Abstract
Description
この出願は、2007年9月26日に出願された、係属中の米国仮特許出願第60/955,348号に対する優先権を主張する。上記出願は、あたかもそれが本明細書に完全に再度記載されているように、参考として援用される。
本発明は、一般に、炭素源としてスクロースを使用して、Corynebacterium属またはBrevibacterium属の細菌によってアミノ酸を産生するための方法および組成物に関する。より詳細には、本発明は、炭素源としてスクロースを使用した、Corynebacterium glutamicumからのL−リジンの産生に関する。
CorynebacteriumおよびBrevibacteriumの多くの商業的発酵は、炭素源としてグルコースを使用する。したがって、多くの細菌産生培養物が、炭素源としてグルコースを使用して産生速度および/または収量を最適化するように設計されてきた。商業的に重要なアミノ酸であるL−リジンの産生は、最適化の特定の標的である。
実施例1は、ptsF遺伝子(配列番号2)によってコードされるフルクトース−PTS酵素II(配列番号1)の破壊を開示している。これは、スクロース上で増殖する培養物の増殖培地におけるフルクトース濃度の増加をもたらす。このプロセスによって、Moonらによって記載されたものと類似したptsF変異体が生じることになる。
実施例2は、スクロースを含有するリジン発酵培地へのグルコースイソメラーゼの添加を説明する。
実施例3は、C.glutamicumのptsF変異体バックグラウンド菌株におけるフルクトース排出変異体の作製を説明する。

Claims (14)
- 発酵によってアミノ酸を産生する方法であって、
炭素源としてスクロースを含有し、発酵培地においてフルクトースをグルコースに転換するのに有効な量のグルコースイソメラーゼを含有する該発酵培地においてフルクトース−PTS酵素IIのトランスフェラーゼ活性が減弱または遮断されているC.glutamicum菌株を増殖させるステップと、該アミノ酸を産生するために該微生物を発酵させるステップと
を含む方法。 - 前記アミノ酸がL−リジンであることを含む、請求項1に記載の方法。
- 前記フルクトース−PTSの減弱または遮断が、前記菌株のptsF遺伝子の破壊または欠失によってもたらされる、請求項1に記載の方法。
- 前記グルコースイソメラーゼが、前記発酵培地に外因性に添加される、請求項1に記載の方法。
- 前記グルコースイソメラーゼが、前記C.glutamicum菌株中で産生され、該菌株から前記培地中へ排出される、請求項1に記載の方法。
- 前記発酵培地が、該発酵培地において前記スクロースの一部をフルクトースおよびグルコースに転換するのに有効な量のインベルターゼをさらに含む、請求項1に記載の方法。
- 発酵によってアミノ酸を産生する方法であって、
炭素源としてスクロースを含有する発酵培地において、フルクトース−PTS酵素IIのトランスフェラーゼ活性が減弱または遮断されており、C.glutamicum菌株からのフルクトースの排出を減弱または遮断する変異をさらに含む該菌株を増殖させるステップと、
該アミノ酸を産生するために該菌株を発酵させるステップと
を含む、方法。 - 前記菌株においてグルコースキナーゼが過剰に発現される、請求項1に記載の方法。
- 前記菌株においてグルコースイソメラーゼが過剰に発現される、請求項1に記載の方法。
- 前記菌株においてグルコースキナーゼおよびグルコースイソメラーゼが過剰に発現される、請求項1に記載の方法。
- フルクトースのC.glutamicum菌株中への移入を減弱または遮断する、該ptsF遺伝子における第1の変異を有し、該菌株からのフルクトースの排出を減弱または減少させる変異をさらに有するC.glutamicum菌株。
- 前記C.glutamicum菌株においてグルコースイソメラーゼおよびグルコースキナーゼのうちの少なくとも1つを過剰に発現するように構成された手段をコードする組換え型核酸をさらに含む、請求項11に記載のC.glutamicum菌株。
- C.glutamicum菌株がスクロースを含む培地上で増殖した場合、フルクトースの該菌株中への移入を減弱または遮断するptsF遺伝子における第1の変異を有し、かつ、グルコースイソメラーゼを発現し、該グルコースイソメラーゼを該菌株から発酵培地中へ排出するように構成された手段をコードする組換え型核酸を有する、C.glutamicum菌株。
- C.glutamicum菌株からのフルクトース排出を減弱または減少させる変異を有するC.glutamicum菌株。
