Abstract

The invention relates to the cloning and characterisation of a novel gene from M tuberculosis, mpt83 (Sequence ID No 2) encoding the protein MPT83 (Sequence ID No 1). Also disclosed are various substituent parts of the protein and gene including the glycosylation sequence, lipoylation sequence, secretion sequence and promoter region, and analogues and derivatives (generally 70 % homology) of these. The gene and its various aspects have utility in generating recombinant DNA expression systems for use in the transformation of cells e.g. to produce glycosylated or lipoylated products, or which are regulatable e.g. with macrophage factors. The invention also relates to recombinant antigens employing MPT83 or using the mpt83 promoter and vaccines based on the above systems.
