1463448 5 - Phenyl - thiazolidin - 4 - ones WARNER-LAMBERT CO 14 Nov 1975 47043/75 Heading C2C Novel 5-phenyl-thiazolidin-4-ones of the general Formula I wherein R 1 is C 1-6 alkyl, R 2 is C 1-6 alkyl or aralkyl and R 3 is C 1-6 alkyl, aralkyl or in which m is 0, 1, 2 or 3 and each of R 4 and R 5 is C 1 -C 6 alkyl or hydroxyalkyl or NR 4 R 5 is a C 4-6 saturated heterocyolic ring, and, when R 3 contains a basic group, pharmacologically compatible acid addition salts thereof are prepared (a) when R 3 contains an alkylene group connected to the thiazolidine ring, by reaction of a compound of the general Formula II wherein X is H or an alkali metal atom, with VR 6 in which V is a reactive ester radical and R 6 is as R 3 except that m is other than 0; and (b) when R 3 is an amino group connected via a nitrogen atom to the thiazolidine ring, by reaction of a compound (II), wherein X is a halogen atom, with R 4 NHR 5 , followed optionally by salification of the product. Novel thiazolidinones of the general Formula VIII are prepared by reacting a compound of the general Formula VI with NC-CH 2 -COOR 1 in the presence of a basic catalyst and may be alkylated to give the corresponding 3-alkyl derivatives and brominated to form the corresponding 3-alkyl-5-bromo derivatives. Pharmaceutical compositions having hepatoprotective and curative anti-hepatotoxic activity comprise, as active ingredient, at least one 5-phenyl-thiazolidin-4-one (I) or pharmacologically compatible acid addition salts thereof, in admixture with a solid or liquid pharmaceutical diluent or carrier.
