Technical Field
The present application relates to a leuconostoc mesenteroides strain with low gas production and a manufacturing method of kimchi using the same.
Background
Kimchi is a food made by using the fermentation action of microorganisms, in which the food components are decomposed and new components are synthesized by the action of the microorganisms, thereby improving the nutritional value, palatability and storage stability.
The conventional kimchi has problems of expansion, breakage, leakage, etc. of a container due to generation of gas (mainly carbon dioxide) during circulation, and deterioration of taste quality due to strong sour taste caused by over-ripening (ripening).
In order to solve such problems, a method of controlling using various packaging techniques such as a polymer film having high air permeability (korean patent application laid-open No. 10-1999-.
Under such a background, the present inventors have diligently studied to develop a method capable of reducing gas generation in the production process of kimchi. As a result, it was confirmed that when a kimchi was produced using a specific leuconostoc mesenteroides strain, the increase in sour taste was reduced and the gas generation was also reduced, thereby completing the present application.

Technical problem
The purpose of the application is to provide a leuconostoc mesenteroides (KCTC13043BP) strain with low gas production.
It is another object of the present application to provide a starter composition comprising leuconostoc mesenteroides CJLM119 strain.
It is still another object of the present application to provide a kimchi containing leuconostoc mesenteroides CJLM119 strain or the starter composition of the present application.
Another object of the present application is to provide a method for manufacturing kimchi, including the steps of: leuconostoc mesenteroides CJLM119 strain or the starter composition of the present application is brought into contact with a fermentation object.
Technical scheme
The following specifically describes the contents of the present application. In addition, the same matters as those described in the one aspect and the embodiments disclosed in the present application can be applied to the description and the embodiments of the other aspect. Moreover, all combinations of the various elements disclosed in this application are within the scope of this application. Furthermore, the specific description described below should not be construed as limiting the scope of the present application.
To achieve the object of the present application, as one aspect, the present application provides Leuconostoc mesenteroides CJLM119 strain (KCTC13043BP) with low gas production.
According to other embodiments of the present application, the strain of the present application may be a strain with a low acid production amount, and specifically, the acid of the present application may be lactic acid. Therefore, when the strain of the present application is used to produce kimchi, the increase in sourness can be further reduced compared to the case of using another leuconostoc mesenteroides strain to produce kimchi, and the taste quality of kimchi can be maintained.
According to yet another embodiment of the present application, the strain of the present application may be a strain having a high production amount of mannitol. Mannitol imparts a cool and refreshing taste to kimchi, and mannitol inhibits not only sourness but also propagation of rancidity bacteria, thereby preventing excessive fermentation of kimchi.
According to still another embodiment of the present application, the strain of the present application shows negative in all of the gelatin liquefaction reaction, the production of harmful metabolites (e.g., ammonia), the deamination of phenylalanine, and the examination of the phenomenon of hemolyzing, and thus is safe even when applied to food (see example 5).

According to an embodiment of the present application, the starter composition of the present application may be in the form of a liquid or a powder.
According to another embodiment of the present application, the starter composition of the present application may further comprise a cryoprotectant, and more particularly, may further comprise at least one cryoprotectant selected from the group consisting of glycerol, trehalose, maltodextrin, skim milk powder, and starch. The cryoprotectant of the present application may be contained in an amount of 0.01 wt% to 20 wt% or 0.01 wt% to 10 wt% with respect to the total weight of the starter composition of the present application, and specifically, in the starter composition of the present application, glycerol may be contained in an amount of 5 wt% to 20 wt%, trehalose may be contained in an amount of 2 wt% to 10 wt%, maltodextrin is contained in an amount of 2 wt% to 10 wt%, skim milk powder may be contained in an amount of 0.5 wt% to 2 wt%, and starch may be contained in an amount of 0.1 wt% to 1 wt%.
According to yet another embodiment of the present application, the starter composition of the present application may further comprise an excipient. Specifically, the excipient of the present application may be at least one selected from the group consisting of glucose, dextrin, and skim milk. More specifically, the excipient herein may be present in an amount of 75 to 95% by weight or 85 to 95% by weight, relative to the total weight of the starter composition herein.
As yet another aspect, the present application provides a kimchi containing the leuconostoc mesenteroides CJLM119 strain (KCTC13043BP) of the present application or the starter composition of the present application.
The term "kimchi" in the present application refers to a food obtained by salting vegetables (e.g., chinese cabbage, radish, onion, mustard, cucumber, etc.) and then mixing seasonings (e.g., paprika, garlic, ginger, fish sauce, etc.) and fermenting them.
The kimchi of the present application may further include known additives acceptable as foods, and specifically, may further include: natural perfumes such as plum, lemon, pineapple, vanilla, and the like; natural pigments such as natural fruit juices, chlorophyllin, flavonoids, and the like; sweet components such as fructose, honey, sugar alcohol, white sugar, etc.; or acidulants such as citric acid, sodium citrate, and the like.
As another aspect, the present application provides a method for manufacturing kimchi, including the steps of: the leuconostoc mesenteroides CJLM119 strain (KCTC13043BP) of the present application or the starter composition of the present application is contacted with a fermentation object.
According to still another embodiment of the present application, 0.01 to 3 wt%, 0.1 to 3 wt%, 0.5 to 2 wt% of the leuconostoc mesenteroides CJLM119 strain (KCTC13043BP) of the present application or the starter composition of the present application may be contacted with the fermentation object based on the total weight of the fermentation object.
Further, the contacting of the present application can be carried out at 3 ℃ to 10 ℃ or 5 ℃ to 10 ℃ for 1 day to 90 days, 10 days to 60 days, 20 days to 60 days, or 20 days to 40 days.
In still another aspect, the present application provides kimchi produced by the production method of the present application.
Effects of the invention
Leuconostoc mesenteroides CJLM119 strain (KCTC13043BP) of the present application has the following effects: since the amount of gas generated is low, when kimchi is produced using the above-mentioned strain or its culture as a starter, the amount of gas generated during circulation is reduced to prevent problems such as breakage of packaging due to gas generation, thereby enabling to stabilize the circulation quality of kimchi. Further, the following effects are also provided: the taste quality of kimchi is improved by maintaining a predetermined sour taste due to a low amount of acid produced and by having an excellent potential for producing mannitol.
Detailed Description
The present application will be described more specifically with reference to the following examples. However, these examples are only intended to aid the understanding of the present application, and the scope of the present application is not limited to these examples in any sense.
Example 1: isolation of strains with lower gas production
1-1) isolation and identification of the Strain
Various kimchi purchased from a large supermarket is aged at a low temperature of 5 ℃, and the kimchi reaching a ph range of 3.8 to 4.5 is used as a kimchi sample. The kimchi sample was diluted 10-fold with 0.85% saline solution, inoculated onto a PES agar medium (phenethyl alcohol sucrose agar; tryptone 5g, yeast extract 0.5g, sucrose 20g, ammonium sulfate 2g, dipotassium hydrogen phosphate 1g, magnesium sulfate 0.244g, phenethyl alcohol 2.5ml, agar 15 g/distilled water 1L) plate, and then smeared with a spreader. Then, after the above plates were cultured in an incubator at 25 ℃ for 24 hours, the individual colonies generated were streaked (streaking) onto other PES agar plates to be isolated as single colonies.
1-2) screening of strains with lower gas production
Colonies of the strain isolated from the above example 1-1 were inoculated into 10ml of MRS broth [ difco. MRS broth (broth) in a test tube containing a dellam tube having a height of 30mm, respectively; bacterial peptone (bactopeptone)10g, beef extract (beef extract)10g, yeast extract 5g, glucose 20g, tween80 (tween80)1g, ammonium citrate 2g, dipotassium hydrogen phosphate 2g, sodium acetate 5g, manganese sulfate 0.1g, magnesium sulfate 0.05g, and distilled water 1L ], followed by static culture at 25 ℃ for 24 hours. The degree of gas production was measured by measuring the height of the gas trapped in the Delamm tube, and strains having a gas production of 10mm or less were selected.
1-3) screening of strains with reduced acid production
The bacterial colonies isolated from the above examples 1-2 were inoculated into 10ml of MRS broth (Difco. MRS broth; Bacto peptone 10g, beef extract 10g, Yeast extract 5g, glucose 20g, Tween 801 g, ammonium citrate 2g, dipotassium hydrogen phosphate 2g, sodium acetate 5g, manganese sulfate 0.1g, magnesium sulfate 0.05g, distilled water 1L), respectively. To measure the degree of acid production, after resting culture at 25 ℃ for 24 hours, the pH was measured using a pH meter (SevenCompact/IonS 220, Mettler Toledo) and strains having a pH of at least 4.4 were selected.
1-4) screening of mannitolStrain with high production amount
The colonies of the strains isolated from the above examples 1 to 3 were respectively inoculated onto 10ml of a minimum medium (bactopeptone 10g, fructose 20g, tween 801 g, ammonium citrate 2g, dipotassium hydrogen phosphate 2g, sodium acetate 5g, manganese sulfate 0.1g, magnesium sulfate 0.05 g/distilled water 1L) containing 2% fructose (fructose), and the amount of mannitol produced was measured after static culture at 25 ℃ for 24 hours. Mannitol production was measured by HPLC, and strains producing mannitol in an amount of at least 16000mg/L were selected.
1-5) identifying the selected strains
The name of the strain selected in examples 1 to 4 was named CJLM119, and its 16srRNA base sequence (SEQ ID NO: 1) was analyzed. As a result, it was confirmed that the 16s rRNA base sequence of CJLM119 strain was 99% identical to the 16s rRNA base sequence of Leuconostoc mesenteroides NRIC 1517 (SEQ ID NO: 2). Thus, the above CJLM119 strain was named "Leuconostoc mesenteroides CJLM 119" and deposited at 10/6 in 2016 to Korean institute of Life engineering (Korean Type culture Collection for Type Cultures) and was given the accession number KCTC13043 BP.
Example 2: comparative gas generating Capacity
In order to compare the gas production degree of leuconostoc mesenteroides CJLM119 screened in example 1 with other leuconostoc mesenteroides strains, leuconostoc mesenteroides KCTC3100 and KCTC3722, which are standard strains of leuconostoc mesenteroides, were used as a control group and the gas production degree was measured. Each strain was inoculated into 10ml of MRS broth in a tube containing a dellam tube having a height of 30mm, and after 24 hours of culture at 25 ℃, the degree of gas production was compared by measuring the height of gas trapped in the dellam tube.
The degree of gas generation is expressed as: "-" in the case of no gas generation, "+" in the case of a gas height of 1mm to 5mm, "+ +", in the case of a height of 6mm to 10mm, "+ + + + + + + +", in the case of a height of 11mm to 15mm, "+ + + + + + + + + +", in the case of a height of 16mm to 25mm, and "+ + + + + + + + +", in the case of a height of 25mm or more.
As a result, it was confirmed that the gas production level of leuconostoc mesenteroides CJLM119 was "++" and significantly decreased compared to "+++++" or "++++" as the gas production level of the control group strain (table 1).

Example 3: comparison of acid Generation ability
The acid production degree of leuconostoc mesenteroides CJLM119 screened in example 1 was compared with that of leuconostoc mesenteroides KCTC3100 and KCTC3722 as control groups. Each strain was inoculated into 10ml of MRS broth, respectively, and in order to measure the degree of acid production, the pH after stationary culture at 25 ℃ for 24 hours was measured using a pH meter (SevenCompact/Ion S220, MettlerToledo).
As a result, it was confirmed that the acid production amount of leuconostoc mesenteroides CJLM119 was significantly reduced compared to the control strain (table 2).

Example 4: comparison of mannitol Generation ability
The mannitol-producing ability of leuconostoc mesenteroides CJLM119 screened in example 1 was compared with that of leuconostoc mesenteroides KCTC3100 and KCTC3722 as control groups. Each strain was inoculated into 10ml of a minimum medium containing 2% fructose (bactopeptone 10g, fructose 20g, Tween 801 g, ammonium citrate 2g, dipotassium hydrogen phosphate 2g, sodium acetate 5g, magnesium sulfate 0.1g, magnesium sulfate 0.05 g/distilled water 1L) respectively, and after culturing at 25 ℃ for 24 hours, the mannitol content in the supernatant was measured by HPLC.
As a result, it was confirmed that the production amount of mannitol of Leuconostoc mesenteroides CJLM119 was increased by 147% and 187%, respectively, as compared with the control group strain (Table 3).

Example 5: evaluation of safety of Strain
In order to confirm whether the leuconostoc mesenteroides CJLM119 screened in example 1 can be used as a starter in the production of kimchi, the harmfulness of the strain was analyzed. Specifically, Hemolysis (haemolysis), gelatin liquefaction (gelitiniquefaction), confirmation of harmful metabolite (Ammonia) Production (Ammonia Production) and Phenylalanine deamination (Phenylalanine deaminase) were performed according to the safety evaluation test method proposed by the korean society for biological risk investment.
As a result, it was confirmed that leuconostoc mesenteroides CJLM119 strain was negative in all of the gelatin liquefaction reaction, the production of harmful metabolite (ammonia), the deamination of phenylalanine, and the hemolysis test, and thus the leuconostoc mesenteroides CJLM119 strain was a safe strain that can be administered to human bodies, and the strain could be used in food production (table 4).

Example 6: preparation method of spicy Chinese cabbage
6-1) production of kimchi using Leuconostoc mesenteroides CJLM119 strain
About 10g of a medium sterilized by mixing 2.5g of sucrose, 2.0g of sodium citrate, 1.5g of peptone, 1.0g of glucose, 1.0g of yeast extract, 0.5g of fructose, 0.5g of sodium acetate and 1L of distilled water was inoculated9CFU/ml of leuconostoc mesenteroides CJLM119 strain, and cultured at 25 ℃ for 24 hours, thereby preparing a strain broth, wherein the inoculated leuconostoc mesenteroides CJLM119 strain was 1 wt% with respect to the total weight of the medium.
Then, a seasoning was prepared by adding the strain culture solution prepared above to a general kimchi seasoning obtained by mixing chili powder (2.5 wt%), garlic (2 wt%), ginger (0.4 wt%), green Chinese onion (1 wt%), radish (18 wt%), and fish sauce (5 wt%), and then mixing with pickled cabbage to produce kimchi (experimental example 1), wherein the added strain culture solution was 0.1 wt% with respect to the total weight of the kimchi.
6-2) preparing kimchi by using standard strain culture solution
Kimchi was produced in the same manner as in example 6-1 using the culture solutions of the standard strains KCTC3100 and KCTC3722 of leuconostoc mesenteroides, respectively (KCTC 310 culture solution: comparative example 1 and KCTC3722 culture solution: comparative example 2).
6-3) preparing kimchi without adding strain culture solution
Kimchi was produced in the same manner as in example 6-1 above, except that no strain culture fluid was added to the kimchi seasoning (comparative example 3).
Example 7: analysis of spicy cabbage characteristics
7-1) comparison of acid production amounts
The kimchi of the above experimental example 1 and comparative examples 1 to 3 was stored at 7 ℃ for 30 days and the respective lactic acid production amounts were measured. Specifically, a kimchi juice was prepared by grinding a predetermined amount of each kimchi and filtering the crushed kimchi once with gauze, and in order to analyze the amount of lactic acid in the kimchi juice, 3ml of kimchi juice was extracted and subjected to centrifugal analysis (10000g, 10 minutes), and then filtered twice with a 0.2 μm filter, and the content of lactic acid was analyzed by HPLC.
As a result, it was confirmed that the lactic acid production amount of the experimental example 1 reached a level of 65% to 67% as compared with the kimchi of the comparative example 3 not produced using the strain culture solution and the kimchi of the comparative examples 1 and 2 produced using the standard strain culture solution of the same species, and that the increase in sour taste was reduced and the taste quality of the kimchi was maintained in the case of the production of the kimchi using the leuconostoc mesenteroides CJLM119 strain culture solution (table 5).

7-2) comparison of gas production
A predetermined amount of the kimchi of the above experimental example 1 and comparative examples 1 to 3 was packed in an aluminum bag (Al pouch) to which no gas absorbent was attached and stored at 7 ℃ for 30 days, and the gas generation amount of each kimchi was measured by measuring the increased volume.
As a result, it was confirmed that the gas production amount of the experimental example 1 was as high as 57% to 62% compared to the comparative examples 1 and 2, which are kimchi produced using the same standard strain culture fluid, and was lower than that of the comparative example 3, which is kimchi produced without using the strain culture fluid, that the gas production amount was significantly reduced and the convenience of circulation was increased in the case of the kimchi produced using the leuconostoc mesenteroides CJLM119 strain culture fluid (table 6).

7-3) comparison of mannitol production
The kimchi of the above experimental example 1 and comparative examples 1 to 3 was stored at 7 ℃ for 30 days and the respective mannitol production amounts were measured. Specifically, a kimchi juice was prepared by grinding a predetermined amount of each kimchi and then filtering with gauze, and 3ml of the above kimchi juice was extracted and centrifuged (10000g, 10 minutes), and then filtered twice with a 0.2 μm filter, and then the mannitol content was analyzed by HPLC.
As a result, it was confirmed that the production amount of mannitol in experimental example 1 reached a level of 167% to 199% compared to comparative example 3 which was not produced using a strain culture solution and comparative examples 1 and 2 which were produced using a standard strain culture solution of the same species, and it was confirmed that kimchi having excellent storage stability and taste quality due to the inclusion of a large amount of mannitol was produced in the case of producing kimchi using a leuconostoc mesenteroides CJLM119 strain culture solution (table 7).

Claims (6)

1. A Leuconostoc mesenteroides CJLM119 strain having a lower gas production amount as compared with Leuconostoc mesenteroides KCTC3100 or KCTC3722, wherein the Leuconostoc mesenteroides CJLM119 strain has a accession number of KCTC13043 BP.
2. A starter composition comprising Leuconostoc mesenteroides CJLM119 strain or a culture thereof, wherein the Leuconostoc mesenteroides CJLM119 strain has a accession number of KCTC13043 BP.
3. A starter composition according to claim 2 wherein,
the starter composition comprises 107The Leuconostoc mesenteroides CJLM119 strain of cfu/ml or more, wherein the Leuconostoc mesenteroides CJLM119 strain has a accession number of KCTC13043 BP.
4. A starter composition according to claim 2 wherein,
the starter composition further comprises at least one cryoprotectant selected from the group consisting of glycerol, trehalose, maltodextrin, skim milk powder and starch.
5. A kimchi comprising the leuconostoc mesenteroides CJLM119 strain according to claim 1 or the starter composition according to any one of claims 2 to 4, wherein the leuconostoc mesenteroides CJLM119 strain has a accession number of KCTC13043 BP.
6. A method for preparing kimchi comprises the following steps: the Leuconostoc mesenteroides CJLM119 strain according to claim 1 or the starter composition according to any one of claims 2 to 4 is contacted with a fermentation object, wherein the Leuconostoc mesenteroides CJLM119 strain has a accession number of KCTC13043 BP.
