Abstract

1333420 Determination of amylase XEROX CORP 1 Dec 1970 56964/70 Heading B1X Amylase is determined in body fluids using a reagent combination comprising (A) a tetrazolium salt and (B) an alkaline buffer capable of inactivating amylase in a serum-starch substrate solution. The reagent used in the test comprise a composition comprising soluble starch as substrate (C), a buffer (D) to maintain the sample-substrate solution at pH 6 to 8, and an amylaseactivating substance (E) which may be sodium chloride or potassium chloride, together with the reagent combination as above. The alkaline buffer (B) may comprise diethylamine hydrochloride and lithium hydroxide in proportions to give a pH of at least 10. The reagent may be in the form of three tablets, comprising (C), (D), (E) and surfactant; lithium hydroxide, (E), and surfactant; diethylamine hydrochloride, (A), (E) and surfactant. In one method of carrying out the test, a serum sample is incubated with the first tablet, the second and third tablets added, and incubation continued. Optical density is then read periodically at 560 mu. There is reference to a conventional procedure wherein serum is incubated with starch substrate solution, the mixture is acidified and sodium tungstate solution added then an alkaline copper solution, the mixture is incubated and an arsenomolybdate colour forming reagent added.
