Abstract
A composition for detecting urea in body and other fluids comprises urease, a buffer compound producing ammonium ions and a colorixmetric indicator. The urease is preferably water-soluble. Specified buffer compounds are ammonium salts of citric, lactic, oxalic, benzoic, acetic, salicylic, stearic, propionic, butyric or phosphoric acids, or mixtures thereof. Specified indicators are bromthymol blue, bromocresol purple, dichlorosulphonphthalein, 6:8-dinitro-2:4-quinazolinedione, alizarin, and 2-(2:4-dinitrophenylazo) -1- naphthol-3:6-disulphonic acid. In addition, as colour stabilisers there may be present, albuminoid proteins (bovine albumin, egg albumin) and if desired in addition a heteropolysaccharide (gums arabic, tragacanth or mesquite, Damson or Indian gum); as sensitising agents there may be present an aliphatic amide of M.P. below 150 DEG C. (acetamide, propionamide, butramide, acrylamide); gelatin may be present as thickening agent; polyvinyl alcohol or a solid polyethylene glycol may be present as a wetting agent; and inert dyes may be present to give a colour background. The composition may be in the form of tablets, pellets or powder and may be obtained by lyophilising a solution or suspension of the ingredients. The ingredients may alternatively be incorporated in a bibulous strip and if desired a dialysing polymeric film coated thereon to permit urea present in a liquid, e.g. blood, to be tested to come into contact with the reagents whilst larger molecules e.g. haemoglobin are held back and do not mask the colour. Ethyl cellulose dissolved in benzene may be used to form the coating.
