Claims (1)
- 【特許請求の範囲】 1.DNAの鋳型指令重合を触媒し、3’−5’エキソヌクレアーゼ(プルーフ リーディング)活性を有し、ピロコッカス・フリオサス(Pyrococcus furiosu s)から得られるDNAポリメラーゼより少なくとも2倍高い複製忠実度を示 すことを特徴とする、テルモコッカス・ゴルゴナリウス(Thermococcus gorgo narius)から得られる、精製された熱安定性DNAポリメラーゼ。 2.安定剤の存在下に約95℃で2時間インキュベーション後に、その活性の約 90%を保持する、請求項1に記載の精製された熱安定性DNAポリメラーゼ 。 3.前記ポリメラーゼが約92000〜96000ダルトンの見かけの分子量を 有する、請求項1〜2のいずれか1項に記載のポリメラーゼ。 4.前記ポリメラーゼが大腸菌(E.coli)から得られる、請求項1〜3のいず れか1項に記載のポリメラーゼ。 5.請求項1〜4のいずれか1項に記載のポリメラーゼおよび安定剤からなる、 安定化された組成物。 6.前記安定剤が非イオン性界面活性剤である、請求項5に記載の組成物。 7.Thesitおよび/またはノニデットP40が安定剤として作用する、請 求項5または6に記載の組成物。 8.テルモコッカス・ゴルゴナリウス(Thermococcus gorgonarius)から得られ る請求項1〜4のいずれか1項に記載のポリメラーゼをコードする、単離され たDNA配列。 9.プラスミドpBTac2Tgo内に含有される、請求項8に記載のポリメラ ーゼをコードする単離されたDNA配列。 10.プラスミドpBTac2Tgoの約2.3kBのEcoRI/PstI制限 断片内に含有される、請求項9に記載の単離されたDNA配列。 11.配列番号6に示す式で表される、単離されたDNA配列。 12.請求項8〜11のいずれか1項に記載の単離されたDNA配列を含有するベク ター。 13.プラスミドpBTac2Tgoである、請求項12に記載のベクター。 14.以下の特徴の一部またはすべてを提供する請求項12および13に記載のベクタ ー: (1)プロモーターまたは転写開始部位 (2)遺伝子発現の開始または停止に使用されるオペレーター (3)翻訳を高めるためのリボソーム結合部位 (4)転写または翻訳終結部位。 15.請求項12〜14に記載のベクターで形質転換された微生物宿主。 16.大腸菌(E.coli)LE392pUBS520に由来し、大腸菌(E.coli) pBTac2Tgoと呼ばれる、請求項15に記載の微生物宿主。 17.請求項1〜7のいずれか1項に記載のDNAポリメラーゼの調製方法であっ て、 (a)天然株テルモコッカス・ゴルゴナリウス(Thermococcus gorgonarius) を培養し、 (b)天然株の細胞を緩衝液中に懸濁し、 (c)細胞を破砕し、 (d)いくつかのクロマトグラフィー工程によりDNAポリメラーゼを精製す る、 ことを含んでなる上記方法。 18.請求項15または16に記載の微生物宿主株を増殖させ、それからDNAポリメ ラーゼを精製することを含んでなる、請求項1〜7のいずれか1項に記載のD NAポリメラーゼの調製方法。 19.請求項1〜7のいずれか1項に記載の熱安定性DNAポリメラーゼを使用す ることを特徴とするDNAの増幅方法。 20.3’−5’エキソヌクレアーゼ活性が不活性化されている、請求項1〜7の いずれか1項に記載の熱安定性DNAポリメラーゼを使用することを特徴とす る、DNA塩基配列決定法またはDNA標識法。 21.請求項1〜7のいずれか1項に記載の熱安定性DNAポリメラーゼを使用す ることを特徴とする、第2cDNAクローニングおよびDNA塩基配列決定の 方法。 22.請求項1〜7のいずれか1項に記載の熱安定性DNAポリメラーゼを使用す ることを特徴とする、DNA塩基配列決定法。
