Abstract

The circulatory hormone callicrein is obtained from animal pancreas or submaxillary glands by forming an aqueous mixture of the animal substance, adjusting the hydrogen ion concentration of the mixture to pH 4 to 7, heating the aqueous mixture to 45 to 60 DEG C, and removing the aqueous component from the separated fatty and proteinaceous material. According to the example, fresh hog pancreas is minced, suspended in water, brought to a pH 4.6 by the addition of hydrochloric acid and stirred on e.g. a water bath until a temperature of 55 DEG C has been reached. The hot solution is centrifuged or allowed to stratify, and the aqueous solution removed. It is preferred to use the pancreas or submaxillary glands of the hog, and a thermolyis temperature of 55 DEG C, as in the example, when substantially all the hormone is liberated from the animal tissue within minutes of reaching this temperature. At 50 DEG C ten minutes is required, and at 48 DEG C one hour is required to ensure that the hormone is fully extracted into the aqueous phase.
