Abstract
1506017 Fluorometric sample detection INTERNATIONAL DIAGNOSTIC TECHNOLOGY Inc 26 Feb 1975 [4 March 1974] 8059/75 Headings G1A and G1B A fluorometric system comprising means for exciting and means for receiving fluorescent light and a body 10 with an elongate handle joined to a portion having a continuous surface carrying a reagent 11 selectively reactive with the biologically derived substance 14 to be detected. Sample 11 may be an antigen and substance 14 an antibody or vice versa. The body may have two samples one of which is a standard, the intensity of fluorescence being proportional to the quantity of antigen. Alternatively the samples may comprise different antigens (or antibodies), the labels being different fluorescent substances. The sample areas may comprise bands. The fluorometric system may comprise such a body and a monochromatic light source between 0À2 and 1À0Ám and means, as light guide 24, to convey the light to the body of a detector of fluorescence. Nylon sphere 10 of diameter 10mm bears a dried antibody film 11. Glass cuvette is racked for 5 mins with added serum 13 so that antigen 14 present binds to 11. The cuvette is rinsed, incubated with solution of antibody 23 tagged with sodium fluorescein isothyocyanate and resinsed. Ultraviolet light is then passed through filter 28 of 460 nm. Fluorescence from antibody 23 reaches a photo-multiplier via a filter and a fibre optic cable. The electric signal is filtered, rectified, linearized, amplified, converted to digital and displayed on a digital meter calibrated in titre. Two fibre optic cables each with filters and a chopper wheel may supply the photo-multiplier with fluorescence alternatively from the two bands. Two photomultipliers may be used without a chopper. The substrate may be paddle shaped, a cube or other polyhedron with a handle and with a different sample on each of the vertical faces. The fluorometer may have a filter wheel with a selected fluorescence wavelength for each face. A modified fluorometer may have a single fibre optic cable 86 dividing into two or into four as in Fig. 9. Substrate 87 has fluorescent sample 88. Branches 81, 82 transmit light from lamps 89 and 90 and branches 83 and 84 conduct the fluoroescence through lenses 91 and 92 to solid state detectors or photo-multipliers 93, 94. Surface 88 may include a plurality of randomly dispersed fluorescent samples, tagged with different exciting wavelengths and excited from a single lamp via a plurality of filters.
