ここに、J:誘導電流密度[A/m2],σ:培地導電率[S/m],π:円周率,f:周波数[Hz],B:磁束密度[T],r:半径[m]をそれぞれ表す。
細胞非接着基質:アガロースゲル
細胞接着基質 :ポリエチレンイミン(PEI)
鋳型の材質 :ポリジメチルシロキサン(PDMS)
培地 :Neurobasal medium(+2% B−27 supplement,1% Glutamax supplement,1% Pen−Strep)(20 ml で培養,2乃至3日に一度半量の培地交換を実施)
細胞 :ラット大脳皮質から採取された神経細胞
培養条件<1> :鋳型を付けた状態で7日間培養し、
その後に鋳型を取り外した状態で39日間培養
培養条件<2> :鋳型を付けた状態で21日間培養し、
その後に鋳型を取り外した状態で25日間培養

Claims (4)
- 培養容器内に細胞非接着基質の領域が形成され、細胞培養スポットが形成されている鋳型が前記細胞非接着基質の領域に設置されると共に前記鋳型の前記細胞培養スポット内に細胞接着基質の領域が形成され、その上で前記培養容器へと培地並びに細胞と組織とのうちの少なくとも一方が投入され、前記細胞や前記組織の培養が行われた後に前記鋳型が取り外されることを特徴とする培養細胞供試体の作製方法。
- 前記鋳型として磁性を有する材料が添加された鋳型が用いられることを特徴とする請求項1記載の培養細胞供試体の作製方法。
- 前記細胞非接着基質の領域を形成する物質として温度応答性高分子が添加された物質が用いられ、加熱されることによって前記細胞接着基質の領域が前記細胞非接着基質から剥離することを特徴とする請求項1記載の培養細胞供試体の作製方法。
- 前記細胞接着基質の領域を形成する物質として温度が変化することによって前記細胞や前記組織の接着性を有する性質から前記細胞や前記組織の接着性を有しない性質へと変質する物質が用いられ、加熱若しくは冷却されることによって前記細胞接着基質の領域から前記細胞や前記組織が分離して回収されることを特徴とする請求項1記載の培養細胞供試体の作製方法。
