Compound with anticancer effect and anti-helicobacter pylori activity, preparation method and application thereof
The present application is a divisional application of a patent application having application number 201480023381.3 and entitled "new compound and method for producing same, and use thereof" filed 2, 24/2014.
Technical Field
The present invention relates to a compound having an anticancer effect and an anti-Helicobacter pylori activity, a method for producing the same, and a composition, an anticancer agent, and an anti-Helicobacter pylori (Helicobacter pylori) agent containing the compound.
Background
Cancer tissue is composed of a mixture of cancer cells and surrounding normal tissue called stroma. The following is increasingly evident: the stroma is composed of various factors such as blood vessels, extracellular matrix, and fibroblast-like cells (also simply referred to as "stromal cells"), and is closely involved in the growth of cancer. Among the above-mentioned mesenchymes, in particular, mesenchyme cells are known to control the proliferation of cancer cells positively and negatively through adhesion and secretion factors (see, for example, non-patent document 1). Under such circumstances, the search for new useful anticancer agents has been conducted, and the rapid provision thereof has been strongly demanded.
In gastric and duodenal lesions such as gastric ulcer and duodenal ulcer, it is known that the lesions are caused by Helicobacter pylori (Helicobacter pylori). Then, quinolinone compounds have been proposed as compounds having anti-helicobacter pylori activity (see, for example, non-patent document 2 and patent document 1). However, the proposed quinolinone compounds cannot be said to be sufficient as a pharmaceutical product, and new compounds having anti-helicobacter pylori activity are required.

The present invention has been made in view of the above-described conventional techniques, and an object thereof is to achieve the following objects. That is, an object of the present invention is to provide a novel compound having an excellent anti-cancer effect or an excellent anti-helicobacter pylori activity, a method for producing the novel compound, a compound-containing composition using the novel compound, an anti-cancer agent, and an anti-helicobacter pylori agent.
Means for solving the problems

After stirring at room temperature for 12 hours, a 10% aqueous hydrochloric acid solution was added dropwise thereto in an ice bath. After the addition was complete, sodium hydroxide was added in an ice bath to adjust the pH to 7. The organic layer was separated and the aqueous layer was extracted with diethyl ether. The organic layers were combined and dried over sodium sulfate and the solvent was distilled off. The obtained residue can be purified by silica gel chromatography (hexane: ethyl acetate ═ 6: 1) to obtain a compound represented by structural formula (14).

Sodium methyl mercaptide was added thereto, and further stirred for 1 hour. Aqueous ammonium chloride solution was added thereto, followed by extraction with ethyl acetate. The alkali metal hydride is not particularly limited and can be appropriately selected according to the purpose, and for example, sodium hydride can be mentioned.

Further, octanoyl chloride as an acid chloride was added dropwise in an ice bath, and the mixture was stirred at room temperature. The reaction was stopped with 0.1N hydrochloric acid, extracted with dichloromethane, and washed with a saturated aqueous sodium bicarbonate solution and brine. The combined organic layers were dried over sodium sulfate and the solvent was distilled off. Production of the compound represented by the formula (20) -
The compound represented by the structural formula (20) can be produced, for example, as follows.
Sodium hydroxide is added to a 0.1M dioxane solution of the compound represented by the structural formula (15), and the mixture is stirred at 110 ℃ for 1 to 2 hours. The reaction solution was returned to room temperature, water was added, and 1N hydrochloric acid was further added until the pH became 7. Further, hexane was added thereto and ultrasonic waves were applied thereto, whereby a solid precipitated. The solid was filtered with suction and washed with water, hexane or hexane/ethyl acetate ═ 1: 1. It can be washed and then dried to obtain the compound represented by the structural formula (20).

The compound represented by the structural formula (20) was dissolved in THF under argon atmosphere, a THF solution of lithium tert-butoxide was added, and the mixture was stirred at room temperature for 20 minutes. The reaction conditions, the compound used, the amount thereof used, the solvent, and the like in the production method of the compound represented by each structural formula are not particularly limited as long as the effect of the present invention is not impaired, and can be appropriately selected according to the purpose.
Whether or not each of the compounds has a structure represented by each structural formula can be confirmed by appropriately selected various analysis methods, and examples thereof include analysis methods such as the mass spectrometry, the ultraviolet spectrometry, the infrared spectrometry, the proton nuclear magnetic resonance spectrometry, and the carbon-13 nuclear magnetic resonance spectrometry.

Other ingredients-
The other components are not particularly limited, and may be appropriately selected from pharmacologically acceptable carriers according to the purpose, and examples thereof include additives, adjuvants, and water. These may be used alone or in combination of two or more.

The isotonic agent is not particularly limited and may be appropriately selected according to the purpose, and examples thereof include sodium chloride and glucose.
The content of the other component in the compound-containing composition is not particularly limited, and can be appropriately selected according to the purpose within a range that does not impair the effects of the compound represented by any one of the structural formulae (1) to (13).
Application-
The compound-containing composition contains a compound represented by any one of the structural formulae (1) to (13), and therefore has an excellent anticancer effect, an excellent activity against helicobacter pylori, and high safety, and can be preferably used in, for example, pharmaceutical compositions, anticancer agents, anti-helicobacter pylori agents, and the like.
The compound-containing composition may be used alone or in combination with a pharmaceutical preparation containing other ingredients as active ingredients. The compound-containing composition may be used in a state of being incorporated in a pharmaceutical product containing another component as an active ingredient.
< anticancer agent >
The anticancer agent of the present invention contains at least a compound represented by any one of the structural formulae (1) to (13), and further contains other components as necessary.

Application-
The anticancer agent contains a compound represented by any one of the structural formulae (1) to (13), and therefore has an excellent anticancer effect and high safety, and can be suitably used as a prophylactic or therapeutic agent for a wide range of cancers such as gastric cancer, prostate cancer, lung cancer, colorectal cancer, pancreatic cancer, and breast cancer. Among these, the use in gastric cancer and colorectal cancer is particularly preferable.
The anticancer agent may be used alone or in combination with a drug containing other components as active ingredients. The anticancer agent may be used in a state of being incorporated in a pharmaceutical product containing other components as active ingredients.
As shown in test examples described later, the compounds represented by the structural formulae (1) to (13) of the present invention can further inhibit the proliferation of cancer cells in the presence of normal mesenchymal cells.
The anticancer agent contains a compound represented by any one of the structural formulae (1) to (13), and therefore, when administered to an individual, it is possible to prevent carcinogenesis in the individual or treat the individual suffering from cancer. Therefore, the present invention also relates to a method for preventing or treating cancer characterized by administering the anticancer agent to an individual.
< anti-helicobacter pylori agent >
The anti-helicobacter pylori agent of the present invention contains at least a compound represented by any one of the structural formulae (1) to (13), and further contains other components as required.
One compound represented by any one of the structural formulae (1) to (13) may be used alone, or two or more compounds may be used in combination.
The content of the compound represented by any one of the structural formulae (1) to (13) in the anti-helicobacter pylori agent is not particularly limited and can be appropriately selected according to the purpose. The anti-helicobacter pylori agent may be a compound represented by any one of the structural formulae (1) to (13) itself.
Other ingredients-
The other components are not particularly limited and may be appropriately selected according to the purpose, and examples thereof include the same components as those described in the compound-containing composition. These may be used alone or in combination of two or more.

Application-
The agent for controlling helicobacter pylori contains a compound represented by any one of the structural formulae (1) to (13), and therefore, has excellent activity against helicobacter pylori and high safety, and can be preferably used as a preventive or therapeutic agent for gastric and duodenal lesions caused by helicobacter pylori, such as gastric ulcer and duodenal ulcer.
The anti-helicobacter pylori agent may be used alone or in combination with a drug containing other ingredients as active ingredients. The anti-helicobacter pylori agent may be used in a state of being incorporated in a pharmaceutical product containing other components as active ingredients.
The anti-helicobacter pylori agent contains a compound represented by any one of the structural formulae (1) to (13), and therefore, by administering to an individual, it is possible to prevent the individual from being infected with helicobacter pylori or to treat the individual infected with helicobacter pylori. Therefore, the present invention also relates to a method for preventing or treating an infection caused by helicobacter pylori, which is characterized by administering the anti-helicobacter pylori agent to an individual.
In addition, by administering the anti-helicobacter pylori agent to an individual, it is possible to prevent the occurrence of gastric and duodenal lesions caused by helicobacter pylori, or to treat an individual suffering from gastric and duodenal lesions caused by helicobacter pylori. Therefore, the present invention also relates to a method for preventing or treating gastric and duodenal damages caused by helicobacter pylori, characterized by administering the anti-helicobacter pylori agent to an individual.
Dosage form
The dosage forms of the compound-containing composition, the anticancer agent, and the anti-helicobacter pylori agent are not particularly limited and can be appropriately selected according to the purpose, and examples thereof include a solid agent, a semisolid agent, and a liquid agent. The compound-containing composition, the anticancer agent, and the anti-helicobacter pylori agent in these dosage forms can be produced according to conventional methods.
Solid agents
The solid agent is not particularly limited, and may be appropriately selected according to the purpose, and when used as an internal preparation, examples thereof include tablets, chewable tablets, effervescent tablets, orally disintegrating tablets, troches, drops, hard capsules, soft capsules, granules, powders, pills, dry syrups, infusions, and the like.
When the solid agent is used as an external preparation, examples thereof include suppositories, poultices, plasters and the like.
Semi-solid agents-
The semisolid agent is not particularly limited, and may be appropriately selected according to the purpose, and when used as an internal preparation, examples thereof include licking agents (creams), masticatory agents, fluffy gels (jellies), and the like.
When the semisolid agent is used as an external preparation, for example, an ointment, a cream, a mousse, an inhalant, a Nazar gel, and the like can be given.
Liquid agents
The liquid agent is not particularly limited and may be appropriately selected according to the purpose, and when used as an internal preparation, examples thereof include syrup, drink, suspension, spirit, and the like.
When the liquid preparation is used as an external preparation, examples thereof include a liquid preparation, an eye drop, an aerosol, and a spray.
< administration >
The compound-containing composition, the method of administration of the anticancer agent and the anti-helicobacter pylori agent, the dose, the administration time, and the administration subject are not particularly limited, and can be appropriately selected depending on the purpose.
Examples of the administration method include a topical administration method, an enteral administration method, a non-oral administration method, and the like.
The dose is not particularly limited, and can be appropriately selected in consideration of various factors such as age, body weight, body constitution, symptoms of the subject to be administered, and whether or not a drug or medicament containing another component as an active ingredient is administered.
The species of the animal to be administered is not particularly limited, and can be appropriately selected according to the purpose, and examples thereof include humans, monkeys, pigs, cows, sheep, goats, dogs, cats, mice, rats, birds, and the like, and among these, humans are preferably used.

The present invention will be specifically described below by way of production examples and test examples of the present invention, but the present invention is not limited to these production examples and test examples. The data of the following test examples are representative data of experiments performed twice or three times independently, and the same results were obtained. Aminobenzonitrile (15.0g, 128mmol) was dissolved in 300mL of anhydrous tetrahydrofuran (hereinafter sometimes referred to as "THF") under an argon atmosphere, and ethylmagnesium bromide (127mL, 383mmol) was added dropwise under an ice bath. After stirring at room temperature for 12 hours, a 10% aqueous hydrochloric acid solution (100mL) was added dropwise thereto while cooling on ice. The diluted cell suspension was added to 50. mu.L/well of HI medium containing the test sample at each concentration, and the mixture was allowed to stand and cultured at 37 ℃ for 18 hours under aerobic conditions containing 5% carbonic acid gas.
After the culture is finished, whether the bacteria proliferate is judged by turbidity vision, and the MIC of the strain is obtained. The results are shown in FIG. 3.

As shown in Table 3, each of the compounds showed anti-helicobacter pylori activity. Among these, the compound represented by the structural formula (3), the compound represented by the structural formula (11), and the compound represented by the structural formula (12) all show anti-helicobacter pylori activity at low concentrations.
On the other hand, each of the compounds described above has weak antibacterial activity against other common infectious pathogenic bacteria.
The compounds represented by any one of structural formulae (1) to (13) of the present invention have an excellent anticancer effect or an excellent activity against helicobacter pylori, and are highly safe, and therefore, can be preferably used as active ingredients of pharmaceutical compositions, anticancer agents, anti-helicobacter pylori agents, and the like.

Claims (6)

1. A compound represented by any one of the following structural formulae (1) or (2),
wherein, in the structural formula (1), Me represents methyl.
2. A process for producing a compound represented by the following formula (1), which comprises reacting a compound represented by the following formula (21) with at least one of an alkali metal carbonate and an alkali metal hydride with methyl bromoacetate,
wherein, in the structural formula (1), Me represents methyl.
3. A method for producing a compound represented by the following structural formula (2), characterized in that a compound represented by the following structural formula (1) is reacted with at least one of an alkali metal hydroxide and an alkaline earth metal hydroxide, and then the pH of the reaction product is made acidic,
wherein, in the structural formula (1), Me represents methyl.
4. A composition comprising a compound of claim 1.
5. An anticancer agent comprising the compound according to claim 1.
6. An anti-helicobacter pylori agent comprising the compound according to claim 1.
