Abstract
1331513 Microbial polysaccharides KUREHA KAGAKU KOGYO KK 14 Oct 1970 [15 Oct 1969] 48870/70 Heading C3U [Also in Division A5] Polysaccharides and polysaccharide-protein complexes possessing anti-carcinogenic properties are recovered from a liquid e.g. hot water, extract of a mycelium of a species of Basidiomycetes or a cultured broth of the species in a conventional fashion, after first removing free protein, free nucleic acids, reducing sugars, various ions and vitamins to give a greyish white amorphous solid characterized by the following properties: water-soluble, non-hydroscopic and non-dialyzable, gives a positive result when tested for the presence of glucose after hydrolysis with IN sulphuric acid; gives negative results when subjected to the ferric chloride reaction for determining the presence of phenols and to the Fehling reaction for determining the presence of reducing sugars; gives positive results when subjected to the anisaldehyde-sulphuric acid reaction, the Molisch reaction with α-naphthol the anthrone-sulphuric acid reaction and the tryptophanesulphuric acid reaction for detecting the presence of any saccharide, gives a positive result when subjected to the chromotropic acidsulphuric acid reaction for detecting the presence of a hexose, the aniline-hydrochloric acid reaction for detecting the presence of a pentose, the resorcinol-hydrochloric acid reaction (Seliwanoff's reaction) for detecting the presence of a ketohexose and pentose, the carbazole-cysteinesulphuric acid reaction for detecting the presence of a ketose, the Tollens reaction, and the thiogylcol-sulphuric acid reaction for detecting a ketose and uronic acid; shows only one spot at the anode side when subjected to electrophoresis in a 0À05 mol sodium borate solution for 90 minutes using a cellulose acetate membrane at 20-25 V/cm; and shows no antimicrobial action to bacteria, fungi and yeasts such as Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Aspergillus niger and Candida albicans. It is in this last respect stated to be differentiated from anti-bacterial anticarcinogenic substances obtained from species of Actinomycetes. The 19 examples describe detailed recovery procedures and specifically mention a very large number of species and strains of Basidiomycetes.
