Claims (1)
- 【特許請求の範囲】 1.多糖類を生体高分子に共有結合させる方法であって、臭化シアンで該多糖 類を活性化し、続いて該活性化された多糖類を式1のアミノチオールリンカー: H2N-[(CH2)m-CHR1-CR2R3-A]q-CHR4-(CHR5)p-CHR6-S-R7 1 (ここでAは直接結合又は式-{Z-(CH2)m-CHR1-CHR2R3}nZ-を有する基である; mは、0〜5の整数である; nは、0〜3の整数である; pは、0〜2の整数である; qは、0又は1の整数である; R1は、水素、又は必要に応じてアミノ、ヒドロキシル、カルボキシル、C1〜 C4アルコキシカルボニル、カルバモイル、モノ-若しくはジ-C1〜C4アルキル カルバモイル若しくはN-(α-カルボキシアルキル)カルバモイルで置換された C1〜C6アルキル、又はmが0でなければ、R1はヒドロキシル、アミノ、若し くはペプチジルアミノである; R2及びR3は、独立に、水素若しくはC1〜C4アルキルであるか、又は両者で オキソ基を形成する; R4は、水素、C1〜C4アルキル、カルボキシル、C1〜C4アルコキシカルボニル、 カルバモイル、モノ-若しくはジ-C1〜C4アルキルカルバモイル若しくはN-(α- カルボキシアルキル)カルバモイルである; R5は、水素、メチル、ヒドロキシ、又はC1〜C7アシルオキシである; R6は、水素又はメチルである; R7は、水素又はチオール保護基又は式:-S-CHR6-(CHR5)p-CHR4-[A-CR2R3- CHR1-(CH2)m]qNH2を有する基である; Zは、イミノ、メチルイミノ、酸素、又は硫黄である)と反応させて、式2 Ps-O-C(=NH)-NH-[(CH2)m-CHR1-CR2R3-A]q-CHR4-(CHR5)p-CHR6-S-R7 2 (ここでPsは多糖類残基であり、A、m、p、q、R1、R2、R3、R4、R5、R6、及 びR7は上に定義した通りである)を有するチオール化多糖類を産生せしめた後、 必要に応じて保護基R7を除去し、該チオール化多糖類を活性化生体高分子と反応 させることを具備する方法。 2.多糖類を生体高分子に共有結合させる方法であって、式3 H2N-(CH2)m-CHR1-CR2R3-A-CHR4-(CHR5)p-CHR6-S-R7 3 (ここで、A、m、p、R1、R2、R3、R4、R5、R6及びR7は請求項1の定義通りで ある)で表わされるアミノ‐チオールリンカーを用いることを具備する方法。 3.R7が、式:-S-CHR6-(CHR5)p-CHR4-[A-CR2R3-CHR1-(CH2)m]q-NH2を有する 基である請求項1又は2に記載の方法。 4.前記チオール保護基R7が-C(=O)-R、-C(=S)-R、-C(=NR)-R、-C(=O)-SR、-C (=S)-NHR、-SO2-OR又は-P(=O)(OR)2のようなアシル、チオアシル又はイミノアシ ル基であり、Rが水素、又はメチル、エチル、アリル、tert-ブチル、フェニル 、ベンジル等のC1-C7ヒドロカルビルである請求項1又は2に記載の方法。 5.R1がα-アミノ酸の側鎖、特に水素、C1-C4アルキル又はα-ヒドロキシC1- C4アルキルである、請求項1‐4のいずれかに記載の方法。 6.R2とR3がオキソ基を形成している請求項1〜5の何れかに記載の方法。 7.Aが、-(NH-CHR1-CO)nNH-の式を持つ基であり、nが0から2までの整数 である、請求項1〜6のいずれかに記載の方法。 8.R4とR5が、水素又はメチル、好ましくは水素であり、p=0又は1である、 請求項1〜7のいずれかに記載の方法。 9.式:-C(=NH)-NH-[(CH2)m-CHR1-CR2R3-A]q-CHR4-(CHR5)p-CHR6-S-(ここで 、A、m、p、q、R1、R2、R3、R4、R5、R6は請求項1〜8の何れか1項で定義し たとおりである)を有する基によって共有結合された2つの生体高分子の複合体 。 10.式:-NH-(CH2)m-CHR1-CR2R3-A-CHR4-(CHR5)p-CHR6-S-(ここで、A、m 、p、R1、R2、R3、R4、R5及びR6は請求項1〜8の何れか1項で定義したとおりで ある)を有する基によって共有結合された2つの生体高分子の複合体。 11.前記2つの生体高分子が多糖類とペプチド又はタンパクである、請求項 10又は11に記載の複合体。 12.請求項9〜11のいずれかに記載の複合体を含有するワクチン。
