Description
<実験1−1:サイトカイン類の産生異常動物実験モデル>
3週齢のC57/BL6雌マウス(日本チャールスリバー株式会社から購入)(以下、単に「マウス」と言う。)16匹を各群4匹からなる4群(A群乃至D群)に分け、市販の標準飼料『MF固形飼料』(オリエンタル酵母株式会社販売)を用いて1週間予備飼育した。次いで、A群のマウスを正常マウス群(対照1)として、MF固形飼料を与えて26週間飼育した。又、B群(対照2)、C群(試験1)及びD群(試験2)のマウスには、高脂血症を惹起させて、サイトカイン類(IL−1β、IFN−γ、及びIL−4)の産生異常を誘発するための後述するHF飼料を与えて26週間飼育した。飼育期間中、A群及びB群のマウスには、通常の飲み水を自由摂取させる一方、C群及びD群のマウスには、後述する糖転移ルチンを添加した飲み水、及び糖転移ヘスペリジンを添加した飲み水をそれぞれ与えて飼育した。尚、A群乃至D群に於いて、本飼育実験期間中、各群間に各群のマウスが自由摂取する水量、及び栄養学的な観点から見た飼料摂取量に統計学上の有意差は認められなかった。
MF固形飼料に対し、コレステロール1.25%(w/w)、コール酸ナトリウム0.5%(w/w)、及びココアバター15%(w/w)を添加し、放射線滅菌し、高脂肪飼料(以下、「HF飼料」と言う。)を得た。このHF飼料は、マウスに摂取させて高脂血症を惹起させ、サイトカイン類(IL−1β、IFN−γ、及びIL−4)産生異常を誘発させるための飼料である。
実験1−1の26週間飼育後のA群(対照1)、B群(対照2)、C群(試験1)、及びD群(試験2)の各群のマウスをエーテル麻酔して、脾臓をそれぞれ摘出し、その一部をそれぞれ機械的に細断し、細胞濃度5×106個/mlとなるように10%(v/v)のウシ胎児血清を補足したRPMI1640培地中に懸濁し、これを24穴マイクロプレートに1.5ml/穴の割合で播種した。次いで、各穴当たり、リポポリサッカライド(LPS)を10μg/ml又はコンカナバリンA(ConA)を5μg/mlとなるように添加し、37℃、5%(v/v)CO2インキュベータ内で24時間静置培養した。その後、A群乃至D群の各群マウス由来の脾細胞培養液をそれぞれそれ個別に回収し、遠心分離(3,000r.p.m.×10分)し、上清を回収し、サイトカイン類(IL−1β、IFN−γ、及びIL−4)産生量を常法に従ってエンザイム・イムノアッセイ(ELISA)法によりそれぞれ定量した。尚、有意差検定は、スチューデント・t−テスト(Student’s t−test)法で行った。その結果を図1乃至図3に示す。
<実験2−1>
7週齢のdd系マウスを用いて、糖転移ビタミンP類として、特開平3−27293号公報に開示された方法に従って調製した、ルチン1モルにD−グルコースが1モル結合したα−グルコシル ルチンを経口投与して急性毒性試験をしたところ、マウス体重1kg当たり5gまでの投与量では死亡例は見られなかった。また、具体的なデータは示さないが、ルチン1モルにD−グルコースが2乃至5モル結合したα−グルコシル ルチンについても同様の結果が予想される。
実験2−1で用いた糖転移ルチンを、特開平3−27293号公報に開示された方法に従って調製した、ヘスペリジン1モルにD−グルコースが1モル結合したα−グルコシル ヘスペリジン(糖転移ヘスペリジン)で置き換えた以外は、実験2−1と同様に経口投与試験したところ、何れの糖転移ヘスペリジンについても、マウス体重1kg当たり5gまでの投与量では死亡例は見られなかった。また、具体的なデータは示さないが、ヘスペリジン1モルにD−グルコースが2乃至5モル結合したα−グルコシル ヘスペリジンについても同様の結果が予想される。
α,α−トレハロース(登録商標『トレハ』、株式会社林原商事販売)20質量部、プルラン2質量部、精製水50質量部、糖転移ビタミンP類として粉末糖転移ヘスペリジン(商品名『αGヘスペリジンH』(固形分質量当りの全ヘスペリジン含量22乃至26質量%、株式会社林原商事販売)10質量部、糖転移ビタミンC粉末(商品名『AA−2G』、株式会社林原商事販売)0.5質量部、クエン酸1質量部、及び適量の着色料と着香料とをそれぞれ混合し、濾過滅菌して、ガラス製容器に充填して、飲料タイプの健康食品を得た。
ガムベース3質量部を柔らかくなるまで加熱融解し、これにα,α−トレハロース含量約50質量%の粉末緑黄色野菜(商品名『ニューミックス』、株式会社H+Bライフサイエンス販売)を7質量部、及び緑茶エキス1質量部を加え、さらに、適量の着色料及び着香料とともに、糖転移ビタミンP類として粉末糖転移ルチン(商品名『αGルチンP』(固形分質量当りの全ルチン含量40乃至46%、株式会社林原商事販売)を0.5質量部加えた後、常法により練り合わせ、成型し、包装して糖転移ビタミンPを含有するチューインガムタイプの健康食品を得た。
脱脂乳85質量部、脱脂粉乳3質量部、α,α−トレハロース含有シロップ(登録商標『トレハスター』、株式会社林原商事販売)9質量部、寒天0.1質量部、糖転移ビタミンP類として粉末糖転移ルチン(商品名『αGルチンP』(固形分重量当りの全ルチン含量40乃至46%、株式会社林原商事販売)3質量部、粉末糖転移ヘスペリジン(商品名『αGヘスペリジンPA』(固形分質量当りの全ヘスペリジン含量74乃至78%、株式会社林原商事販売)4質量部、ブルーベリーエキス1質量部、及び精製水2質量部を調合タンクに入れ、撹拌しながら55℃に加熱して完全に溶解した。次いで、常法にしたがって混合物を均質化し、殺菌冷却器により殺菌し、スターターを3%(w/w)接種し、プラスチック容器に充填した後、37℃で5時間発酵させて糖転移ビタミンPを含有するヨーグルトタイプの健康食品を得た。
糖転移ビタミンP類として粉末糖転移ヘスペリジン(商品名『αGヘスペリジンPA』(固形分質重部当りの全ヘスペリジン含量74乃至78%、株式会社林原商事販売)1質量部と結晶性α,α−トレハロース粉末(登録商標『トレハオース』、株式会社林原商事販売)9質量部、及び少量の香料を均一に混合した後、遮光性黒色ガラス瓶に50gずつ充填して粉末タイプの健康補助食品を得た。
糖転移ビタミンC粉末(商品名『AA−2G』、株式会社林原商事販売)10質量部に粉末糖転移ヘスペリジン(商品名『αGヘスペリジンPA』((固形分質量当りの全ヘスペリジン含量74乃至78%、株式会社林原商事販売)300質量部、結晶性トレハロース粉末(登録商標『トレハオース』、株式会社林原商事販売)17質量部をそれぞれ均一に混合した後、常法により打錠して、1錠当り糖転移ビタミンPを30mg含有する錠剤を得た。
塩化ナトリウム6質量部、塩化カリウム0.3質量部、塩化カルシウム0.2質量部、乳酸ナトリウム3.1質量部、結晶性トレハロース粉末(登録商標『トレハオース』、株式会社林原商事販売)44質量部、粉末糖転移ルチン(商品名『αGルチンP』(固形分質量当りの全ルチン含量40乃至46%、株式会社林原商事販売)0.5質量部、粉末糖転移ヘスペリジン(商品名『αGヘスペリジンPS』(固形分質量当りの全ヘスペリジン含量80乃至84%、株式会社林原商事販売)1.5質量部及び糖転移ビタミンC(商品名『AA−2G』、株式会社林原商事販売)0.5質量部を、精製水1,000質量部に溶解し、常法にしたがって、膜濾過した後、滅菌したプラスチック製容器に30mlずつ充填して糖転移ビタミンPを含有する液剤を得た。
