Abstract
A composition, for use in reducing the cholesterol level in patients, comprises soya protein and 2 to 3% by weight, on a dry solids weight basis, of pectin, the composition being in the form of granules. The composition can be administered as it is or in a (liquid) foodstuff or beverage; administration is easier than for conventional powdered compositions.
Description
SPECIFICATION
Hypo - p - liproproteinaemic dietetic composition
The invention relatesto a hypo-6- lipoproteinaemic dietetic composition which contains vegetable protein and pectin.
It is known that, in contrast to proteins of animal origin, vegetable proteins, in particular soya protein (soya bean protein) act to reduce elevated serum cholesterol levels in animals and humans; they are also valuable in weight-reducing diets.
It is also known that pectin causes a moderate reduction of elevated cholesterol levels. Pectin has other therapeutically valuable properties; for example, administration of pectin shortens the passage time in the gastrointestinal tract, increases the tonus of the gastrointestinal tract and thus suppression of the sensation of hunger, and prevents digestion disorders, such as constipation and fermentative dyspepsias and adsorption of toxic intestinal contents.
Pectin can accordingly be used for the prophylaxis of gallstone illnesses and of colon diverticulosis.
A "cholesterol-minus diet" composition in powder form, which contains whole soya and apple pectin is alsb known, but this composition has a very high fat content (11 grams per 100 grams of composition).
We have found that this known powdered composition can be taken and swallowed only with difficulty.
We have now developed an improved dietetic composition containing soya protein and pectin which is unexpectedly easier to administer than eitherthe separate ingredients or the known "cholesterol-minus diet" composition referred to above.
The dietetic composition according to the invention comprises soya protein and 2 to 3% by weight, on a dry solids weight bases, of pectin, the composition being in the form of granules.
Administration of the composition according to the invention leads to reduction of the cholesterol level by reduction of the LDL (low density lipoprotein) cholesterol content. From this, it can be concluded that the particularly atherogenic LDL lipoprotein levels are also reduced. Elevated triglyceride levels are also reduced. Overloading of the pepsin/trypsin system by a protein-rich diet alone can be prevented or at least moderated by administration of the composition according to the invention. The pectin present in the composition swells in the gastrointestinal tract; thus both the sensation of hunger is reduced and an effect necessary for digestion is produced, so that the composition can be used effectively as part of a weight-reducing diet.Compared with other known agents for reducing cholesterol levels, the composition according to the invention is particularly well tolerated physiologically.
As a result of its granular form, the composition according to the invention can be taken considerably more easily than the known "choiesterol-minus diet". It can be swallowed with or without a liquid, or chewed and the swelling properties of the pectin, which interfere with the acceptance of the known diet referred to, cannot be detected. Thanks to its very much lower fat content, the dietetic composition according to the invention is physiologically more favourable than the known dietetic powder.
The composition according to the invention preferably contains about 45 to 90 /O by weight of pure protein (preferably soya protein). Commercially available products with protein contents of, for example, 60 to 90% by weight can be used as the soya protein. Accordingly, the dietetic composition preferably contains about 70 to 97% by weight of soya protein (in the form in which it is commercially available).
Citrus pectin or dried apple pectin are suitable for use as the pectin, citrus pectin being preferred.
The composition according to the invention may, if desired, also contain other active compounds, in particular one or more vitamins and/or one or more mineral salts.
Suitable vitamins are, for example, vitamin A and derivatives (such as esters) thereof, such as vitamin
A palmitate; vitamin B 1 and derivatives thereof (for example, salts thereof, such as the chloridehydrochloride or the nitrate); vitamin B 2 and derivatives thereof (for example, esters thereof, such as riboflavin 5'-phosphate); vitamin B 6 and derivatives thereof (for example, salts thereof, such as pyridoxine hydrochloride); vitamin B 12 and derivatives thereof (for example, hydroxocobalamin); biotin; nicotinic acid and derivatives thereof, for example, nicotinamide; folic acid and derivatives thereof; pantothenic acid and derivatives thereof, for example, salts thereof, such as sodium pantothenate or calcium pantothenate, and panthenol; ascorbic acid (vitamin C) and salts thereof, for example, sodium ascorbate, calcium ascorbate or iron-ll ascorbate; vitamin E and derivatives thereof, for example, a tocopherol acetate and a - tocopherol succinate; rutin and derivatives thereof, for example, sodium salts of rutoside-sulphuric acid esters; vitamin D 2 and D 3 and derivatives thereof; and vitamin K and derivatives thereof, for example, menadiol diacetate.
Suitable mineral salts are, for example, salts of potassium, such as potassium chloride or citrate, of magnesium, such as basic magnesium carbonate or magnesium sulphate, citrate, levulinate or hydrogen phosphate, of calcium, such as calcium citrate, fluoride, hydrogen phosphate, gluconate, lactategluconate or lactobionate, of copper, such as copper sulphate or gluconate, of zinc, such as zinc sulphate, of molybdenum, such as sodium molybdate, of manganese, such as manganese chloride, sulphate or gluconate, of iron, such as iron-ll ascorbate, gluconate, fumarate, phosphate or sulphate, of cobalt, such as cobalt chloride, sulphate or gluconate, and of phosphorus, such as disodium hydrogen phosphate.
The dietetic composition according to the invention may also contain other additives, for example flavour correctors. Examples of suitable flavour correctors are sugars, such as beet sugar (sucrose), glucose, fructose and lactose, sugar substitutes, such as sorbitol, manitol, xylitol and glycerol, or sweeteners, such as saccharines and salts thereof, for example sodium saccharinate; flavour-improving acids, such as citric acid or tartaric acid; spices and aroma substances.
The composition may also contain auxiliaries, for
example binders and disintegrating agents, such as
starch, and lubricants, such as alginic acid, adipic
acid or salts thereof.
The composition according to the invention can be
made in granular form by conventional granulation
processes (for example with the aid of a granulating
roller or by spray-granulation, pelleting or calender
ing). The granules preferably have particle sizes of
from 0.5 to 5 mm, more preferably 1 to 3 mm.
The granules can be provided with any desired
coatings to improve the flavour and/or their life and
for facilitating their consumption. This is advantage
ously effected by a method conventionally used to form dragees using sugar or a sugar substitute, or by
other customary processes, for example, by spray
ing. Any conventional substance permitted by the food laws can be employed for such coating. If an
enrichment of vitamins, mineral substances, trace
elements or other active compounds, or of aroma substances, to improve the flavour, is desired, these can be incorporated into the coating.A particularly advantageous coating from the point of view of the physiology of nutrition is, for example, a vitamincontaining coating which contains all the essential vitamins, and/or a dragee shell, which is applied for protection of the granules and for achieving a smooth surface to facilitate eatjng. If sugar substitutes, such as sorbitol, xylitol or manitol, are used instead of sugar in the preparation of the dragee shell, a further sweetener can be added to intensify the sweetening power. In principle, any dragee coating which is suitable for other foodstuffs or pharmaceutical formulations can be used.
The composition according to the invention can be administered in the dry state or, for example, with cold or hot water, and also, of course, together with other drinks or foodstuffs, which should be as low in calories as possible, for example with fruit juices or vegetable juices, such as orange juice, lemon juice, blackcurrant juice, tomato juice or carrot juice; with milk, in particular skimmed milk or buttermilk; with coffee or tea; with lean meat broth; or with apple puree oryoghurt.
A unit dose of the dietetic composition according to the invention, which should be taken in place of one meal, preferably contains about 15 to 40, preferably 20 to 30, g. of soya protein (corresponding to 80-120 calories) and accordingly about 0.4 to 1.2, preferably 0.5 to 1, g of pectin. With three meals daily, this corresponds to a daily dose of about 60 to 90 g of soya protein and 1.5 to 3 g of pectin.
The unit dose preferably additionally contains 1,500 to 2,500 (preferably 2,000) international units of vitamin A,
0.3 to 0.7 (preferably 0.5) mg of vitamin B1,
0.5 to 1 (preferably 0.75) mgofvitamin B 2,
0.5 to 1 (preferably 0.75) mg of vitamin B 6,
1.2 to 3 (preferably 2) ,ag of vitamin B 12, 20 to 60 (preferably 35) mg of vitamin C,
5 to 20 (preferably 12) mg of vitamin E,
3 to 10 (preferably 5) mg of nicotinamide,
2 to 8 (preferably 4) mg of pantothenic acid (preferably in the form of its calcium salt),
0.1 to 0.2 (preferably 0.15) mg of folic acid,
200 to 500 (preferably 350) mg of calcium in the
form of calcium salts, and
3 to 10 (preferably 5) mg of iron in the form of iron
salts.
In order that the invention may be more fully
understood, the following Examples are given by
way of illustration only.
Example 1
29.1 kg of powdered soya protein (of purity 70%) is
mixed with 0.9 kg of powdered citrus pectin and 3.0
kg of powdered sucrose and the mixture is spray
granulated with water in fluidised bed drier (initial
temperature 60"C).
A unit dose of the resulting granules is 33 g. Such
granules can be administered in liquid carriers such
as water, juice, yoghurt or soup.
Example 2
29.1 kg of soya protein (of purity 70 /O) is mixed with
0.9 kg of citrus pectin in a kneader and 33 litres of
water is added. The moist mass is processed on a
roller granulatorto sausage-shaped granules (length
about 2 mm, diameter about 1.5 mm) and the granules are dried in a fluidised bed at 60"C and sieved (Sieve No.4, mesh width about 0.8 mm).
Example 3
30 kg of granules prepared according t6 Example 2
is sprayed with an aqueous solution containing 9.54
kg of beet sugar, 0.06 kg of citric acid and 0.40 kg of
methyl cellulose (initial temperature 80"C).
A unit dose of the resulting granules is 40 g.
Example 4
12 kg of granules prepared according to Example 2 is moistened with a beet sugar syrup in a coating kettle and 0.60 kg of a vitamin/mineral salt mixture sprinkled in, coating being repeated using a beet sugar/powdered cocoa mixture such that the final composition contains 3.20 kg of beet sugar and 0.20 kg of cocoa powder.
The vitamin/mineral salt mixture used has the following composition: 2.8 g of vitamin A acetate, 0.2 g of thiamine mononitrate, 0.12 g of riboflavin, 0.4 g of pyridoxole hydrochloride, 0.2 mg of cyanocobalamin, 0.04 g of folic acid, 1.2 g of calcium D - (+) panthothenate, 1.6 g of nicotinamide, 12 g of ascorbic acid, 12 g of dried vitamin E powder (25% pure), 21.48 g of calcium hydrogen phosphate, 0.16 g of copper sulphate, 8 g of iron-ll sulphate, 8 g of basic magnesium carbonate and 532 g of beet sugar.
A unit dose of the resulting coated granules is 40 g.
Example 5
A mixture of 29 kg of soya bean protein,0.8 of dried apple pectin, 2.5 kg of sorbitol and 0.5 kg of aroma substances is ground (particle size: about 50 to 200,u), passed through a high-speed sieve (sieve size 4), mixed and sieved again. The pulverulent mixture is then spray-granulated in a fluidised bed drier with a solution, warmed to about 50 , of 160 g of pectin and 40 g of sorbitol in 8 litres of water.
The resulting dry granules (unit dose: 33 g) may
be administered by stirring into cold or warm water ortaken by the spoonful with a liquid (at least 150 ml of liquid).
Sorbitol can be replaced by the same amounts of
beet sugar, glucose, fructose, manitol, xylitol or glycerol.
Example 6
Example 5 is repeated, but pure water is used for the spray-granulation instead of the pectin/sorbitol solution.
Claims (9)
1. A hypo -,p - lipoproteinaemic dietetic composition, which comprises soya protein and 2 to 3% by weight, on a dry solids weight basis, of pectin, the composition being in the form of granules.
2. A composition according to claim 1,which has a pure protein content of 60 to 90% by weight.
3. A composition according to claim 1 or 2, in which the granules have particle sizes of 0.5 to 5 mm.
4. A composition according to any of claims 1 to 3, which contains one or more ingredients selected from vitamins, mineral salts, flavouring agents, binders, disintegrating agents and lubricants.
5. A composition according to any of claims 1 to 4, in which the granules have a coating thereon.
6. A composition according to any of claims 1 to 5, which is in the form of a unit dose.
7. A composition according to claim 1, substantially as herein described in any of examples 1 to 6.
8. Afoodstuff or beverage which contains a composition according to any of claims 1 to 7.
9. For use in reducing the cholesterol level in humans or animals, a composition according to any of claims 1 to 7.
