Abstract

A gonadotropic hormone active both as a follicle-stimulant hormone, and as a luteinic hormone and devoid of antigen properties, is prepared by treating fresh urine obtained from women in menopausal or post-menopausal stage, at a pH of 4 to 4.5 with kaolin purified by washing with dilute mineral acid, separating the kaolin with adsorbed hormone, washing, extracting with aqueous ammonia of pH 11 to 11.3, centrifuging and adjusting the pH of the liquid so obtained to pH 8.5 by the addition of acetic acid, discarding the precipitate, lowering the pH of the residual liquid to pH 5 to 5.5 and adding acetone to precipitate the crude hormone which is further purified, repeated if necessary, by chromatographic adsorption on a cationic exchange zeolite, previously acidified to a pH of 3.7 to 3.8, followed by washing with water to neutrality, next with aqueous ethanol of 70 to 80% concentration, then with aqueous ethanol of 70 to 80% concentration containing ammonium acetate and finally eluting with aqueous ethanol of 35 to 45% concentration containing ammonium acetate, the hormone being precipitated from the eluate by increasing the ethanol content of the eluate, and is then washed and dried. It is preferred to precipitate the hormone by adjusting to an ethanol concentration of 80%.
