペプチドをHot−start PCRに使用する用途。
- 請求項1〜5のいずれか一つに記載のポリペプチドをHot−start PCR組成物の製造に使用する用途。
