Abstract
1338579 Electro-phoresis F R ELEVITCH 16 Nov 1970 [17 Nov 1969] 54511/70 Heading C7B [Also in Divisions B1 and B5] In preparation for the electro-phoresis of a specimen (e.g. of proteins or blood serum) in a gel, the gel is shaped and stored ready for use in apparatus comprising an impermeable base member 11 (which may be in the form of a multiplicity of segments or a continuous strip) and a cover member 10. The gel, which is in the form of a layer of aqueous permeable gel optionally containing reagent, is located between the members and is adherent to the base member, the inner surface of the cover member being uniformly coated with a normally solid thermoplastic essentially hydrocarbon material which is in surface contact with the gel and non-adherent to it. Thereby, the gel can be stored and the cover be easily separated there from when ready to perform the electro-phoresis. The layer of gel may be not more than 1 mm thick and this permits rapid migration under an applied voltage of 10 to 15 volts per cm. In the embodiment shown, the inner surface of cover member 10 has ribs 19 and projections 24a, 24b; in assembling the apparatus the edges of cover 10 are heat or adhesive-sealed to base member 11, gel is poured in through a vent 26 and the vent is capped or sealed. If the base member is a continuous strip it may then be cut into segments. For use, the cover member is stripped off and sample introduced into the wells formed in the gel by ribs 19 and projections 24. The base and/or cover member may be of polystyrene, polyethylene or polypropylene. The inner surface of the cover member may be pre-coated with a paraffin wax solution. The gel formulation may be 10% sucrose, 1 to 1.2% agarose in an aqueous solution of sodium barbital /HCL pH 8.6, optionally containing 0.50% to 0.2% polyvinyl alcohol. Other gels comprise starch, agar or acrylamide. In another embodiment Fig.10 (not shown) a package 110 comprises a plurality of base members each carrying a layer of gel without wells formed in it, one face of each base member being non-adherent to the adjacent gel layer and so forming the cover member for that gel layer.
