Abstract

A composition for the determination of aldolase, e.g. in body liquids of animals and man, comprises fructose- 1:6-diphosphate, glyceraldehyde phosphate dehydrogenase, an alkali metal arsenate, nicotinic acid adenine dinucleotide, 2:6-dichloroindophenol, phenazine methosulphate and a triethanolamine buffer of pH 7-8.5. In the presence of aldolase, the fructose-1:6-diphosphate is hydrolysed to 3-phosphoglyceraldehyde and in the presence of glyceraldehyde phosphate dehydrogenase and arsenate, oxidation takes place, whilst the dinucleotide is reduced: the dyestuff system (2:6 -dichloroindophenol, phenazine methosulphate) is then reduced by the reduced dinucleotide. The materials may be put up as a test pack in the form of the following aqueous solutions:-(1) triethanolamine, nicotinic acid adenine dinucleotide, dichloroindophenol and arsenate, (2) phenazine methosulphate, (3) fructose- 1:6-diphosphate, (4) glyceraldehyde phosphate dehydrogenase and (5) triethanolamine buffer. Alternatively the composition may be impregnated in a suitable carrier, e.g. filter paper strips, and the product lyophilised.
