Description
本出願は、本明細書中、その全体が参照として含まれる、2007年7月11日出願の米国シリアル番号第60/949,142号の恩典を主張する。
イオンチャンネルは、薬学及び生命工学業界、特に心臓、肺及び胃腸の健康分野での最も大きな種類の治療標的の一つを構成する。細胞の内外へイオン流動を制御する際に関与するタンパク質の治療的標的化(therapeutic targeting)は患者の健康に劇的な効果がある。イオンチャンネル標的を調節するその能力に関して化合物を試験するのは困難であり、時間の浪費となることがある。パッチクランプアッセイ(patch−clamp assay)は、イオンチャンネルモジュレータの生物学的作用の非常に高感度の測定法である。しかしながらパッチクランプ法は複雑であり、試験化合物の処理量が少ない。
本発明のバイオセンサは比色共鳴反射バイオセンサである。たとえばCunninghamら、“Colorimetric resonant reflection as a direct biochemical assay technique,”、Sensors and Actuators B、第81巻、316−328頁、2002年1月5日;米国特許公開(U.S. Pat. Publ.)第2004/0091397号を参照されたい。比色共鳴バイオセンサは、表面プラズモン共鳴(SPR)バイオセンサではない。SPRバイオセンサは、銀、金、銅、アルミニウム、ナトリウム及びインジウムなどの薄い金属層をもつ。この金属は、好適な波長で光と共鳴し得る伝導帯電子を持たねばならない。光に暴露されたSPRバイオセンサ表面は純粋な金属でなければならない。酸化物、硫化物及び他のフィルムはSPRと干渉する。比色共鳴バイオセンサは金属層を持たない。むしろこれらはTiO2などの高屈折率材料の誘電体コーティングを有する。
たとえば、物理的吸着または化学的結合により、一つ以上の細胞をバイオセンサ上に固定化することができる。細胞は非粘着性細胞でも粘着性細胞であってもよい。細胞は生来、一つ以上のイオンチャンネルまたは一つ以上のチャンネル成分を発現することができる。あるいは細胞は、組み換え的に一つ以上のイオンチャンネルまたはイオンチャンネル成分を発現することができる。細胞は哺乳類細胞、たとえばヒト細胞であり得る。
本発明の一態様では、イオンチャンネルのモジュレータの識別法を提供する。イオンチャンネルのモジュレータは、イオンチャンネルの機能活性に影響を与える。機能活性における影響としては、イオンチャンネル活性の遮断または抑制が挙げられる。遮断または抑制は、イオンチャンネルの刺激剤の存在下、またはこれに応答して起きることができる。あるいはモジュレータは、イオンチャンネルの機能活性を促進または増進することができる。促進は、イオンチャンネルを遮断する分子の存在下で、またはこれに応答して起きることができる。本発明の方法は組み換え細胞と共に使用することができるが、イオンチャンネルタンパク質またはサブユニットを組み換え的に過剰発現する細胞は、本発明の方法で要求されていない。細胞は、比色共鳴反射光バイオセンサ表面の第一の位置に適用することができる。細胞はバイオセンサに固定化できるか、またはこれらは単にバイオセンサ表面上で成長することができる。場合により、細胞は、約1、5、10、30分若しくはそれ以上、または約1、2、5、24、48、36時間若しくはそれ以上の間、本発明の方法の任意の段階でバイオセンサ表面でインキュベートすることができる。アッセイは、約25、30または37℃の温度(または約25〜37℃の間の任意の範囲の温度)で実施することができる。
Claims (12)
- イオンチャネルのアンタゴニストまたはアゴニストを識別する方法であって、
(a)比色共鳴反射光バイオセンサの表面上の第一の位置と第二の位置に細胞を適用すること;
(b)試験試薬を前記第一の位置に適用すること;
(c)前記第二の位置に細胞の既知のイオンチャネルアンタゴニストまたはアゴニストを適用すること;及び
(d)前記第一の位置に関しては比色共鳴反射光第一ピーク波長値(peak wavelength value:PWV)と、前記第二の位置に関しては第二のPWVを検出すること、
を含み、
ここで第一及び第二のPWVが同一または類似であれば、試験試薬はイオンチャネルのアンタゴニストまたはアゴニストであり;第一及び第二のPWVが実質的に異なれば、試験試薬はアンタゴニストでもアゴニストでもない、前記方法。 - 前記第一の位置に細胞を適用後;前記第一の位置に試験試薬を適用後;前記第二の位置に細胞を適用後;既知のイオンチャネルアンタゴニスト若しくはアゴニストを前記第二の位置に適用後;またはこれらの組み合わせの後、前記細胞を一定期間インキュベートする、請求項1に記載の方法。
- 前記PWVは、約2マイクロメートル〜約200マイクロメートルの下限ピクセルサイズをもつレンズを備えたスキャナを使用して検出する、請求項1に記載の方法。
- 前記比色共鳴反射光バイオセンサ表面上の前記第一の位置と第二の位置は、マイクロタイターウェル、マイクロタイタープレート、試験管、ペトリ皿、ミクロフルイドチャネル及びマイクロアレイからなる群から選択される容器の内部表面である、請求項1に記載の方法。
- 前記細胞、試験試薬及びイオンチャネルアンタゴニストまたはアゴニストは、検出標識を含まない、請求項1に記載の方法。
- 約25、30または37℃の温度で前記方法を実施する、請求項1に記載の方法。
- イオンチャネルのモジュレータを識別する方法であって、
(a)比色共鳴反射光バイオセンサの表面上の第一の位置に細胞、試験試薬及び細胞の既知のイオンチャネルアンタゴニストまたはアゴニストを適用すること;
(b)前記第一の位置に関して第一のPWVを検出すること;
(c)比色共鳴反射光バイオセンサ表面上の第二の位置に、細胞と細胞の既知のイオンチャネルアンタゴニストまたはアゴニストを適用すること;及び
(d)前記第二の位置に関して第二のPWVを検出すること;
を、含み、
ここで第一及び第二のPWVが異なっていれば、試験試薬はイオンチャネルのモジュレータであり;第一及び第二のPWVが同一または類似であれば、試験試薬はイオンチャンネルのモジュレータではない、前記方法。 - 前記第一の位置に細胞を適用後;前記第一の位置に試験試薬を適用後;前記第一の位置に既知のイオンチャネルアンタゴニスト若しくはアゴニストを適用後;前記第二の位置に細胞を適用後;前記第二の位置に既知のイオンチャネルアンタゴニストまたはアゴニストを適用後;またはこれらの組み合わせの後、前記細胞を一定期間インキュベートする、請求項7に記載の方法。
- 前記PWVは、約2マイクロメートル〜約200マイクロメートルの下限ピクセルサイズをもつレンズを備えたスキャナを使用して検出する、請求項7に記載の方法。
