Claims (1)
- 【特許請求の範囲】 1.マトリックス重合体を用いて固体表面に生物学的結合パートナーを結合さ せる方法であって;下記ステップ、すなわち 核酸とマトリックス重合体を含んでなるマトリックス溶液を調製し; 上記マトリックス溶液を固体表面と接触させて、生物学的結合パートナーを結 合させて、前記生物学的結合パートナーが第二分子を特異的に認識して第二分子 を捕捉できるようにする; ステップを含んでなる方法。 2.請求の範囲1記載の、マトリックス重合体を用いて固体表面に生物学的結 合パートナーを結合させる方法であって; マトリックス溶液を製造するステップが、生物学的結合パートナーを含んでな る水溶液を、マトリックス重合体を含んでなる第二溶液と接触させることからな る方法。 3.請求の範囲1記載の、マトリックス重合体を用いて固体表面に生物学的結 合パートナーを結合させる方法であって; 生物学的結合パートナーが核酸であり、その結合パートナーを固体表面に結合 させて、その核酸が相補的核酸と特異的にハイブリッドを形成することができる ようにする方法。 4.請求の範囲1記載の、マトリックス重合体を用いて固体表面に生物学的結 合パートナーを結合させる方法であって; 固体表面がスライドガラスである方法。 5.請求の範囲1記載の、マトリックス重合体を用いて固体表面に生物学的結 合パートナーを結合させる方法であって; 固体表面が光ファイバーのセンサ末端である方法。 6.請求の範囲1記載の、マトリックス重合体を用いて固体表面 に生物学的結合パートナーを結合させる方法であって; マトリックス重合体がニトロセルロースである方法。 7.請求の範囲1記載の、マトリックス重合体を用いて固体表面に生物学的結 合パートナーを結合させる方法であって; 第二溶液がDMSOに溶解したニトロセルロースを含有している方法。 8.請求の範囲1記載の、マトリックス重合体を用いて固体表面に生物学的結 合パートナーを結合させる方法であって; さらに、マトリックス溶液を固体表面に接触させた後、そのマトリックス溶液 を乾燥させるステップを含んでなる方法。 9.試料中の複数の生物学的結合パートナーを感知する光ファイバー束を構築 する方法であって;下記ステップすなわち、 各ファイバーがセンサ末端と伝送末端を有する複数の光ファイバーを提供し、 各ファイバーには、そのセンサ末端に、一つの種の生物学的結合パートナーを結 合させ、その結合パートナーはマトリックス重合体を用いて光ファイバーのセン サ末端に結合させて、その生物学的結合パートナーが第二分子を特異的に認識し て第二分子を捕捉できるようにし; 異なる結合パートナーを有する前記ファイバーを組み合わせて光ファイバーア レイを製造し、前記ファイバーが、試料を同時に検定するためセンサ末端がとも に整列しており;そして 組み合わされた別個のファイバーの伝送末端を、問合せに対してアドレス指定 して、試料に対する光ファイバーセンサを製造する; ステップを含んでなる方法。 10.請求の範囲9記載の、試料中の複数の結合パートナーを感知する光ファイ バー束を構築する方法であって; マトリックス重合体がニトロセルロースである方法。 11.請求の範囲9記載の、試料中の複数の結合パートナーを感知する光ファイ バー束を構築する方法であって; 生物学的結合パートナーが核酸でありそしてその結合パートナーをセンサ末端 に結合させて、核酸が相補的核酸とハイブリッドを形成することができるように する方法。 12.請求の範囲9記載の、試料中の複数の結合パートナーを感知する光ファイ バー束を構築する方法であって;下記ステップ、すなわち 各ファイバーがセンサ末端と伝送末端を有する複数の光ファイバーを提供し; 複数の光ファイバーを、複数のファイバー群中に入れ、その各ファイバー群は 同時に処理するためセンサ末端をともに整列させ; 各バッチが、光ファイバーの選択された群の光ファイバーのともに整列させた センサ末端に、処理によって結合させるのに適した単一種の生物学的結合パート ナーを含有している複数の異なるバッチを提供し; 異なる生物学的結合パートナーを含有する異なるバッチ内に、異なるファイバ ー群のともに整列させたセンサ末端を入れて処理し、同じ種の生物学的結合パー トナーを有する各群の光ファイバーのセンサ末端および異なる種の生物学的結合 パートナーを有する異なる群の光ファイバーのセンサ末端を有する複数群の光フ ァイバーを製造し;次いで 各ファイバー群からファイバーを選択して、各ファイバーがセンサ末端と伝送 末端を有し、そして各ファイバーがそのセンサ末端に一つの種の生物学的結合パ ートナーが結合されている複数の光ファイバーを提供する; ステップを含んでなる方法。 13.請求の範囲9記載の、試料中の複数の結合パートナーを感知する光ファイ バー束を構築する方法であって; 異なる結合パートナーを有する分離されたファイバーを、センサ末端をともに 整列させて組み合わせることが、そのセンサ末端をランダムに集めることを含ん でなる方法。 14.請求の範囲9記載の、試料中の複数の結合パートナーを感知する光ファイ バー束を構築する方法であって; 異なる結合パートナーを有する分離されたファイバーを、センサ末端をともに 整列させて組み合わせることが、そのセンサ末端を集めて、階段型のセンサ面を 形成することを含んでなる方法。 15.請求の範囲9記載の、試料中の複数の結合パートナーを感知する光ファイ バー束を構築する方法であって; 異なる結合パートナーを有する分離されたファイバーを、センサ末端をともに 整列させて組み合わせることが、前記センサ末端を集めて平坦なセンサ面を形成 することを含んでなる方法。 16.請求の範囲9記載の、試料中の複数の結合パートナーを感知する光ファイ バー束を構築する方法であって; 再び組み合わせた別個のファイバーの伝送末端を問合せのためにアドレス指定 することが、別個のファイバー各々の伝送末端を光アレイにアドレス指定するこ とを含んでなる方法。 17.請求の範囲9記載の、試料中の複数の結合パートナーを感知する光ファイ バー束を構築する方法であって; 類似の結合パートナーを有する各群の光ファイバーの伝送末端が、結合パート ナーに対応する伝送末端を区別する類似のマーキングを有している方法。 18.請求の範囲9記載の、試料中の複数の結合パートナーを感知する光ファイ バー束を構築する方法であって; 異なる結合パートナーの複数のバッチを提供することが、試料中の核酸がハイ ブリッドを形成できる核酸の結合パートナーのバッチを含んでなる方法。 19.請求の範囲18記載の、試料中の複数の結合パートナーを感知する光ファイ バー束を構築する方法であって; 核酸結合パートナーが各々、染色体の特定の領域に対応している方法。 20.請求の範囲18記載の、試料中の複数の結合パートナーを感知する光ファイ バー束を構築する方法であって; 核酸結合パートナーが各々、発現される配列に対応している方法。
