Abstract

1384051 Endo-beta-glucanase NOVO TERAPEUTISK LABORATORIUM AS 18 Jan 1972 [26 Jan 1971] 3232/71 Headings C3H and C3U [Also in Division C6] Endo-beta-glucanase is obtained by aerobic cultivation of Bacillus pumilus on a conventional nutrient medium and then isolating the enzyme for example by treating the purified and/or concentrated broth with salts for example sodium or ammonium sulphate or with a watermiscible solvent for example acetone or ethanol followed by drying the precipitated enzyme preferably by spray- or freeze-drying. The enzyme is used as a viscosity-reducing agent in brewer's wort. Strains of Bacillus pumilus exemplified:-A.T.C.C. Nos. 6632; 12,140; 98; 70; 72; 71; 6,631; 945; 4,520; 14,884; 15,477; 18; 1; 4,522 and NCIB Nos. 7,576, 8,982, 8,600, 8,738. Beta glucan is obtained from an aqueous extract of milled barley, destarched by treatment with bacterial amylase containing no betaglucanase activity, precipitation with acetone, redissolution in water and then re-precipitation with 30% ammonium sulphate. The preparation is then exhaustively dialyzed against distilled water and freeze-dried.
