Abstract

Claims (19)
- 1つ以上の試料の特定の対象領域中の1つ以上のターゲット分析物の存在を検出する方法であって、
複数の物質および1つ以上のターゲット分析物を含む1つ以上の試料を提供することであって、前記ターゲット分析物のうちの少なくとも一部は蛍光体でラベルが付けられ、前記1つ以上の試料中の前記物質の少なくとも一部に結合すること、
蛍光誘起光で前記1つ以上の試料を照射すること、
前記1つ以上の試料の1つ以上の領域から蛍光光を集めること、
前記1つ以上の試料の少なくとも1回の異方性計測を行うことによって、1つ以上のターゲット分析物が前記物質に結合される1つ以上の対象領域を特定すること、および
前記対象領域からの前記集められた蛍光光を分析することによって、前記1つ以上の試料中の前記物質に結合されたターゲット分析物の存在を決定すること

前記対象領域は、所定のスレッショルド値を超える計測された異方性値を有する前記試料の領域として特定され、

- 請求項1〜7のいずれかに記載の方法であって、前記提供することは、洗浄ステップを伴わないホモジニアスな環境のもとで実行される方法。
- 請求項6に記載の方法であって、前記第2親和性部位は、抗体、抗原、レセプター、リガンド、核酸、酵素、基質、インヒビター、および類似部位からなるグループから選択される方法。
- 請求項1〜9のいずれかに記載の方法であって、前記ターゲット分析物は、抗体、抗原、レセプター、リガンド、タンパク質、ペプチド、酵素、核酸、薬物、ホルモン、化学物質、病原体、毒素、バクテリア、またはウイルスの少なくとも1つを含む方法。
- 請求項1〜10のいずれかに記載の方法であって、前記集めることおよび分析することのステップは、約毎秒20,000物質に達するまで実行される方法。
- 請求項5に記載の方法であって、それぞれの光学的にエンコードされた物質は、その放出した蛍光光に基づいて個別に分類可能である方法。
- 請求項1〜12のいずれかに記載の方法であって、前記集めるステップは、蛍光強度値、蛍光偏光値、蛍光異方性値、回転相関時間、および蛍光寿命のうち1つ以上を集めることを含む方法。
- 請求項1〜13のいずれかに記載の方法であって、前記照射すること、集めることおよび分析することのステップは、複数の蛍光波長領域において実行される方法。
- 請求項1〜14に記載の方法であって、分析することは、前記集められた蛍光光の時間についての変化を計測することによって反応情報を提供することをさらに含む方法。
- 請求項15に記載の方法であって、前記集められた蛍光光の強度値における時間に対しての計測された変化に基づいて、前記1つ以上のターゲット分析物についての濃度を決定することをさらに含む方法。
- 請求項15に記載の方法であって、前記集められた蛍光光の異方性値における時間に対しての計測された変化に基づいて、前記1つ以上のターゲット分析物についての濃度を決定することをさらに含む方法。
- 請求項15に記載の方法であって、前記集められた蛍光光の異方性値における時間に対しての計測された変化に基づいて、結合したターゲット分析物を含む前記複数の物質中の個別の物質を決定することをさらに含む方法。
- 請求項1〜18のいずれかに記載の方法であって、
複数の物質を提供することであって、それぞれの物質は既知の異方性を有することによって、内部異方性レファレンスが形成されることをさらに含み、
少なくとも1つの異方性計測を行うことは、前記計測された異方性を、既知の異方性を持つ前記物質の異方性と比較することによって前記結合されたターゲット分析物の改善された異方性計測を得ることを含む方法。
