Abstract
1401297 Analysis using enzyme bound hapten etc SYNTEX ENERGY RESEARCH Inc and VARIAN ENERGY 15 May 1972 [14 May 1971] 22778/72 Heading G1B A method of detecting a ligand in a medium comprises contacting in a liquid phase the medium, an enzyme-bound ligand and a soluble receptor for the ligand, the enzyme-bound ligand being capable of competing with the ligand for the reception sites of the receptor and comparing the enzyme activity then obtained with that obtained for a mixture of the same amounts of enzymebound ligand and of receptor. The interaction of the enzyme-bound ligand with receptor reduces the enzyme activity. The ligand may be a monomeric or polymeric material, especially a drug, a cell or virus. The receptor may be an enzyme, antibody (obtained when the ligand is hapten using as antigen the ligand linked to protein), RNA, DNA, or a naturally occurring receptor such as thyrobinding globulin. Examples disclose the detection of codeine using carboxymethyl morphine-amylase and morphine antiserum assaying the amylase using a suspension of dyed amylose, the measurement of Pysozyme activity in morphine-lysozyme using a suspension of dried bacteria, Micrococcus lysodeikticus in sodium phosphate pH 7.0, the effect of addition of meperidine on the inhibition of activity in lysozyme-meperidine by addition of BSA-meperidine antiserum, the effect of addition of phenobarbital on the inhibition of activity in a phenobarbital-lysozyme BSA-N-carboxymethyl phenobarbital antiserum, the detection of testosterone using testosterone-3-carboxymethyl oxime-maleate dehydrogenase and antibody, assaying using oxalacetic acid and NADH, the assay of barbiturate in a synthetic urine (described) using phenobarbital - or secobarbital-lysozyme and antibodies to BSA, conjugates of analogues to these barbitals, a substrate for determination of horse-radish peroxidase (HRP) prepared by forming a leuco-dye by reaction of toluic acid p,p' -dimethylamino benzophenone, converting this to the mixed anhydride and adding this with pyridine to a DMF solution of polyvinylalcohol at 100C, the resulting polymer giving a blue colour with HRP. The preparation of other enzyme-bound ligands is described.
