Abstract

An improved Castle's intrinsic factor concentrate is obtained from desiccated and defatted hog pyloric or stomach mucosa by treating with sodium chloride to produce a precipitate, separating the liquor therefrom, adjusting the pH of the liquor to about 9, acidifying the liquor to about pH 1.5 and removing the precipitate thus formed, adjusting the pH of the supernatent to about 4.5, adding portionwise thereto seven successive portions of solid ammonium sulphate in an amount such that a concentration of at least 35% by weight is eventually achieved, separating and collecting the resultant precipitates formed upon each addition, mixing each with water and dialysing the mixture and isolating the resulting intrinsic factor concentrate, e.g. by freeze-drying. The concentrate obtained by precipitation at between about 35 and 60% ammonium sulphate saturation is stated to be particularly potent.
