1382606 o-Aminoalkylbenzoice acid drivatives F HOFFMANN-LA ROCHE & CO AG 22 Sept 1972 43993/72 Heading C2C Novel compounds of Formula I and salts wherein R 1 represents a hydrogen atom or a hydroxy, lower alkoxy or lower alkyl group, R 2 represents a hydrogen atom or a lower alkyl group, R 3 represents a lower alkyl, aralkyl or (lower alkyl)-aryl group, R 4 represents a hydrogen atom or a lower alkyl, aralkyl, (lower alkyl)-aryl or (lower alkoxy)-(lower alkyl) group and R 5 represents a lower alkyl, aralkyl, (lower alkyl)-aryl or (lower alkoxy)-(lower alkyl) group or R 4 and R 5 together with the nitrogen atom to which they are attached represent a saturated five- or six-membered heterocyclic ring which may contain an oxygen atom or a further nitrogen atom which may carry a lower alkyl group (lower denotes groups containing up to 7 carbon atoms), are prepared either (a) by esterification, usually in situ with R 3 OH of the free acid which is obtained by hydrolysis of compounds of Formula VI (b) by substituting the amino group in a compound of Formula III with R 4 and/or R 5 by known methods. Intermediates of Formula VI above are obtained from a m-trifluoromethylphenyl-magnesium Grignard reagent by reaction with an appropriate alkylene oxide followed by halogenation of the resulting alcohol and reacting the halide with HNR 4 R 5 . Pharmaceutical compositions in conventional forms for oral or parenteral administration and having the effect of enhancing learning and memory or having cholinergic and spasmolytic activity comprise an above novel compound and a carrier or diluent.
