Abstract

1356794 Tissue cells DEFENCE SECRETARY OF STATE FOR 21 Feb 1972 [20 Nov 1970] 55268/70 Heading C6F [Also in Division B1] A method for the production of single tissue cells from subdivided tissue (chicken embryo, rabbit, kidney, brain) suspended with concomitant red blood cells in isotonic saline solution in a container 1, Fig. 1, involves adding isotonic solution through line 4 and removing isotonic solution and red bloood cells through rotary filter 19, hollow shaft 15 and line 14 under the suction created by a peristaltic pump (not shown). Thereafter, the flow of liquid is stopped while the tissue is enzymatically disaggregated by trypsin and the single tissue cells produced are likewise removed through the filter. The filter element has a pore size which permits only particles having a mean diameter up to three times and generally about twice the diameter of red blood cells in the tissue suspension to be drawn through. The single cells are subsequently recovered from suspension by centrifugation. Preferably a collagen-digesting enzyme, such as pepsin and/or deoxyribonuclease, is used with the trypsin in order to digest the gelatinous fibrin which would otherwise obstruct the filter.
