試験例1
セリンの美白効果を検証するため、動物の経口摂取による皮膚色素沈着抑制試験を実施した。
(方法)
セリンを通常のモルモット飼料に適量添加し、セリンが0.167重量%配合するモルモット飼料を作成した。ネガティブコントロールとして通常のモルモット飼料を使用した。また、ポジティブコントロールとして美白成分として知られているシステイン(特開2004−203812号公報に開示)が0.267重量%配合するモルモット飼料を作成し、使用した。
茶色モルモット(ウェルターメイプル種)1群7匹のモルモットのそれぞれの群に試験飼料、又は比較対照飼料(ネガティブコントロール、ポジティブコントロール)を自由摂取にて28日間与えた後、背部を剃毛して紫外線(UVB強度:1J/cm2)を、隔日で計3日間照射し、色素沈着の作成を行った。紫外線照射期間および照射後それぞれの群にさらに試験飼料、又は比較対照飼料を自由摂取にて与え、紫外線照射前および紫外線照射後4、7、14、21、28、35日後の色素沈着を以下に示す判定基準にしたがって肉眼判定した。また同時に、色彩色差計(MINOLTA製CR−200)を用いて色素沈着部位の明度差(試験前後のL値)を測定した。各群の色素沈着度(%)は、照射後のL値と照射前のL値の差を求め、ネガティブコントロールにおいて最も色素沈着した値(35日後)を100%として算出した。
(色素沈着の判定基準)
−;色素沈着が認められない、±;わずかな色素沈着が認められる、+;中等度の色素沈着が認められる、++;強度の色素沈着が認められる
(結果)
表1に示すように、セリンを配合した飼料は皮膚色素沈着抑制効果を示した。Test example 1

セリンを通常のモルモット飼料に適量添加し、セリンが0.167重量%配合するモルモット飼料を作成した。コントロールとして通常のモルモット飼料を使用した。1群7匹の茶色モルモット(ウェルターメイプル種)のそれぞれの群に試験飼料、又は比較対照飼料(コントロール)を自由摂取にて与え、飼料摂取後68日後の皮膚水分量を皮膚水分計(courage+khazak製cumeometer CM825)にて測定した。皮膚水分量(%)をコントロール群の皮膚水分量を100%として算出した。
(結果)
表2に示すように、L−セリンが配合した飼料は皮膚水分量増加効果を示した。Test example 2

クリーンベンチ内において培養フラスコ(25cm2)に、牛胎児血清1ml、イーグル最少栄養培地9mlを添加し、そこに3×104個の色素細胞(B16 mouse Melanoma)をまき、炭酸ガス培養器において5%の炭酸ガスを含有する空気下37℃で培養した。細胞が付着したところで、フラスコに試料を、各々最終濃度が50μg/ml、100μg/mlになるように添加し、3日間培養した。比較例としてアミノ酸の1種であるアラニン、ロイシン、プロリンを使用した。なお、ネガティブコントロールは試料を添加せず培養したものを使用した。また、ポジティブコントロールとして、既に本試験系において白色化することが知られているブドウ種子エキスを使用した。3日間培養した色素細胞を顕微鏡観察した後、培養液を除去し、0.02%EDTA(エチレンジアミン テトラアセテート)、次いで0.25%トリプシンを含むダルベッコのリン酸緩衝液を加え剥離した。次いで、剥離した色素細胞にリン酸緩衝液9mlを加え、1000gで5分間遠心して得られた色素細胞の細胞ペレットにより白色化の程度を肉眼により比較した。
(結果)
表3に示すように、セリンにおいて色素細胞の白色化効果が確認された。なお、いずれの系も細胞毒性がないことを肉眼、および細胞数計測で確認した。Test example 3

セリンの美容効果を検証するため、ヒトによる飲用試験を実施した。

実施例1の美容ドリンクを1日1本、35歳以上の女性14名に連続10日間飲用してもらい、飲用前との比較において飲用後の美容実感をアンケートした。
(結果)
肌の状態について飲用前後の違いを7段階で評価したところ、表4に示すように、14人中9人以上が、飲用前よりも「少し良くなった」以上を回答した。また、肌の状態がどのように改善したかとの回答を求めたところ、表5の結果が得られた。
以上のようにセリンを配合した実施例1のドリンクの美容効果が示された。Test example 4
