Abstract
Crude hirudin, obtained by acetone extraction of alcohol-dehydrated leech heads followed by pptn. from the acetone solution, is purified by: a) precipitating the hirudin from an aqueous solution thereof by addition of a water-miscible solvent, b) stirring a solution of the precipitated hirudin in 0.01M Amm. acetate soln. with oxycellulose (cellulose which has been oxidized by treatment with NO2), c) eluting the hirudin from the oxycellulose with 0.55M Amm. acetate buffer, d) absorbing the hirudin on a weakly acid cation exchange resin (crosslinked styreneacrylic acid copolymer containing free COOH groups) and e) eluting with 5% NH4OH solution.
